Abstract: Anaerobic batch experiments were conducted to investigate the effect of magnetite-supplementation (7 mM) on methane production from digested sludge undergoing two different microbial growth phases, namely fresh sludge (exponential growth phase) and degassed sludge (endogenous decay phase). Three different particle sizes were assessed: small (50 - 150 nm), medium (168 – 490 nm) and large (800 nm - 4.5 µm) particles. Results show that, in the case of the fresh sludge, magnetite significantly enhanced the methane production rate (up to 32%) and reduced the lag phase (by 15% - 41%) as compared to the control, regardless of the particle size used. However, the cumulative methane produced at the end of the incubation was comparable in all treatment and control bottles. In the case of the degassed sludge, only the medium-sized magnetite particles increased significantly the methane production rate (12% higher) as compared to the control. Small and large particles had little effect on the methane production rate but did result in an extended lag phase which led to significantly lower cumulative methane production at the end of the incubation period. These results suggest that magnetite produces a clear and positive effect on methane production only when an active and balanced microbial community is present in the anaerobic digester. It is concluded that, (i) the effect of magnetite particle size on increasing the methane production rate and reducing lag phase duration is strongly influenced by the initial metabolic state of the microbial consortium, and (ii) the particle size would positively affect the methane production if it is provided within the nanometer size range.
Abstract: Yeast cells are generally used as a model system of eukaryotes due to their complex genetic structure, rapid growth ability in optimum conditions, easy replication and well-defined genetic system properties. Thus, yeast cells increased the knowledge of the principal pathways in humans. During fermentation, carbohydrates (hexoses and pentoses) degrade into some toxic by-products such as 5-hydroxymethylfurfural (5-HMF or HMF) and furfural. HMF influences the ethanol yield, and ethanol productivity; it interferes with microbial growth and is considered as a potent inhibitor of bioethanol production. In this study, yeast single cell behavior under HMF application was monitored by using a continuous flow single phase microfluidic platform. Microfluidic device in operation is fabricated by hot embossing and thermo-compression techniques from cyclo-olefin polymer (COP). COP is biocompatible, transparent and rigid material and it is suitable for observing fluorescence of cells considering its low auto-fluorescence characteristic. The response of yeast cells was recorded through Red Fluorescent Protein (RFP) tagged Nop56 gene product, which is an essential evolutionary-conserved nucleolar protein, and also a member of the box C/D snoRNP complexes. With the application of HMF, yeast cell proliferation continued but HMF slowed down the cell growth, and after HMF treatment the cell proliferation stopped. By the addition of fresh nutrient medium, the yeast cells recovered after 6 hours of HMF exposure. Thus, HMF application suppresses normal functioning of cell cycle but it does not cause cells to die. The monitoring of Nop56 expression phases of the individual cells shed light on the protein and ribosome synthesis cycles along with their link to growth. Further computational study revealed that the mechanisms underlying the inhibitory or inductive effects of HMF on growth are enriched in functional categories of protein degradation, protein processing, DNA repair and multidrug resistance. The present microfluidic device can successfully be used for studying the effects of inhibitory agents on growth by single cell tracking, thus capturing cell to cell variations. By metabolic engineering techniques, engineered strains can be developed, and the metabolic network of the microorganism can thus be manipulated such that chemical overproduction of target metabolite is achieved along with the maximum growth/biomass yield.
Abstract: Biscuit is one of the most consumed cereal foods in Nigeria and research has shown that locally available tropical crops like cassava, sweet potato can be made into flour and used in the production of biscuits and other pastries. This study investigates some quality parameters in biscuits made from blends of wheat, cassava and carrot flour. The values of result of samples increased with increasing percentage substitution of cassava and carrot flour in some quality parameter like fiber, ash, gluten content, and carbohydrate. The protein content reduced significantly (P < 0.05) with increasing percentage substitution of cassava and carrot flour which ranged from 14.80% to 11.80% compared with the control sample which had 15.60%. There was a recorded significant increase (P < 0.05) in some mineral composition such as calcium, magnesium, sodium, iron, phosphorus, and vitamin A and C composition as the percentage substitution of cassava and carrot flour increased. During storage stability test, samples stored in the fridge and freezer were found to be the best storage location to preserve the sensory attributes and inhibit microbial growth when compared with storage under the sun and on the shelf. Biscuit made with blends of wheat, cassava and carrot flour can therefore serve as an alternative to biscuits made from 100% wheat flour, as they are richer in vitamin A, vitamin C, carbohydrate, dietary fiber and some essential minerals.
