Abstract: Chlorophytum borivillianum root extract (CBE) was chosen as a reducing agent to fabricate silver nanoparticles with the aim of studying its radioprotective efficacy. The formation of synthesized nanoparticles was characterized by UV–visible analysis (UV–vis), Fourier transform infra-red (FT-IR), Transmission electron microscopy (TEM), Scanning electron microscope (SEM). TEM analysis showed particles size in the range of 20-30 nm. For this study, Swiss albino mice were selected from inbred colony and were divided into 4 groups: group I- control (irradiated-6 Gy), group II- normal (vehicle treated), group III- plant extract alone and group IV- CB-AgNPs (dose of 50 mg/kg body wt./day) administered orally for 7 consecutive days before irradiation to serve as experimental. CB-AgNPs pretreatment rendered significant increase in body weight and testes weight at various post irradiation intervals in comparison to irradiated group. Supplementation of CB-AgNPs reversed the adverse effects of gamma radiation on biochemical parameters as it notably ameliorated the elevation in lipid peroxidation and decline in glutathione concentration in testes. These observations indicate the radio-protective potential of CB-AgNPs in testicular constituents against gamma irradiation in mice.
Abstract: Type 2 Diabetes (T2DM) and Alzheimer's disease (AD) are two main health problems influencing millions of people in the world. Neuron loss and synaptic impairment that interfere with cognition and memory cause for the behavioral indications of AD. While it is now accepted that insulin has central neuromodulatory purpose, it was contemplated for many years that brain is insusceptible to insulin, involving its function in memory and learning, which are impaired in AD. The common characteristics of both AD and T2D are impaired insulin signaling, oxidative stress, the excitation of inflammatory pathways and unqualified glucose metabolism. This review summarizes how the recognition of these mechanisms may lead to the development of alternative therapeutic approaches. Abstract: Oxidative stress associated with semen cryopreservation may result in lipid peroxidation (LPO), DNA damage and apoptosis, leading to decreased sperm motility and fertilization ability. Curcumin (CUR), a natural phenol isolated from Curcuma longa Linn. has been presented as a possible supplement for a more effective semen cryopreservation because of its antioxidant properties. This study focused to evaluate the effects of CUR on selected oxidative stress parameters in cryopreserved bovine semen. 20 bovine ejaculates were split into two aliquots and diluted with a commercial semen extender containing CUR (50 μmol/L) or no supplement (control), cooled to 4 °C, frozen and kept in liquid nitrogen. Frozen straws were thawed in a water bath for subsequent experiments. Abstract: Tartrazine is an organic azo dyes food additive widely used in foods, drugs, and cosmetics. The present study aimed to investigate the toxic effects of tartrazine on kidneys and liver biomarkers in addition to the investigation of oxidative stress and change of histopathological structure of liver and kidneys in 30 male rats. Tartrazine was orally administrated daily at dose 200 mg/ kg bw (1/ 10 LD50) for sixty days. Serum and tissue samples were collected at the end of the experiment to investigate the underlying mechanism of tartrazine through assessment oxidative stress (Glutathione (GSH), Superoxide dismutase (SOD) and malondialdehyde (MDA) and biochemical markers (alanine aminotransferase (ALT), aspartate aminotransferase (AST), Total protein and Urea). Liver and kidneys tissue were collected and preserved in 10% formalin for histopathological examination. The obtained values were statistically analyzed by one way analysis of variance (ANOVA) followed by multiple comparison test. Biochemical analysis revealed that tartrazine induced significant increase in serum ALT, AST, total protein, urea level compared to control group. Tartrazine showed significant decrease in liver GSH and SOD where their values when compared to control group. Tartrazine induced increase in liver MDA compared to control group. Histopathology of the liver showed diffuse vacuolar degeneration in hepatic parenchyma, the portal area showed sever changes sever in hepatoportal blood vessels and in the bile ducts. The kidneys showed degenerated tubules at the cortex together with mononuclear leucocytes inflammatory cells infiltration. There is perivascular edema with inflammatory cell infiltration surrounding the congested and hyalinized vascular wall of blood vessel. The present study indicates that the subchronic effects of tartrazine have a toxic effect on the liver and kidneys together with induction of oxidative stress by formation of free radicals. Therefore, people should avoid the hazards of consuming tartrazine.
