Abstract: The speed profiles, gas holdup (eG) and global oxygen transfer coefficient (kLa) from a stirred airlift bioreactor using water as the fluid model, was investigated by computational fluid dynamics modeling. The parameters predicted by the computer model were validated with the experimental dates. The CFD results were very close to those obtained experimentally. During the simulation it was verified a prevalent impeller effect at low speeds, propelling a large volume of fluid against the walls of the vessel, which without recirculation, results in low values of eG and kLa; however, by increasing air velocity, the impeller effect is smaller with the air flow being greater, in the region of the riser, causing fluid recirculation, which explains the increase in eG and kLa.
Abstract: The aim of the study is to improve removal of trace organic contaminants dissolved in activated sludge by the process of filtration with membrane bioreactor combined with modified activated carbon, for a maximum removal of organic compounds characterized by low molecular weight. Special treatment was conducted in laboratory on activated carbon. Tow reaction parameters: the pH of aqueous middle and the type of granular activated carbon were very important to improve the removal and to motivate the electrostatic Interactions of organic compounds with modified activated carbon in addition to physical adsorption, ligand exchange or complexation on the surface activated carbon. The results indicate that modified activated carbon has a strong impact in removal 21 of organic contaminants and in percentage of 100% of the process.
Abstract: The performance of airlift bioreactors are closely related with their geometry, especially the gas-liquid separator design. In this study, the influence of the gas-liquid separator geometry on oxygen transfer and gas hold-up was evaluated in 10-L concentric-tube airlift bioreactor operating with distilled water and xanthan gum solution. The specific airflow rate (ɸAIR) exhibited the higher effect on the oxygen transfer coefficient (kLa) for both fluids. While the gas-liquid separator openness angle (α) and liquid volume fraction on the gas-liquid separator (VGLS) have presented opposite effects on oxygen mass transfer, they affected negatively the global gas hold-up of distilled water system. The best degassing zone geometry corresponded to a 90° openness angle with 10% of the liquid on it.
Abstract: This work deals with the synthesis and the determination of some surface properties of a new anionic surfactant belonging to sulfonamide derivatives. The interest in this new surfactant is that its behavior in aqueous solution is interesting both from a fundamental and a practice point of view. Indeed, it is well known that this kind of surfactant leads to the formation of bilayer structures, and the microstructures obtained have applications in various fields, ranging from cosmetics to detergents, to biological systems such as cell membranes and bioreactors. The surfactant synthesized from pure n-alkane by photosulfochlorination and derivatized using N-ethanol amine is a mixture of position isomers. These compounds have been analyzed by Gas Chromatography coupled to Mass Spectrometry by Electron Impact mode (GC -MS/IE), and IR. The surface tension measurements were carried out, leading to the determination of the critical micelle concentration (CMC), surface excess and the area occupied per molecule at the interface. The foaming power has also been determined by Bartsch method, and the results have been compared to those of commercial surfactants. The stability of the foam formed has also been evaluated. These compounds show good foaming power characterized in most cases by dry foam.
Abstract: A predictive clustering hybrid regression (pCHR)
approach was developed and evaluated using dataset from H2-
producing sucrose-based bioreactor operated for 15 months. The aim
was to model and predict the H2-production rate using information
available about envirome and metabolome of the bioprocess. Selforganizing
maps (SOM) and Sammon map were used to visualize the
dataset and to identify main metabolic patterns and clusters in
bioprocess data. Three metabolic clusters: acetate coupled with other
metabolites, butyrate only, and transition phases were detected. The
developed pCHR model combines principles of k-means clustering,
kNN classification and regression techniques. The model performed
well in modeling and predicting the H2-production rate with mean
square error values of 0.0014 and 0.0032, respectively.
Abstract: A Laboratory-scale packed bed reactor with microbial
cellulose as the biofilm carrier was used to investigate the
denitrification of high-strength nitrate wastewater with specific
emphasis on the effect the nitrogen loading rate and hydraulic
retention time. Ethanol was added as a carbon source for
denitrification. As a result of this investigation, it was found that up
to 500 mg/l feed nitrate concentration the present system is able to
produce an effluent with nitrate content below 10 ppm at 3 h
hydraulic retention time. The highest observed denitrification rate
was 4.57 kg NO3-N/ (m3 .d) at a nitrate load of 5.64 kg NO3-
N/(m3 .d), and removal efficiencies higher than 90% were obtained
for loads up to 4.2 kg NO3-N/(m3 .d). A mass relation between COD
consumed and NO3-N removed around 2.82 was observed. This
continuous-flow bioreactor proved an efficient denitrification system
with a relatively low retention time.
