Abstract: The alternative technique for sterilization of culture
medium to replace autoclaving was carried out. For sterilization of
culture medium without autoclaving, some commercial pure essential
oils, bergamot oil, betel oil, cinnamon oil, lavender oil and turmeric
oil, were tested alone or in combinations with some disinfectants,
10% povidone-iodine and 2% iodine + 2.4% potassium iodide. Each
essential oil or combination was added to 25-mL Murashige and
Skoog (MS) medium before medium was solidified in a 120-mL
container, kept for 2 weeks before evaluating sterile conditions.
Treated media, supplemented with essential oils, were compared to
control medium, autoclaved at 121 degree Celsius for 15 min. In
vitro sterile conditions were found 20 – 100% from these treated
media compared to 100% sterile condition from autoclaved medium.
Treated media obtained 100% sterile conditions were chosen for
culturing chrysanthemum shoots. It was found that 10% povidoneiodine
in combination with cinnamon oil (3:1) and 2% iodine + 2.4%
potassium iodide in combination with lavender oil (1:3) at the
concentration of 36 3L/25 mL medium provided the promising
growth of shoot explants.
Abstract: This paper presents a novel method for inferring the
odor based on neural activities observed from rats- main olfactory
bulbs. Multi-channel extra-cellular single unit recordings were done
by micro-wire electrodes (tungsten, 50μm, 32 channels) implanted in
the mitral/tufted cell layers of the main olfactory bulb of anesthetized
rats to obtain neural responses to various odors. Neural response
as a key feature was measured by substraction of neural firing rate
before stimulus from after. For odor inference, we have developed a
decoding method based on the maximum likelihood (ML) estimation.
The results have shown that the average decoding accuracy is about
100.0%, 96.0%, 84.0%, and 100.0% with four rats, respectively. This
work has profound implications for a novel brain-machine interface
system for odor inference.
Abstract: This study was carried out to reveal the bacterial composition of aerosol in the studied abattoirs. Bacteria isolated were characterized according to microbiological standards. Factors such as temperature and distance were considered as variable in this study. The isolation was carried out at different temperatures such as 27oC, 31oC and 29oC and at various distances of 100meters and 200meters away from the slaughter sites. Result obtained showed that strains of Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Lactobacillus alimentarius and Micrococcus sp. were identified. The total viable counts showed that more microorganisms were present in the morning while the least viable count of 388cfu was recorded in the evening period of this study. This study also showed that more microbial loads were recorded the further the distance is to the slaughter site. Conclusively, the array of bacteria isolated suggests that abattoir sites may be a potential source of pathogenic organisms to commuters if located within residential environment.
Abstract: The objective of this research was to investigate biodegradation of water hyacinth (Eichhornia crassipes) to produce bioethanol using dilute-acid pretreatment (1% sulfuric acid) results in high hemicellulose decomposition and using yeast (Pachysolen tannophilus) as bioethanol producing strain. A maximum ethanol yield of 1.14g/L with coefficient, 0.24g g-1; productivity, 0.015g l-1h-1 was comparable to predicted value 32.05g/L obtained by Central Composite Design (CCD). Maximum ethanol yield coefficient was comparable to those obtained through enzymatic saccharification and fermentation of acid hydrolysate using fully equipped fermentor. Although maximum ethanol concentration was low in lab scale, the improvement of lignocellulosic ethanol yield is necessary for large scale production.
Abstract: The study aimed to investigate characteristics of
vegetative tissue for taxonomic purpose and possibly trend of waste
application in industry. Stems and branches of 15 species in Solanum
found in Thailand were prepared for fiber and examined by light
microscopy. Microstructural characteristic data of fiber i.e. fiber
length and width, fiber lumen diameter and fiber cell wall thickness
were recorded. The longest average fiber cell length (>3.9 mm.) were
obtained in S. lycopersicum L. and S. tuberosum L. Fiber cells from
S. lycopersicum also revealed the widest average diameter of whole
cell and its lumen at >45.5 μm and >29 μm respectively. However
fiber cells with thickest wall of > 9.6 μm were belonged to the
ornamental tree species, S. wrightii Benth. The results showed that
the slenderness ratio, Runkel ratio, and flexibility coefficient, with
potentially suitable for feedstock in paper industry fell in 4 exotic
species, i.e. Solanumamericanum L., S. lycopersicum, S.