Abstract: Cisplatin (CIS) is one of the most effective an anticancer drug and also toxic to cells by activating oxidative stress. Oleuropein (OLE) has key role against oxidative stress in mammalian cells, but the role of this antioxidant in the toxicity of CIS remains unknown. The aim of the present study was to investigate the efficacy of OLE on CIS-induced liver damages in male rats. With this aim, male Sprague Dawley rats were randomly assigned to one of eight groups: Control group; the group treated with 7 mg/kg/day CIS; the groups treated with 50, 100 and 200 mg/kg/day OLE (i.p.); and the groups treated with OLE for three days starting at 24 h following CIS injection. After 4 days of injections, serum was provided to assess the blood AST, ALT and LDH values. The liver tissues were removed for histological, biochemical (TAC, TOS and MDA) and genotoxic evaluations. In the CIS treated group, the whole liver tissue showed significant histological changes. Also, CIS significantly increased both the incidence of oxidative stress and the induction of 8-hydroxy-deoxyguanosine (8-OH-dG). Moreover, the rats taking CIS have abnormal results on liver function tests. However, these parameters reached to the normal range after administration of OLE for 3 days. Finally, OLE demonstrated an acceptable high potential and was effective in attenuating CIS-induced liver injury. In this trial, the 200 mg/kg dose of OLE firstly appeared to induce the most optimal protective response.
Abstract: Background: Uncontrolled inflammation may cause serious inflammatory diseases if left untreated. Non-steroidal anti-inflammatory drug (NSAIDs) is commonly used to inhibit pro-inflammatory enzymes, thus, reduce inflammation. However, long term administration of NSAIDs leads to various complications. Medicinal plants are getting more attention as it is believed to be more compatible with human body. One of them is a flavonoid-containing medicinal plants, Strobilanthes crispus which has been traditionally claimed to possess anti-inflammatory and antioxidant activities. Nevertheless, its anti-inflammatory activities are yet to be scientifically documented. Objectives: This study aimed to examine the anti-inflammatory activity of S. crispus by investigating its effects on intracellular oxidative stress and prostaglandin E2 (PGE2) levels. Materials and Methods: In this study, the Maximum Non-toxic Dose (MNTD) of methanol extract of both leaves and stems of S. crispus was first determined using 3-(4,5-dimethylthiazolyl-2)-2,5-diphenytetrazolium Bromide (MTT) assay. The effects of S. crispus extracts at MNTD and half MNTD (½MNTD) on intracellular ROS as well as PGE2 levels in 1.0 µg/mL LPS-stimulated RAW 264.7 macrophages were then be measured using DCFH-DA and a competitive enzyme immunoassay kit, respectively. Results: The MNTD of leaf extract was determined as 700µg/mL while for stem was as low as 1.4µg/mL. When LPS-stimulated RAW 264.7 macrophages were subjected to the MNTD of S. crispus leaf extract, both intracellular ROS and PGE2 levels were significantly reduced. In contrast, stem extract at both MNTD and ½MNTD did not significantly reduce the PGE2 level, but significantly increased the intracellular ROS level. Conclusion: The methanol leaf extract of S. crispus may possess anti-inflammatory properties as it is able to significantly reduce the intracellular ROS and PGE2 levels of LPS-stimulated cells. Nevertheless, further studies such as investigating the interleukin, nitric oxide and cytokine tumor necrosis factor-α (TNFα) levels has to be conducted to further confirm the anti-inflammatory properties of S. crispus.