Abstract: Thirty six samples from each (aerobic and anoxic)
activated sludge were collected from two wastewater treatment plants
with MBRs in Berlin, Germany. The samples were prepared for count
and definition of fungal isolates; these isolates were purified by
conventional techniques and identified by microscopic examination.
Sixty tow species belonging to 28 genera were isolated from
activated sludge samples under aerobic conditions (28 genera and 58
species) and anoxic conditions (26 genera and 52 species). The
obtained data show that, Aspergillus was found at 94.4% followed by
Penicillium 61.1 %, Fusarium (61.1 %), Trichoderma (44.4 %) and
Geotrichum candidum (41.6 %) species were the most prevalent in all
activated sludge samples. The study confirmed that fungi can thrive
in activated sludge and sporulation, but isolated in different numbers
depending on the effect of aeration system. Some fungal species in
our study are saprophytic, and other a pathogenic to plants and
animals.
Abstract: In this study, cometabolic biodegradation of
chloroform was experimented with mixed cultures in the presence of
various organic solvents like methanol, ethanol, isopropanol, acetone,
acetonitrile and toluene as these are predominant discharges in
pharmaceutical industries. Toluene and acetone showed higher
specific chloroform degradation rate when compared to other
compounds. Cometabolic degradation of chloroform was further
confirmed by observation of free chloride ions in the medium. An
extended Haldane model, incorporating the inhibition due to
chloroform and the competitive inhibition between primary
substrates, was developed to predict the biodegradation of primary
substrates, cometabolic degradation of chloroform and the biomass
growth. The proposed model is based on the use of biokinetic
parameters obtained from single substrate degradation studies. The
model was able to satisfactorily predict the experimental results of
ternary and quaternary mixtures. The proposed model can be used for
predicting the performance of bioreactors treating discharges from
pharmaceutical industries.
Abstract: The fluorescent pseudomonad strain R81 is a root
colonizing rhizobacteria which promotes the growth of many plants
by various mechanisms. Its broth containing siderophore (ironchelating
compound) and 2,4- diacetyl phloroglucinol (DAPG) is
used for preparing bioinoculant formulations for agronomical
applications. Glycerol was found to be the best carbon source for
improved biomass production. Splitting of nitrogen source to NH4Cl
and urea had a stabilizing effect on pH during batch cultivation. Ltryptophan
at 0.5 % in the medium increased the siderophore
production to 850 mg/l. During batch cultivation of the strain in a
bioreactor, a maximum of 4 g/l of dry cell mass, 1.8 g/l of
siderophore and 20 mg/l of DAPG was achieved when glycerol was
15 g/l and C/N ratio was maintained at 12.5. In case of intermittent
feeding of fresh medium during fed-batch cultivation, the dry cell
mass was increased to 25 g/l with improved production of DAPG to
70 mg/l.
Abstract: This work aims to investigate a potential of
microalgae for utilizing industrial wastewater as a cheap nutrient for
their growth and oil accumulation. Wastewater was collected from
the effluent ponds of agro-industrial factories (cassava and ethanol
production plants). Only 2 microalgal strains were isolated and
identified as Scenedesmus quadricauda and Chlorella sp.. However,
only S. quadricauda was selected to cultivate in various wastewater
concentrations (10%, 20%, 40%, 60%, 80% and 100%). The highest
biomass obtained at 6.6×106 and 6.27×106 cells/ml when 60%
wastewater was used in flask and photo-bioreactor. The cultures gave
the highest lipid content at 18.58 % and 42.86% in cases of S.
quadricauda and S. obliquus. In addition, under salt stress (1.0 M
NaCl), S. obliquus demonstrated the highest lipid content at 50%
which was much more than the case of no NaCl adding. However, the
concentration of NaCl does not affect on lipid accumulation in case
of S. quadricauda.
Abstract: The performance of a sucrose-based H2 production in
a completely stirred tank reactor (CSTR) was modeled by neural
network back-propagation (BP) algorithm. The H2 production was
monitored over a period of 450 days at 35±1 ºC. The proposed model
predicts H2 production rates based on hydraulic retention time
(HRT), recycle ratio, sucrose concentration and degradation, biomass
concentrations, pH, alkalinity, oxidation-reduction potential (ORP),
acids and alcohols concentrations. Artificial neural networks (ANNs)
have an ability to capture non-linear information very efficiently. In
this study, a predictive controller was proposed for management and
operation of large scale H2-fermenting systems. The relevant control
strategies can be activated by this method. BP based ANNs modeling
results was very successful and an excellent match was obtained
between the measured and the predicted rates. The efficient H2
production and system control can be provided by predictive control
method combined with the robust BP based ANN modeling tool.
