Abstract: This paper discusses about the conceptual design and development of the control system software using Programmable logic controller (PLC) for autonomous driving and agricultural operations of Multipurpose Agricultural Robot Platform (MARP). Based on given initial conditions by field analysis and desired agricultural operations, the structural design development of MARP is done using modelling and analysis tool. PLC, being robust and easy to use, has been used to design the autonomous control system of robot platform for desired parameters. The robot is capable of performing autonomous driving and three automatic agricultural operations, viz. hilling, mulching, and sowing of seeds in the respective order. The input received from various sensors on the field is later transmitted to the controller via ZigBee network to make the changes in the control program to get desired field output. The research is conducted to provide assistance to farmers by reducing labor hours for agricultural activities by implementing automation. This study will provide an alternative to the existing systems with machineries attached behind tractors and rigorous manual operations on agricultural field at effective cost.
Abstract: The robot manipulator is an equipment that stands out for two reasons: Firstly because of its characteristics of movement and reprogramming, resembling the arm; secondly, by adding several areas of knowledge of science and engineering. The present work shows the development of the prototype of a robotic manipulator driven by a Programmable Logic Controller (PLC), having two degrees of freedom, which allows the movement and displacement of mechanical parts, tools, and objects in general of small size, through an electronic system. The aim is to study direct and inverse kinematics of the robotic manipulator to describe the translation and rotation between two adjacent links of the robot through the Denavit-Hartenberg parameters. Currently, due to the many resources that microcomputer systems offer us, robotics is going through a period of continuous growth that will allow, in a short time, the development of intelligent robots with the capacity to perform operations that require flexibility, speed and precision.
Abstract: Egg yolk oil is a concentrated source of egg bioactive compounds, such as fat-soluble vitamins, phospholipids, cholesterol, carotenoids and others. To extract lipids and other fat-soluble nutrients from liquid egg yolk, a two-step extraction process involving polar (ethanol) and non-polar (hexane) solvents were used. This extraction technique was based on egg yolk bioactive compounds polarities, where non-polar compound was extracted into non-polar hexane, but polar in to polar alcohol/water phase. But many egg yolk bioactive compounds are not strongly polar or non-polar. Egg yolk phospholipids, cholesterol and pigments are amphipatic (have both polar and non-polar regions) and their behavior in ethanol/hexane solvent system is not clear. The aim of this study was to clarify the behavior of phospholipids, cholesterol and carotenoids during extraction of egg yolk oil with ethanol and hexane and determine the loss of these compounds in egg yolk oil. Egg yolks and egg yolk oil were analyzed for phospholipids (phosphatidylcholine (PC) and phosphatidylethanolamine (PE)), cholesterol and carotenoids (lutein, zeaxanthin, canthaxanthin and β-carotene) content using GC-FID and HPLC methods. PC and PE are polar lipids and were extracted into polar ethanol phase. Concentration of PC in ethanol was 97.89% and PE 99.81% from total egg yolk phospholipids. Due to cholesterol’s partial extraction into ethanol, cholesterol content in egg yolk oil was reduced in comparison to its total content presented in egg yolk lipids. The highest amount of lutein and zeaxanthin was concentrated in ethanol extract. The opposite situation was observed with canthaxanthin and β-carotene, which became the main pigments of egg yolk oil.
Abstract: This paper presents a configuration software solution in Linux, which is used for the control of space environment simulator. After introducing the structure and basic principle, it is said that the developing of QT software frame and the dynamic data exchanging between PLC and computer. The OPC driver in Linux is also developed. This driver realizes many-to-many communication between hardware devices and SCADA software. Moreover, an algorithm named “Scan PRI” is put forward. This algorithm is much more optimizable and efficient compared with "Scan in sequence" in Windows. This software has been used in practical project. It has a good control effect and can achieve the expected goal.
Abstract: In the present study, highly effective iTLC single strip method for the determination of radiochemical purity (RCP) of 68Ga-BCA-peptides was developed (with no double-developing, changing of eluents or other additional manipulation). In this method iTLC-SG strips and commonly used eluent TFAaq. (3-5 % (v/v)) are used. The method allows determining each of the key radiochemical forms of 68Ga (colloidal, bound, ionic) separately with the peaks separation being no less than 4 σ. Rf = 0.0-0.1 for 68Ga-colloid; Rf = 0.5-0.6 for 68Ga-BCA-peptides; Rf = 0.9-1.0 for ionic 68Ga. The method is simple and fast: For developing length of 75 mm only 4-6 min is required (versus 18-20 min for pharmacopoeial method). The method has been tested on various compounds (including 68Ga-DOTA-TOC, 68Ga-DOTA-TATE, 68Ga-NODAGA-RGD2 etc.). The cross-validation work for every specific form of 68Ga showed good correlation between method developed and control (pharmacopoeial) methods. The method can become convenient and much more informative replacement for pharmacopoeial methods, including HPLC.