Abstract: Intense oxidative stress, increased glycated hemoglobin and mineral imbalance represent risk factors for complications in diabetic patients. Cardiovascular complications are most common in these patients, including nephropathy. This study was conducted in 2015 at the Procardia Laboratory in Tîrgu Mureș, Romania on 40 type 2 diabetic adults. Routine biochemical tests were performed on the Konleab 20XTi analyzer (serum glucose, total cholesterol, LDL and HDL cholesterol, triglyceride, creatinine, urea). We also measured serum uric acid, magnesium and calcium concentration by photometric procedures, potassium, sodium and chloride by ion selective electrode, and chromium by atomic absorption spectrometry in a group of patients. Glycated hemoglobin (HbA1c) dosage was made by reflectometry. Urine analysis was performed using the HandUReader equipment. The level of oxidative stress was measured by serum malondialdehyde dosage using the thiobarbituric acid reactive substances method. MDRD (Modification of Diet in Renal Disease) formula was applied for calculation of creatinine-derived glomerular filtration rate. GraphPad InStat software was used for statistical analysis of the data. The diabetic subject included in the study presented high MDA concentrations, showing intense oxidative stress. Abstract: The main cause of Alzheimer disease (AD) was
believed to be mainly due to the accumulation of free radicals owing
to oxidative stress (OS) in brain tissue. The mechanism of the
neurotoxicity of Aluminum chloride (AlCl3) induced AD in
hippocampus Albino wister rat brain tissue, the curative & the
protective effects of Lipidium sativum group (LS) water extract were
assessed after 8 weeks by attenuated total reflection spectroscopy
ATR-IR and histologically by light microscope. ATR-IR results
revealed that the membrane phospholipid undergo free radical
attacks, mediated by AlCl3, primary affects the polyunsaturated fatty
acids indicated by the increased of the olefinic -C=CH sub-band area
around 3012 cm-1 from the curve fitting analysis. The narrowing in
the half band width (HBW) of the sνCH2 sub-band around 2852 cm-1
due to Al intoxication indicates the presence of trans form fatty acids
rather than gauch rotomer. The degradation of hydrocarbon chain to
shorter chain length, increasing in membrane fluidity, disorder, and
decreasing in lipid polarity in AlCl3 group indicated by the detected
changes in certain calculated area ratios compared to the control.
Administration of LS was greatly improved these parameters
compared to the AlCl3 group. Al influences the Aβ aggregation and
plaque formation, which in turn interferes to and disrupts the
membrane structure. The results also showed a marked increase in
the β-parallel and antiparallel structure, that characterize the Aβ
formation in Al-induced AD hippocampal brain tissue, indicated by
the detected increase in both amide I sub-bands around 1674, 1692
cm-1. This drastic increase in Aβ formation was greatly reduced in the
curative and protective groups compared to the AlCl3 group and
approached nearly the control values. These results supported too by
the light microscope. AlCl3 group showed significant marked
degenerative changes in hippocampal neurons. Most cells appeared
small, shrieked and deformed. Interestingly, the administration of LS
in curative and protective groups markedly decreases the amount of
degenerated cells compared to the non-treated group. In addition, the
intensity of congo red stained cells was decreased. Hippocampal
neurons looked more/or less similar to those of control. This study showed a promising therapeutic effect of Lipidium
sativum group (LS) on AD rat model that seriously overcome the
signs of oxidative stress on membrane lipid and restore the protein
misfolding.
Abstract: Sodium formate is the chemical substance used for
food additive. Catalase is the important antioxidative enzyme in
protecting the cell from oxidative damage by reactive oxygen species
(ROS). The resultant level of oxidative stress in sodium formatetreated
lymphocytes was investigated. The sodium formate
concentrations of 0.05, 0.1, 0.2, 0.4 and 0.6 mg/mL were treated in
human lymphocytes for 12 hours. After 12 treated hours, catalase
activity change was measured in sodium formate-treated
lymphocytes. The results showed that the sodium formate
concentrations of 0.4 and 0.6 mg/mL significantly decreased catalase
activities in lymphocytes (P < 0.05). The change of catalase activity
in sodium formate-treated lymphocytes may be the oxidative damage
marker for detect sodium formate exposure in human.