seaforthianum Andr., and S. tuberosum L
Abstract: Strain M was isolated from the latex of Hevea brasiliensis that grow in the rubber farm area of Malaysia Rubber Board. Strain M was tentatively identified as Bacillus sp. Strain M demonstrated high protease production at pH 9, and this was suitable to be applied in rubber processing that was in alkaline conditions. The right and suitable proportion to be used in applying supernatant into the latex was two parts of latex and one part of enzyme. In this proportion, the latex was stable throughout the 72 hours of treatment. The potential of strain M to degrade protein in the natural rubber latex was proven with the reduction of 79.3% nitrogen in 24 hours treatment. Centrifugation process of the latex before undergoing the treatment had increased the protein degradation in latex. Although the centrifugation process did not achieve zero nitrogen content, it had improved the performance of protein denaturing in the natural rubber.
Abstract: Inferring the network structure from time series data
is a hard problem, especially if the time series is short and noisy.
DNA microarray is a technology allowing to monitor the mRNA
concentration of thousands of genes simultaneously that produces
data of these characteristics. In this study we try to investigate the
influence of the experimental design on the quality of the result.
More precisely, we investigate the influence of two different types of
random single gene perturbations on the inference of genetic networks
from time series data. To obtain an objective quality measure for
this influence we simulate gene expression values with a biologically
plausible model of a known network structure. Within this framework
we study the influence of single gene knock-outs in opposite to
linearly controlled expression for single genes on the quality of the
infered network structure.
Abstract: Tufting carpet is a very suitable substrate for growing
microorganism such as pathogenic microbes, due to the direct touch
with human body, long washing periods and laying on the floor; in
fact there are 3 major problems: To risk human health, Prepare bad
odors and Destruction of the products.. In the presented research, for
investigation of presence most common microbes on polyester
tufting, first goods laid in a public place (in the corridor fair) for 30
days and the existence of some microbes were investigate on it with
two methods of enrichment in nutrient environments such as
thioglycolate and noutrunt brath, and shake the dust off the polyester
tufting onto cultivation mediums such as blood agar and noutrunt
agar. After the microorganism colonics are grown, the colonies were
separated and six microbial tests such as cataloes and sitrat were
carried out in five phases on the colonics for identifying the varieties
of bacteria. As a result of tests, 5 type of bacteria, such as
Escherichia coli, staphylococcus saprophytic as were identified. Each
of the mentioned bacteria can be seriously harmful for the heath of
human.
Abstract: Studies were carried out on the comparative study of the production of Avicelase enzyme using sugarcane bagasse-SCB in two different statuses (i.e. treated and untreated SCB) by thermophilic Geobacillus stearothermophilus at 50ºC. Only four thermophilic bacterial isolates were isolated and assayed for Avicelase production using UntSCB and TSCB. Only one isolate selected as most potent and identified as G. stearothermophilus used in this study. A specific endo-β-1,4-D-glucanase (Avicelase EC 3.2.1.91) was partially purified from a thermophilic bacterial strain was isolated from different soil samples when grown on cellulose enrichment SCB substrate as the sole carbon source. Results shown that G. stearothermophilus was the better Avicelase producer strain. Avicelase had an optimum pH and temperature 7.0 and 50ºC for both UntSCB and TSCB and exhibited good pH stability between "5-8" and "4-9", however, good temperature stability between (30-80ºC) for UntSCB and TSCB, respectively. Other factors affecting the production of Avicelase were compared (i.e. SCB concentration, inoculum size and different incubation periods), all results observed and obtained were revealed that the TSCB was exhibited maximal enzyme activity in comparison with the results obtained from UntSCB, so, the TSCB was enhancing the Avicelase production.
Abstract: A new sythetic gene coding for a Human
Elastin-Like Polypeptide was constructed and expressed. The
recombinant product was tested as coating agent to realize a
surface suitable for cell growth. Coatings showed peculiar
features and different human cell lines were seeded and
cultured. All cell lines tested showed to adhere and proliferate
on this substrate that has been shown also to exert a specific
effect on cells, depending on cell type.