Abstract: Lactulose is a synthetic disaccharide which has remarkable applications in food and pharmaceutical fields. Lactulose is not found in nature and it is produced by isomerization reaction of lactose in an alkaline environment. It should be noted that this reaction has a very low yield since significant amount of lactose stays un-reacted in the system. Basically, purification of lactulose is difficult and costly. Previous studies have revealed that solubility of lactose and lactulose are significantly different in ethanol. Considering the fact that solubility is also affected by temperature itself, we investigated the effect of ethanol and temperature on separation process of lactose from the syrup containing lactose and lactulose. For this purpose, a saturated solution containing lactulose and lactose was made at three different temperatures; 25⁰C (room temperature), 31⁰C, and 37⁰C first. Five samples containing 2g saturated solution was taken and then 2g, 3g, 4g, 5g, and 6g ethanol separately was added to the sampling tubes. Sampling tubes were kept at respective temperatures afterward. The concentration of lactose and lactulose after separation process measured and analyzed by High Performance Liquid Chromatography (HPLC). Results showed that ethanol has such a greater impact than operating temperature on purification process. Also, it was observed that the maximum rate of separation occurred at initial amount of added ethanol.
Abstract: Even its content is rich in antioxidants ϒ-oryzanol, rice bran is not used properly as functional food. This research aims to (1) extract ϒ-oryzanol; (2) determine the solubility of ϒ-oryzanol in supercritical CO2 based on phase equilibrium theory; and (3) study the effect of process variables on solubility. Extraction experiments were carried out for rice bran (5 g) at various extraction pressures, temperatures and reaction times. The flowrate of supercritical fluid through the extraction vessel was 25 g/min. The extracts were collected and analysed with high-pressure liquid chromatography (HPLC). The conclusion based on the experiments are as: (1) The highest experimental solubility was 0.303 mcg/mL RBO at T= 60°C, P= 90 atm, t= 30 min; (2) Solubility of ϒ-oryzanol was influenced by pressure and temperature. As the pressure and temperature increase, the solubility increases; (3) The solubility data of supercritical extraction can be successfully determined using phase equilibrium theory. Meanwhile, tocopherol was found and slightly investigated in this work.
Abstract: Pectinatella magnifica (Leidy, 1851) is an invasive freshwater animal that lives in colonies. A colony of Pectinatella magnifica (a gelatinous blob) can be up to several feet in diameter large and under favorable conditions it exhibits an extreme growth rate. Recently European countries around rivers of Elbe, Oder, Danube, Rhine and Vltava have confirmed invasion of Pectinatella magnifica, including freshwater reservoirs in South Bohemia (Czech Republic). Our project (Czech Science Foundation, GAČR P503/12/0337) is focused onto biology and chemistry of Pectinatella magnifica. We monitor the organism occurrence in selected South Bohemia ponds and sandpits during the last years, collecting information about physical properties of surrounding water, and sampling the colonies for various analyses (classification, maps of secondary metabolites, toxicity tests). Because the gelatinous matrix is during the colony lifetime also a host for algae, bacteria and cyanobacteria (co-habitants), in this contribution, we also applied a high performance liquid chromatography (HPLC) method for determination of potentially present cyanobacterial toxins (microcystin-LR, microcystin-RR, nodularin). Results from the last 3-year monitoring show that these toxins are under limit of detection (LOD), so that they do not represent a danger yet. The final goal of our study is to assess toxicity risks related to fresh water resources invaded by Pectinatella magnifica, and to understand the process of invasion, which can enable to control it.
Abstract: Substandard and counterfeit antimalarials is a major problem in malaria endemic areas. The availability of counterfeit/ substandard medicines is not only decreasing the efficacy in patients, but it is also one of the contributing factors for developing antimalarial drug resistance. Owing to this, a pilot study was conducted to survey quality of drugs collected from different malaria endemic areas of India. Artesunate+Sulphadoxine-Pyrimethamine (AS+SP), Artemether-Lumefantrine (AL), Chloroquine (CQ) tablets were randomly picked from public health facilities in selected states of India. The quality of antimalarial drugs from these areas was assessed by using Global Pharma Health Fund Minilab test kit. This includes physical/visual inspection and disintegration test. Thin-layer chromatography (TLC) was carried out for semi-quantitative assessment of active pharmaceutical ingredients. A total of 45 brands, out of which 21 were for CQ, 14 for AL and 10 for AS+SP were tested from Uttar Pradesh (U.P.), Mizoram, Meghalaya and Gujrat states. One out of 45 samples showed variable disintegration and retension factor. The variable disintegration and retention factor which would have been due to substandard quality or other factors including storage. However, HPLC analysis confirms standard active pharmaceutical ingredient, but may be due to humid temperature and moisture in storage may account for the observed result.