Abstract: The potential neuroprotective effect of Phyllantus
nuriri against Fe2+ and sodium nitroprusside (SNP) induced oxidative
stress in mitochondria of rats brain was evaluated. Cellular viability
was assessed by MTT reduction, reactive oxygen species (ROS)
generation was measured using the probe 2,7-dichlorofluoresce
indiacetate (DCFH-DA). Glutathione content was measured using
dithionitrobenzoic acid (DTNB). Fe2+ (10μM) and SNP (5μM)
significantly decreased mitochondrial activity, assessed by MTT
reduction assay, in a dose-dependent manner, this occurred in parallel
with increased glutathione oxidation, ROS production and lipid
peroxidation end-products (thiobarbituric acid reactive substances,
TBARS). The co-incubation with methanolic extract of Phyllantus
nuriri (10-200 μg/ml) reduced the disruption of mitochondrial
activity, gluthathione oxidation, ROS production as well as the
increase in TBARS levels caused by both Fe2+ and SNP in a dose
dependent manner. HPLC analysis of the extract revealed the
presence of gallic acid (20.540.01), caffeic acid (7.930.02), rutin
(25.310.05), quercetin (31.280.03) and kaemferol (14.360.01).
This result suggests that these phytochemicals account for the
protective actions of P. niruri against Fe2+ and SNP -induced
oxidative stress. Our results show that P. nuriri consist important
bioactive molecules in the search for an improved therapy against the
deleterious effects of Fe2+, an intrinsic producer of reactive oxygen
species (ROS), that leads to neuronal oxidative stress and
neurodegeneration.
Abstract: Diminished antioxidant defense or increased
production of reactive oxygen species in the biological system can
result in oxidative stress which may lead to various
neurodegenerative diseases including Alzheimer’s disease (AD).
Microglial activation also contributes to the progression of AD by
producing several proinflammatory cytokines, nitric oxide (NO) and
prostaglandin E2 (PGE2). Oxidative stress and inflammation have
been reported to be possible pathophysiological mechanisms
underlying AD. In addition, the cholinergic hypothesis postulates that
memory impairment in patient with AD is also associated with the
deficit of cholinergic function in the brain. Although a number of
drugs have been approved for the treatment of AD, most of these
synthetic drugs have diverse side effects and yield relatively modest
benefits. Marine algae have great potential in pharmaceutical and
biomedical applications as they are valuable sources of bioactive
properties such as anticoagulation, antimicrobial, antioxidative,
anticancer and anti-inflammatory. Hence, this study aimed to provide

Abstract: In this investigation, we have evaluated the effects of
arsenic trioxide on hepatic function in pregnant and lactating Swiss
albino mice and their suckling pups. Experiments were carried out on
female mice given 175 ppm As2O3 in their drinking water from the
14th day of pregnancy until day 14 after delivery. Our results showed
a significant decrease in plasma levels of total protein and albumin,
cholesterol and triglyceride in As2O3 treated mice and their pups. The
hyperbilirubinemia and the increased plasma total alkaline
phosphatase activity suggested the presence of cholestasis.
Transaminase activities as well as lactate deshydrogenase activity in
plasma, known as biomarkers of hepatocellular injury, were elevated
indicating hepatic cells’ damage after treatment with As2O3.