Abstract: Tannase (tannin acyl hydrolase, E.C.3.1.1.20) is an
important hydrolysable enzyme with innumerable applications and
industrial potential. In the present study, a kinetic model has been
developed for the batch fermentation used for the production of
tannase by A.flavus MTCC 3783. Maximum tannase activity of
143.30 U/ml was obtained at 96 hours under optimum operating
conditions at 35oC, an initial pH of 5.5 and with an inducer tannic
acid concentration of 3% (w/v) for a fermentation period of 120
hours. The biomass concentration reaches a maximum of 6.62 g/l at
96 hours and further there was no increase in biomass concentration
till the end of the fermentation. Various unstructured kinetic models
were analyzed to simulate the experimental values of microbial
growth, tannase activity and substrate concentration. The Logistic
model for microbial growth , Luedeking - Piret model for production
of tannase and Substrate utilization kinetic model for utilization of
substrate were capable of predicting the fermentation profile with
high coefficient of determination (R2) values of 0.980, 0.942 and
0.983 respectively. The results indicated that the unstructured models
were able to describe the fermentation kinetics more effectively.
Abstract: The quality and condition of perishable products
delivered to the market and their subsequent selling prices are
directly affected by the care taken during harvesting and handling.
Mechanical injury, in fact, occurs at all stages, from pre-harvest
operations through post-harvest handling, packing and transport to
the market. The main implications of this damage are the reduction of
the product’s quality and economical losses related to the shelf life
diminution. For most perishable products, the shelf life is relatively
short and it is typically dictated by microbial growth related to the
application of dynamic and static loads during transportation. This
paper presents the correlation between vibration levels and
microbiological growth on strawberries and woodland strawberries
and detects the presence of volatile organic compounds (VOC) in
order to develop an intelligent logistic unit capable of monitoring
VOCs using a specific sensor system. Fresh fruits were exposed to
vibrations by means of a vibrating table in a temperature-controlled
environment. Microbiological analyses were conducted on samples,
taken at different positions along the column of the crates. The values
obtained were compared with control samples not exposed to
vibrations and the results show that different positions along the
column influence the development of bacteria, yeasts and filamentous
fungi.
Abstract: Eucalyptus species are well reputed for their
traditional use in Asia as well as in other parts of the world; therefore,
the present study was designed to investigate the antimicrobial and
antioxidant activities associated with essential oils from different
Eucalyptus species. Essential oils from the leaves of six Eucalyptus
species, including: Eucalyptus woodwardi, Eucalyptus stricklandii,
Eucalyptus salubris, Eucalyptus sargentii, Eucalyptus torquata and
Eucalyptus wandoo were separated by hydrodistillation and dried
over anhydrous sodium sulphate. DPPH, ferric reducing antioxidant
power, and hydroxyl radical scavenging activity assays were carried
out to evaluate the antioxidant potential of the oils. The results
indicate that examined oils exhibit substantial antioxidant activities
relative to ascorbic acid. Previously, these oils were evaluated for
their antimicrobial activities, against wide range of bacterial and
fungal strains, and they were shown to possess significant
antimicrobial activities. In this study, further investigation into the
growth kinetics of oil-treated microbial cultures was conducted. The
results clearly demonstrate that the microbial growth was markedly
inhibited when treated with sub-MIC concentrations of the oils.
Taken together, the results obtained indicate a high potential of the
examined essential oils as bioactive oils, for nutraceutical and
medical applications, possessing significant antioxidant and anti
microbial activities.