Abstract: Anogeissus leiocarpus (Combretaceae) is well known
for its medicinal uses in African traditional medicine, for treating
many human diseases mainly skin diseases and infections. Mycetoma
disease is a fungal and/ or bacterial skininfection, mainly cause by
Madurella mycetomatis fungus. This study was carried out in vitro to
investigate the antifungal activity of Anogeissus leiocarpus leaf
extracts against the isolated pathogenic Madurella mycetomatis, by
using the NCCLS modified method compared to Ketoconazole
standard drug, and MTT assay. The bioactive fraction was subjected
to chemical analysis implementing different chromatographic
analytical methods (TLC, HPLC, and LC-MS/MS). The results
showed significance antifungal activity of A. leiocarpus leaf extracts
against the isolated pathogenic M. mycetomatis, compared to negative
and positive controls. The chloroform fraction showed the highest
antifungal activity. The chromatographic analysis of the chloroform
fraction with the highest activity showed the presence of important
bioactive compounds such as ellagic and flavellagic acids derivatives,
flavonoids and stilbenoid, which are well known for their antifungal
activity.
Abstract: High Performance Liquid Chromatography (HPLC)
method was developed and validated for simultaneous estimation of
6-Gingerol(6G) and 6-Shogaol(6S) in joint pain relief gel containing
ginger extract. The chromatographic separation was achieved by
using C18 column, 150 x 4.6mm i.d., 5μ Luna, mobile phase
containing acetonitrile and water (gradient elution). The flow rate
was 1.0 ml/min and the absorbance was monitored at 282 nm. The
proposed method was validated in terms of the analytical parameters
such as specificity, accuracy, precision, linearity, range, limit of
detection (LOD), limit of quantification (LOQ), and determined
based on the International Conference on Harmonization (ICH)
guidelines. The linearity ranges of 6G and 6S were obtained over 20-
60 and 6-18 μg/ml respectively. Good linearity was observed over the
above-mentioned range with linear regression equation Y= 11016x-
23778 for 6G and Y = 19276x-19604 for 6S (x is concentration of
analytes in μg/ml and Y is peak area). The value of correlation
coefficient was found to be 0.9994 for both markers. The limit of
detection (LOD) and limit of quantification (LOQ) for 6G were
0.8567 and 2.8555 μg/ml and for 6S were 0.3672 and 1.2238 μg/ml
respectively. The recovery range for 6G and 6S were found to be
91.57 to 102.36 % and 84.73 to 92.85 % for all three spiked levels.
The RSD values from repeated extractions for 6G and 6S were 3.43
and 3.09% respectively. The validation of developed method on
precision, accuracy, specificity, linearity, and range were also
performed with well-accepted results.
Abstract: Magnetic Resonance Imaging (MRI) is one of the
most important medical imaging modality. Subjective assessment of
the image quality is regarded as the gold standard to evaluate MR
images. In this study, a database of 210 MR images which contains
ten reference images and 200 distorted images is presented. The
reference images were distorted with four types of distortions: Rician
Noise, Gaussian White Noise, Gaussian Blur and DCT compression.
The 210 images were assessed by ten subjects. The subjective scores
were presented in Difference Mean Opinion Score (DMOS). The
DMOS values were compared with four FR-IQA metrics. We have
used Pearson Linear Coefficient (PLCC) and Spearman Rank Order
Correlation Coefficient (SROCC) to validate the DMOS values. The
high correlation values of PLCC and SROCC shows that the DMOS
values are close to the objective FR-IQA metrics.
Abstract: The consumption of food contaminated with molds
(microscopic filamentous fungi) and their toxic metabolites results in
the development of food-borne mycotoxicosis. The spores of molds
are ubiquitously spread in the environment and can be detected
everywhere. Ochratoxin A is a toxic and potentially carcinogenic
fungal toxin found in a variety of food commodities. In this study, the
mycological quality of various ready-to-eat local and imported pork
meat and meat byproducts sold in Egyptian markets were assessed
and the presence of various molds was determined in pork used as a
raw material, edible organs as liver and kidney as well as in
fermented raw meat by-products. The study assessed the mycological
quality of pork raw meat and their by-products sold in commercial
shops in Cairo, Egypt. Mycological analysis was conducted on
(n=110) samples which included pig’s livers and kidneys from
Egyptian Bassatin slaughter house; local and imported processed
pork meat by-products from Egyptian pork markets. The isolates
were identified using traditional mycological and biochemical tests.