Abstract: An immunomodulator bioproduct is prepared in a
batch bioprocess with a modified bacterium Pseudomonas
aeruginosa. The bioprocess is performed in 100 L Bioengineering
bioreactor with 42 L cultivation medium made of peptone, meat
extract and sodium chloride. The optimal bioprocess parameters were
determined: temperature – 37 0C, agitation speed - 300 rpm, aeration
rate – 40 L/min, pressure – 0.5 bar, Dow Corning Antifoam M-max.
4 % of the medium volume, duration - 6 hours. This kind of
bioprocesses are appreciated as difficult to control because their
dynamic behavior is highly nonlinear and time varying. The aim of
the paper is to present (by comparison) different models based on
experimental data.
The analysis criteria were modeling error and convergence rate.
The estimated values and the modeling analysis were done by using
the Table Curve 2D.
The preliminary conclusions indicate Andrews-s model with a
maximum specific growth rate of the bacterium in the range of
0.8 h-1.
Abstract: Biomass is becoming a large renewable resource for
power generation; it is involved in higher frequency in
environmentally clean processes, and even it is used for biofuels
preparation. On the other hand, hydrogen – other energy source – can
be produced in a variety of methods including gasification of
biomass. In this study, the production of hydrogen by gasification of
biomass waste is examined. This work explores the production of a
gaseous mixture with high power potential from Amazonas´ specie
known as copoazu, using a counter-flow fixed-bed bioreactor.
Abstract: A novel sponge submerged membrane bioreactor
(SSMBR) was developed to effectively remove organics and
nutrients from wastewater. Sponge is introduced within the SSMBR
as a medium for the attached growth of biomass. This paper evaluates
the effects of new and acclimatized sponges for dissolved organic
carbon (DOC) removal from wastewater at different mixed liquor
suspended solids- (MLSS) concentration of the sludge. It was
observed in a series of experimental studies that the acclimatized
sponge performed better than the new sponge whilst the optimum
DOC removal could be achieved at 10g/L of MLSS with the
acclimatized sponge. Moreover, the paper analyses the relationships
between the MLSSsponge/MLSSsludge and the DOC removal efficiency
of SSMBR. The results showed a non-linear relationship between the
biomass parameters of the sponge and the sludge, and the DOC
removal efficiency of SSMBR. A second-order polynomial function
could reasonably represent these relationships.
Abstract: The objective of this study is to evaluate the
occurrence of fungi in aerobic and anoxic activated sludge from
membrane bioreactors (MBRs). Thirty-six samples of both aerobic
and anoxic activated sludge were taken from 2 MBR treating
domestic wastewater. Over a period of eight months 2 samples from
each plant were taken per month. The samples were prepared for
count and definition of fungi. The obtained data show that, sixty
species belonging to 27 genera were collected from activated sludge
samples under aerobic and anoxic conditions. Regarding to the fungi
definition, under aerobic condition the Geotrichum was found at
(8.8%) followed by Penicillium (75.0%), Yeasts (65.7%) and
Trichoderma (55.5%), while Yeasts (77.1%) Geotrichum
candidumand Penicillium (61.1%) species were the most prevalent in
anoxic activated sludge. The results indicate that activated sludge is
habitat for growth and sporulation of different groups of fungi, both
saprophytic and pathogenic.
Abstract: Highly pathogenic avian influenza (HPAI) H5N1 viruses have created demand for a cost-effective vaccine to prevent a pandemic of the disease. Here, we report that Trichoplusia ni (T. ni) larvae can act as a cost-effective bioreactor to produce recombinant HA5 (rH5HA) proteins as an potential effective vaccine for chickens. To facilitate the recombinant virus identification, virus titer determination and access the infected larvae, we employed the internal ribosome entry site (IRES) derived from Perina nuda virus (PnV, belongs to insect picorna like Iflavirus genus) to construct a bi-cistronic baculovirus expression vector that can express the rH5HA protein and enhanced green fluorescent protein (EGFP) simultaneously. Western blot analysis revealed that the 70 kDa rH5HA protein and partially cleaved products (40 kDa H5HA1) were generated in T. ni larvae infected with recombinant baculovirus carrying the H5HA gene. These data suggest that the baculovirus-larvae recombinant protein expression system could be a cost-effective platform for H5N1 vaccine production.