Abstract: Mastitis is one of the most economic disease affecting dairy cows worldwide. Its classic diagnosis using bacterial culture and biochemical findings is a difficult and prolonged method. In this research, using of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) permitted identification of different microorganisms with high accuracy and rapidity (only 24 hours for microbial growth and analysis). During the application of MALDI-TOF MS, one hundred twenty strains of Staphylococcus and Streptococcus species isolated from milk of cows affected by clinical and subclinical mastitis were identified, and the results were compared with those obtained by traditional methods as API and VITEK 2 Systems. 37 of totality 39 strains (~95%) of Staphylococcus aureus (S. aureus) were exactly detected by MALDI TOF MS and then confirmed by a nuc-based PCR technique, whereas accurate identification was observed in 100% (50 isolates) of the coagulase negative staphylococci (CNS) and Streptococcus agalactiae (31 isolates). In brief, our results demonstrated that MALDI-TOF MS is a fast and truthful technique which has the capability to replace conventional identification of several bacterial strains usually isolated in clinical laboratories of microbiology.
Abstract: In the present study, effect of critical medium components (a total of fifteen components) on ethanol production from waste cashew apple juice (CAJ) using yeast Saccharomyces diasticus was studied. A statistical response surface methodology
(RSM) based Plackett-Burman Design (PBD) was used for the design of experiments. The design contains a total of 32 experimental trails. The effect of medium components on ethanol was studied at two different levels such as low concentration level (-) and high concentration levels (+). The dependent variables selected in this study were ethanol concentration (g/L) and cellmass concentration (g/L). Data obtained from RSM on ethanol production were subjected to analysis of variance (ANOVA). In general, initial substrate concentration significantly influenced the microbial growth and product formation. Of the medium components evaluated, CAJ concentration, yeast extract, (NH4)2SO4, and malt extract showed significant effect on ethanol fermentation. A second-order polynomial model was used to predict the experimental data and the model fitted the data with a high correlation coefficient (R2 > 0.98).
Maximum ethanol (15.3 g/L) and biomass (6.4 g/L) concentrations
were obtained at the optimum medium composition and at optimum
condition (temperature-30°C; initial pH-6.8) after 72 h fermentation
using S.diasticus.
Abstract: The shelf life of fish was extended using disinfection
properties of ozone. For this purpose, Trout specimens were exposed
to ozone in the aqueous media for two hours and their microbial
growth and biochemical properties were measured over time.
Microbial growth of ozone treated fish was significantly slower than
control sample, resulting in lower counts of bacteria. According to
the biochemical tests; ozone treatment had no negative effects on fat,
protein and humidity of fish. Peroxide and TVN (Total Volatile
Nitrogen) measurements showed that treatment by ozone increased
the trout shelf life from 4 days to 6 days. According to the sensory
analysis, no changes were observed in color or flavor of the ozone
treated trout.
Abstract: The effects of seawater and slurry ice bleeding methods on the sensory, microbiological and chemical quality changes of cod fillets during chilled storage were examined in this study. The results from sensory evaluation showed that slurry ice bleeding method prolonged the shelf life of cod fillets up to 13-14 days compared to 10-11 days for fish bled in seawater. Slurry ice bleeding method also led to a slower microbial growth and biochemical developments, resulting lower total plate count (TPC), H2S-producing bacteria count, total volatile basic nitrogen (TVB-N), trimethylamine (TMA), free fatty acid (FFA) content and higher phospholipid content (PL) compared to those of samples bled in seawater. The results of principle component analysis revealed that TPC, H2S-producing bacteria, TVB-N, TMA and FFA were in significant correlation. They were also in negative correlation with sensory evaluation (Torry score), PL and water holding capacity (WHC).
Abstract: Acid rain occurs when sulphur dioxide (SO2) and
nitrogen oxides (Nox) gases react in the atmosphere with water,
oxygen, and other chemicals to form various acidic compounds. The
result is a mild solution of sulfuric acid and nitric acid. Soil has a
greater buffering capacity than aquatic systems. However excessive
amount of acids introduced by acid rains may disturb the entire soil
chemistry. Acidity and harmful action of toxic elements damage
vegetation while susceptible microbial species are eliminated. In
present study, the effects of simulated sulphuric acid and nitric acid
rains were investigated on crop Glycine max. The effect of acid rain
on change in soil fertility was detected in which pH of control sample
was 6.5 and pH of 1%H2SO4 and 1%HNO3 were 3.5. Nitrogen nitrate
in soil was high in 1% HNO3 treated soil & Control sample.