All kidney and liver samples were positive to molds growth while all
byproducts were negative. Ochratoxin A levels were quantitatively
analyzed using the high performance liquid chromatography (HPLC)
and the highest results were present in kidney 7.51 part per billion
(ppb) followed by minced meat 6.19 ppb generally the local samples
showed higher levels than the imported ones. To the best of our
knowledge, this is the first report on mycotoxins detection and
quantification from pork by-products in Egypt.
Abstract: The end of the line controls of the finished products in
the automotive industry is important. The control that has been
conducted with the manual methods for the sliding doors tracks is not
sufficient and faulty products cannot be identified. As a result, the
customer has the faulty products. In the scope of this study, the
design criteria of the PLC integrated modular end of line control unit
has been examined, designed and manufactured to make the control
of the 10 different track profile to 2 different vehicles with an
objective to minimize the salvage costs by obtaining more sensitive,
certain and accurate measurement results. In the study that started
with literature and patent review, the design inputs have been
specified, the technical concept has been developed, computer
supported mechanic design, control system and automation design,
design review and design improvement have been made. Laser
analog sensors at high sensitivity, probes and modular blocks have
been used in the unit. The measurement has been conducted in the
system and it is observed that measurement results are more sensitive
than the previous methods that we use.
Abstract: The work aims to develop a robot in the form of
autonomous vehicle to detect, inspection and mapping of
underground pipelines through the ATmega328 Arduino platform.
Hardware prototyping is very similar to C / C ++ language that
facilitates its use in robotics open source, resembles PLC used in
large industrial processes. The robot will traverse the surface
independently of direct human action, in order to automate the
process of detecting buried pipes, guided by electromagnetic
induction. The induction comes from coils that send the signal to the
Arduino microcontroller contained in that will make the difference in
intensity and the treatment of the information, and then this
determines actions to electrical components such as relays and
motors, allowing the prototype to move on the surface and getting the
necessary information. This change of direction is performed by a
stepper motor with a servo motor. The robot was developed by
electrical and electronic assemblies that allowed test your application.
The assembly is made up of metal detector coils, circuit boards and
microprocessor, which interconnected circuits previously developed
can determine, process control and mechanical actions for a robot
(autonomous car) that will make the detection and mapping of buried
pipelines plates. This type of prototype can prevent and identifies
possible landslides and they can prevent the buried pipelines suffer an
external pressure on the walls with the possibility of oil leakage and
thus pollute the environment.
Abstract: Phthalates are ubiquitous environmental pollutants
well known because of their endocrine disrupting activity in human
organism. The aim of our study was, by biological monitoring,
investigate exposure to phthalates of Roma ethnicity group i.e.
children and adults from 5 families (n=29, average age 11.8 ± 7.6
years) living in western Slovakia. Additionally, we analysed some
associations between anthropometric measures, questionnaire data
i.e. socio-economic status, eating and drinking habits, practise of
personal care products and household conditions in comparison with
concentrations of phthalate metabolites. We used for analysis of urine
samples high performance liquid chromatography and tandem mass
spectrometry (HPLC-MS/MS) to determine concentrations of
phthalate metabolites monoethyl phthalate (MEP), mono-n-butyl
phthalate (MnBP), mono-iso-butyl phthalate (MiBP), mono(2-ethyl-
5-hydroxyhexyl) phthalate (5OH-MEHP), mono(2-ethyl-5-oxohexyl)
phthalate (5oxo-MEHP) and mono(2-etylhexyl) phthalate (MEHP).
Our results indicate that ethnicity, lower socioeconomic status and
different housing conditions in Roma population can affect urinary
concentration of phthalate metabolites.