Abstract: Retinal prostheses have been successful in eliciting
visual responses in implanted subjects. As these prostheses progress,
one of their major limitations is the need for increased resolution. As
an alternative to increasing the number of electrodes, virtual
electrodes may be used to increase the effective resolution of current
electrode arrays. This paper presents a virtual electrode technique
based upon time-offsets between stimuli. Two adjacent electrodes are
stimulated with identical pulses with too short of pulse widths to
activate a neuron, but one has a time offset of one pulse width. A
virtual electrode of twice the pulse width was then shown to appear in
the center, with a total width capable of activating a neuron. This can
be used in retinal implants by stimulating electrodes with pulse
widths short enough to not elicit responses in neurons, but with their
combined pulse width adequate to activate a neuron in between them.
Abstract: The most important subtype of non-Hodgkin-s
lymphoma is the Diffuse Large B-Cell Lymphoma. Approximately
40% of the patients suffering from it respond well to therapy,
whereas the remainder needs a more aggressive treatment, in order to
better their chances of survival. Data Mining techniques have helped
to identify the class of the lymphoma in an efficient manner. Despite
that, thousands of genes should be processed to obtain the results.
This paper presents a comparison of the use of various attribute
selection methods aiming to reduce the number of genes to be
searched, looking for a more effective procedure as a whole.
Abstract: Certain sciences such as physics, chemistry or biology,
have a strong computational aspect and use computing infrastructures
to advance their scientific goals. Often, high performance and/or high
throughput computing infrastructures such as clusters and computational
Grids are applied to satisfy computational needs. In addition,
these sciences are sometimes characterised by scientific collaborations
requiring resource sharing which is typically provided by Grid
approaches. In this article, I discuss Grid computing approaches in
High Energy Physics as well as in bioinformatics and highlight some
of my experience in both scientific domains.
Abstract: Themain goal of this article is to find efficient
methods for elemental and molecular analysis of living
microorganisms (algae) under defined environmental conditions and
cultivation processes. The overall knowledge of chemical
composition is obtained utilizing laser-based techniques, Laser-
Induced Breakdown Spectroscopy (LIBS) for acquiring information
about elemental composition and Raman Spectroscopy for gaining
molecular information, respectively. Algal cells were suspended in
liquid media and characterized using their spectra. Results obtained
employing LIBS and Raman Spectroscopy techniques will help to
elucidate algae biology (nutrition dynamics depending on cultivation
conditions) and to identify algal strains, which have the potential for
applications in metal-ion absorption (bioremediation) and biofuel
industry. Moreover, bioremediation can be readily combined with
production of 3rd generation biofuels. In order to use algae for
efficient fuel production, the optimal cultivation parameters have to
be determinedleading to high production of oil in selected
cellswithout significant inhibition of the photosynthetic activity and
the culture growth rate, e.g. it is necessary to distinguish conditions
for algal strain containing high amount of higher unsaturated fatty
acids. Measurements employing LIBS and Raman Spectroscopy were
utilized in order to give information about alga Trachydiscusminutus
with emphasis on the amount of the lipid content inside the algal cell
and the ability of algae to withdraw nutrients from its environment
and bioremediation (elemental composition), respectively. This
article can serve as the reference for further efforts in describing
complete chemical composition of algal samples employing laserablation
techniques.
Abstract: An expressed sequence tag (EST) analysis provideus portions of expressed genes. We have constructed cDNA library and determined randomly sequences from cDNA library clones of T. molitor injected with acholeplasma lysate. We identified the homologous to a galectin gene. As the result of cloning and characterization of novel, we found that the protein has an open reading frame (ORF) of 495 bp, with 164 amino acid residues and molecular weight of 18.5 kDa. To characterize the role of novel Tm-galectin in immune system, we quantified the mRNA level of galectin at different times after treatment with immune elicitors. The galectin mRNA was up-regulated about 7-folds within 18 hrs. This suggests that Tm-galectin is a novel member of animal lectins, and has a role in the process of pathogen recognition. Our study would be helpful for the study on immune defense system and signaling cascade.
Abstract: Gene networks present a graphical view at the level of gene activities and genetic functions and help us to understand complex interactions in a meaningful manner. In the present study, we have analyzed the gene interaction of PPAR-γ (peroxisome proliferator-activated receptor gamma) by search tool for retrieval of interacting genes. We find PPAR-γ is highly networked by genetic interactions with 10 genes: RXRA (retinoid X receptor, alpha), PPARGC1A (peroxisome proliferator-activated receptor gamma, coactivator 1 alpha), NCOA1 (nuclear receptor coactivator 1), NR0B2 (nuclear receptor subfamily 0, group B, member 2), HDAC3 (histone deacetylase 3), MED1 (mediator complex subunit 1), INS (insulin), NCOR2 (nuclear receptor co-repressor 2), PAX8 (paired box 8), ADIPOQ (adiponectin) and it augurs well for the fact that obesity and several other metabolic disorders are inter related.