Abstract: Saccharomyces cerevisiae (baker-s yeast) can exhibit
sustained oscillations during the operation in a continuous bioreactor
that adversely affects its stability and productivity. Because of
heterogeneous nature of cell populations, the cell population balance
models can be used to capture the dynamic behavior of such cultures.
In this paper an unstructured, segregated model is used which is
based on population balance equation(PBE) and then in order to
simulation, the 4th order Rung-Kutta is used for time dimension and
three methods, finite difference, orthogonal collocation on finite
elements and Galerkin finite element are used for discretization of the
cell mass domain. The results indicate that the orthogonal collocation
on finite element not only is able to predict the oscillating behavior of
the cell culture but also needs much little time for calculations.
Therefore this method is preferred in comparison with other methods.
In the next step two controllers, a globally linearizing control (GLC)
and a conventional proportional-integral (PI) controller are designed
for controlling the total cell mass per unit volume, and performances
of these controllers are compared through simulation. The results
show that although the PI controller has simpler structure, the GLC
has better performance.
Abstract: Three reactor types were explored and successfully
used for pigment production by Monascus: shake flasks, and shaken
and stirred miniaturized reactors. Also, the use of dielectric
spectroscopy for the on-line measurement of biomass levels was
explored. Shake flasks gave good pigment yields, but scale up is
difficult, and they cannot be automated. Shaken bioreactors were less
successful with pigment production than stirred reactors.
Experiments with different impeller speeds in different volumes of
liquid in the reactor confirmed that this is most likely due oxygen
availability. The availability of oxygen appeared to affect biomass
levels less than pigment production; red pigment production in
particular needed very high oxygen levels. Dielectric spectroscopy
was effectively used to continuously measure biomass levels during
the submerged fungal fermentation in the shaken and stirred
miniaturized bioreactors, despite the presence of the solid substrate
particles. Also, the capacitance signal gave useful information about
the viability of the cells in the culture.
Abstract: In this study, the designed dual stage membrane
bioreactor (MBR) system was conceptualized for the treatment of
cyanide and heavy metals in electroplating wastewater. The design
consisted of a primary treatment stage to reduce the impact of
fluctuations and the secondary treatment stage to remove the residual
cyanide and heavy metal contaminants in the wastewater under
alkaline pH conditions. The primary treatment stage contained
hydrolyzed Citrus sinensis (C. sinensis) pomace and the secondary
treatment stage contained active Aspergillus awamori (A. awamori)
biomass, supplemented solely with C. sinensis pomace extract from
the hydrolysis process. An average of 76.37%, 95.37%, 93.26 and
94.76% and 99.55%, 99.91%, 99.92% and 99.92% degradation
efficiency for total cyanide (T-CN), including the sorption of nickel
(Ni), zinc (Zn) and copper (Cu) were observed after the first and
second treatment stages, respectively. Furthermore, cyanide
conversion by-products degradation was 99.81% and 99.75 for both
formate (CHOO-) and ammonium (NH4
+) after the second treatment
stage. After the first, second and third regeneration cycles of the C.
sinensis pomace in the first treatment stage, Ni, Zn and Cu removal
achieved was 99.13%, 99.12% and 99.04% (first regeneration cycle),
98.94%, 98.92% and 98.41% (second regeneration cycle) and 98.46
%, 98.44% and 97.91% (third regeneration cycle), respectively.
There was relatively insignificant standard deviation detected in all
the measured parameters in the system which indicated
reproducibility of the remediation efficiency in this continuous
system.
Abstract: Fructooligosaccharides derived from microbial enzyme especially from fungal sources has been received particular attention due to its beneficial effects as prebiotics and mass production. However, fungal fermentation is always cumbersome due to its broth rheology problem that will eventually affect the production of FOS. This study investigated the efficiency of immobilized cell system using rotating fibrous bed bioreactor (RFBB) in producing fructooligosaccharides (FOS). A comparative picture with respect to conventional stirred tank bioreactor (CSTB) and RFBB has been presented. To demonstrate the effect of agitation intensity and aeration rate, a laboratory-scale bioreactor 2.5 L was operated in three phases (high, medium, low) for 48 hours. Agitation speed has a great influence on P. simplicissimum fermentation for FOS production, where the volumetric FOS productivity using RFBB is increased with almost 4 fold compared to the FOS productivity in CSTB that only 0.319 g/L/h. Rate of FOS production increased up to 1.2 fold when immobilized cells system was employed at aeration rate similar to the freely suspended cells at 2.0 vvm.