Abstract: The potential neuroprotective effect of Phyllantus
nuriri against Fe2+ and sodium nitroprusside (SNP) induced oxidative
stress in mitochondria of rats brain was evaluated. Cellular viability
was assessed by MTT reduction, reactive oxygen species (ROS)
generation was measured using the probe 2,7-dichlorofluoresce
indiacetate (DCFH-DA). Glutathione content was measured using
dithionitrobenzoic acid (DTNB). Fe2+ (10μM) and SNP (5μM)
significantly decreased mitochondrial activity, assessed by MTT
reduction assay, in a dose-dependent manner, this occurred in parallel
with increased glutathione oxidation, ROS production and lipid
peroxidation end-products (thiobarbituric acid reactive substances,
TBARS). The co-incubation with methanolic extract of Phyllantus
nuriri (10-200 μg/ml) reduced the disruption of mitochondrial
activity, gluthathione oxidation, ROS production as well as the
increase in TBARS levels caused by both Fe2+ and SNP in a dose
dependent manner. HPLC analysis of the extract revealed the
presence of gallic acid (20.540.01), caffeic acid (7.930.02), rutin
(25.310.05), quercetin (31.280.03) and kaemferol (14.360.01).
This result suggests that these phytochemicals account for the
protective actions of P. niruri against Fe2+ and SNP -induced
oxidative stress. Our results show that P. nuriri consist important
bioactive molecules in the search for an improved therapy against the
deleterious effects of Fe2+, an intrinsic producer of reactive oxygen
species (ROS), that leads to neuronal oxidative stress and
neurodegeneration.
Abstract: The work allowed gaining knowledge about redox and
speciation changes of As, Cr and Sb ionic forms in Klodnica River
water. This kind of studies never has been conducted in this region of
Poland. In study optimized and validated previously HPLC-ICP-MS
methods for determination of As, Sb and Cr was used. Separation
step was done using high-performance liquid chromatograph
equipped with ion-exchange column followed by ICP-MS
spectrometer detector. Preliminary studies included determination of
the total concentration of As, Sb and Cr, pH, Eh, temperature and
conductivity of the water samples. The study was conducted monthly
from March to August 2014, at six points on the Klodnica River. The
results indicate that exceeded at acceptable concentration of total Cr
and Sb was observed in Klodnica River and we should qualify
Klodnica River waters below the second purity class. In Klodnica
River waters dominates oxidized antimony and arsenic forms, as well
as the two forms of chromium Cr(VI) and Cr(III). Studies have also
shown the methyl derivative of arsenic's presence.
Abstract: The influences of cell-free solutions (CFSs) of lactic
acid bacteria (LAB) on cadaverine and other biogenic amines
production by Listeria monocytogenes and Staphylococcus aureus
were investigated in lysine decarboxylase broth (LDB) using HPLC.
Cell free solutions were prepared from Lactococcus lactis subsp.
lactis, Leuconostoc mesenteroides subsp. cremoris, Pediococcus
acidilactici and Streptococcus thermophiles. Two different
concentrations that were 50% and 25% CFS and the control without
CFSs were prepared. Significant variations on biogenic amine
production were observed in the presence of L. monocytogenes and S.
aureus (P < 0.05). The function of CFS on biogenic amine production
by foodborne pathogens varied depending on strains and specific
amine. Cadaverine formation by L. monocytogenes and S. aureus in
control were 500.9 and 948.1 mg/L, respectively while the CFSs of
LAB induced 4-fold lower cadaverine production by L.
monocytogenes and 7-fold lower cadaverine production by S. aureus.
The CFSs resulted in strong decreases in cadaverine and putrescine
production by L. monocytogenes and S. aureus, although remarkable
increases were observed for histamine, spermidine, spermine,
serotonin, dopamine, tyramine and agmatine in the presence of LAB
in lysine decarboxylase broth.
Abstract: Hydrogenated amorphous carbon (a-C:H) films have
been synthesized by a radio frequency plasma enhanced chemical
vapor deposition (rf-PECVD) technique with different bias voltage
from 0.0 to -0.5 kV. The Raman spectra displayed the polymer-like
hydrogenated amorphous carbon (PLCH) film with 0.0 to -0.1 and
a-C:H films with -0.2 to -0.5 kV of bias voltages. The surface chemical
information of all films were studied by X-ray photoelectron
spectroscopy (XPS) technique, presented to C-C (sp2 and sp3) and C-O
bonds, and relative carbon (C) and oxygen (O) atomics contents. The
O contamination had affected on structure and optical properties. The
true density of PLCH and a-C:H films were characterized by X-ray
refractivity (XRR) method, showed the result as in the range of
1.16-1.73 g/cm3 that depending on an increasing of bias voltage. The
hardness was proportional to the true density of films. In addition, the
optical properties i.e. refractive index (n) and extinction coefficient (k)
of these films were determined by a spectroscopic ellipsometry (SE)
method that give formation to in 1.62-2.10 (n) and 0.04-0.15 (k)
respectively. These results indicated that the optical properties
confirmed the Raman results as presenting the structure changed with
applied bias voltage increased.