Abstract: The account of genus Polygonum L. in "Flora of Armenia" was made more than five decades ago. After that many expeditions have been carried out in different regions of Armenia and a huge herbarium material has been collected. The genus included 5 sections with 20 species. Since then many authors accepted the sections as separate genera on the basis of anatomical, morphological, palynological and molecular data. According to the above mentioned it became clear, that the taxonomy of Armenian representatives of Polygonum s. l. also needs revision. New literature data and our investigations of live and herbarium material (ERE, LE) with specification of the morphological characters, distribution, ecology, flowering and fruiting terms brought us to conclusion, that genus Polygonum s. l. has to be split into 5 different genera (Aconogonon (Meisn.) Reichenb., Bistorta (L.) Scop., Fallopia Adans., Persicaria Mill., Polygonum L. s. s.). The number of species has been reduced to 16 species. For each genus new determination keys has been created.
Abstract: This experiment was performed to optimize the medium for tissue culture of Taraxacum kok-saghyz Rodin. Different tissue culture approaches such as shoot regeneration from seed, callus formation from leaf explants and plant regeneration from callus were investigated in this study. All the explants were cultured on MS basal medium supplemented with 20g/l sucrose, 7g/l agar and different plant growth regulators. Seeds of Taraxacum kok-saghyzwere cultured on media containing different levels of BA and 2,4-D (0.5, 1.0 and 3.0mg/L) to direct shoot regeneration study. Leaf explants were cultured in different combination of BA (at three levels: 0.5, 1.0 and 3.0mg/L) and zeatin (at two levels: 0.5 and 1.0mg/L) to examine callus formation. After the callus formation the formed calli were cultured on different combinations of BA and NAA for shoot regeneration. BA at three levels (0.5 and 1.0 and 3.0mg/L) and NAA at two levels (0.5 and 1.0mg/L) in all possible combinations were used for shoot regeneration from callus. The results showed that the treatment containing 1.0mg/L 2,4-D in combination with 1.0mg/L BA was found to be the best one for shoot regeneration from seeds. The treatment with 1.0mg/L BA in combination with 1.0mg/L zeatin were found to be suitable treatments for callus production from leaf explants, as well. Moreover, 0.5mg/L BA alone or in combination with 1.0mg/L NAA were found to be the best treatments for shoot regeneration from callus.
Abstract: Earphones and headphones, which are compact electro-acoustic transducers, tend to have a lot of acoustic absorption materials and porous materials known as dampers, which often have a large number of extremely small holes and narrow slits to inhibit the resonance of the vibrating system, because the air viscosity significantly affects the acoustic characteristics in such acoustic paths. In order to perform simulations using the finite element method (FEM), it is necessary to be aware of material characteristics such as the impedance and propagation constants of sound absorbing materials and porous materials. The transfer function is widely known as a measurement method for an acoustic tube with such physical properties, but literature describing the measurements at the upper limits of the audible range is yet to be found. The acoustic tube, which is a measurement instrument, must be made narrow, and the distance between the two sets of microphones must be shortened in order to take measurements of acoustic characteristics at higher frequencies. When such a tube is made narrow, however, the characteristic impedance has been observed to become lower than the impedance of air. This paper considers the cause of this phenomenon to be the effect of the air viscosity and describes an FEM analysis of an acoustic tube considering air viscosity to compare to the theoretical formula by including the effect of air viscosity in the theoretical formula for an acoustic tube.
Abstract: Spoilage occurs in plant produce due to the action of field and storage microorganisms. The conditions of storage can also cause physiological spoilage. Various methods exist to ensure that these food substances maintain their quality long after harvesting. However, many of these methods either fail to keep the plant for the required period or predispose the plant to other spoilage risks. The major shortcoming posed by the use of many antimicrobials is the chemical residues it deposits in the food substance. The use of plants in preservation has been in use for a long period, though little understood then, it served its purposes. A better understanding of the roles of these plant parts in increasing the shelf life of farm produce has helped in the creation of more effective and safer means of pest and microbial control. This can be extended to plants that have not been used for these purposes initially. Microbial sources should also be investigated as these have provided cheaper sources of secondary metabolites.