Ammonium nitrogen in soil was low in 1% HNO3 & H2SO4 treated
soil. Ammonium nitrogen was medium in control and other samples.
The effect of acid rain on seed germination on 3rd day of germination
control sample growth was 7 cm, 0.1% HNO3 was 8cm, and 0.001%
HNO3 & 0.001% H2SO4 was 6cm each. On 10th day fungal growth
was observed in 1% and 0.1%H2SO4 concentrations, when all plants
were dead. The effect of acid rain on crop productivity was
investigated on 3rd day roots were developed in plants. On12th day
Glycine max showed more growth in 0.1% HNO3, 0.001% HNO3 and
0.001% H2SO4 treated plants growth were same as compare to control
plants. On 20th day development of discoloration of plant pigments
were observed on acid treated plants leaves. On 38th day, 0.1, 0.001%
HNO3 and 0.1, 0.001% H2SO4 treated plants and control plants were
showing flower growth. On 42th day, acid treated Glycine max variety
and control plants were showed seeds on plants. In Glycine max
variety 0.1, 0.001% H2SO4, 0.1, 0.001% HNO3 treated plants were
dead on 46th day and fungal growth was observed. The toxicological
study was carried out on Glycine max plants exposed to 1% HNO3
cells were damaged more than 1% H2SO4. Leaf sections exposed to
0.001% HNO3 & H2SO4 showed less damaged of cells and
pigmentation observed in entire slide when compare with control
plant. The soil analysis was done to find microorganisms in HNO3 &
H2SO4 treated Glycine max and control plants. No microorganism
growth was observed in 1% HNO3 & H2SO4 but control plant showed
microbial growth.
Abstract: This paper presents an integrated knowledge-based
approach to multi-scale modeling of aquatic systems, with a view to
enhancing predictive power and aiding environmental management
and policy-making. The basic phases of this approach have been
exemplified in the case of a bay in Saronicos Gulf (Attiki, Greece).
The results showed a significant problem with rising phytoplankton
blooms linked to excessive microbial growth, arisen mostly due to
increased nitrogen inflows; therefore, the nitrification/denitrification
processes of the benthic and water column sub-systems have
provided the quality variables to be monitored for assessing
environmental status. It is thereby demonstrated that the proposed
approach facilitates modeling choices and implementation option
decisions, while it provides substantial support for knowledge and
experience capitalization in long-term water management.
Abstract: The present study has been taken to explore the
screening of in vitro antimicrobial activities of D-galactose-binding
sponge lectin (HOL-30). HOL-30 was purified from the marine
demosponge Halichondria okadai by affinity chromatography. The
molecular mass of the lectin was determined to be 30 kDa with a
single polypeptide by SDS-PAGE under non-reducing and reducing
conditions. HOL-30 agglutinated trypsinized and glutaraldehydefixed
rabbit and human erythrocytes with preference for type O
erythrocytes. The lectin was subjected to evaluation for inhibition of
microbial growth by the disc diffusion method against eleven human
pathogenic gram-positive and gram-negative bacteria. The lectin
exhibited strong antibacterial activity against gram-positive bacteria,
such as Bacillus megaterium and Bacillus subtilis. However, it did
not affect against gram-negative bacteria such as Salmonella typhi
and Escherichia coli. The largest zone of inhibition was recorded of
Bacillus megaterium (12 in diameter) and Bacillus subtilis (10 mm in
diameter) at a concentration of the lectin (250 μg/disc). On the other
hand, the antifungal activity of the lectin was investigated against six
phytopathogenic fungi based on food poisoning technique. The lectin
has shown maximum inhibition (22.83%) of mycelial growth of
Botrydiplodia theobromae at a concentration of 100 μg/mL media.
These findings indicate that the lectin may be of importance to
clinical microbiology and have therapeutic applications.