Exposure to arsenic led to an increase of liver thiobarbituric acid
reactive substances level along with a concomitant decrease in the
activities of superoxide dismutase, catalase and glutathione

Abstract: This study was conducted to investigate the effect of
the antioxidant activity of germinated African Yam Bean (AYB) on
oxidative stress markers in alloxan induced diabetic rat. Rats were
randomized into three groups; control, diabetic and germinated AYB
– treated diabetic rats. The Total phenol and flavonoid content and
DPPH radical scavenging activity before and after germination were
investigated. The glucose level, lipid peroxidation and reduced
glutathione of the animals were also determined using standard
technique for four weeks. Germination increased the total phenol,
flavonoid and antioxidant activity of AYB extract by 19.14%,

Abstract: The presence of endocrine-disrupting compounds,
such as bisphenol A (BPA), in the environment can cause serious
health problems. However, there are controversial opinions. This
study investigated the reproductive, metabolic, oxidative and
immunologic-disrupting effects of bisphenol A in male rabbits.
Rabbits were divided into five groups. The first four rabbit groups
were administered oral BPA (1, 10, 50, or 100 mg/kg/day) for ten
weeks. The fifth group was administered corn oil as the vehicle. BPA
significantly decreased serum testosterone, estradiol and the free
androgen index (FAI) and significantly increased sex hormone
binding globulin (SHBG) compared with the placebo group. The
higher doses of BPA showed a significant decrease in follicular
stimulating hormone (FSH) and luteinizing hormone (LH). A
significant increase in blood glucose levels was identified in the BPA
groups. The non-significant difference in insulin levels is a novel
finding. The cumulative testicular toxicity of BPA was clearly
demonstrated by the dose-dependent decrease in absolute testes
weight, primary measures of semen quality and a significant increase
in testicular malonaldehyde (MDA). Moreover, BPA significantly
decreased total antioxidant capacity (TAC) and significantly
increased immunoglobulin G (IgG) at the highest concentration. Our
results suggest that BPA, especially at higher doses, is associated
with many adverse effects on metabolism, oxidative stress, immunity,
sperm quality and markers of androgenic action.
Abstract: Fluoroquinolones are a group of antibiotics widely used because of their broad spectrum activity against both Gram-positive and Gram-negative bacteria. In this study, ciprofloxacin and levofloxacin were administered to rats at therapeutic doses to evaluate their effects on plasma arylesterase activity, as well as, on hepatic advanced oxidized protein products (AOPPs) and malondialdehyde (MDA) levels, as measures of oxidative stress. Ciprofloxacin (80 mg/kg body weight) and levofloxacin (40 mg/kg body weight) were administered to male albino rats for 7 and 14 days. The data obtained demonstrated that plasma arylesterase activity was significantly decreased by both drugs with ciprofloxacin administration inhibiting the activity by 29% and 30% while Levofloxacin treatment resulted in 35% and 30% inhibition, after 7 and 14 days treatment respectively. Hepatic AOPP and MDA levels were both elevated by these antibiotics. This study supplies further evidence that fluoroquinolones at therapeutic doses promote oxidative stress.
Abstract: Alongside with antioxidant, pro-oxidant activity is also observed in phytochemical compounds. In the study, Ficus odorata, an endemic medicinal plant in the Philippines, was screened for the potential medical application of its pro-oxidant activity.
Phytochemical screening revealed the presence of terpenes, glycosides and phenolic acids. The crude extract was found to contain low gallic acid and quercetin equivalence. The TLC chromatogram of the crude extract showed that none of the 11 spots obtained has antioxidant activity nor correspond to gallic acid and quercetin standards. Experiments showed that the crude extract has stimulatory activity towards DPPH radicals, hydrogen peroxide, hydroxyl radicals, superoxide anions and nitric oxide. Moreover, the extract exhibited a low ferric reducing power.
The prooxidant activity was evident in the crude ethanolic leaf extract of F. odorata, which may provide a better understanding of the plant’s pharmacological importance in the prevention of diseases.
Abstract: Aim- Study was undertaken to investigate the effect of altered behavioral condition like depression on various oxidative stress markers and biochemical parameters in rats. Methods- Rats were subjected for short (21 days) and long term (84 days) social isolation; the rats displayed an increase in depression on force swim test relative to control. Various markers of oxidative stress like lipid per oxidation (LPO), reduced glutathione (GSH), Supers oxide dismutase (SOD), catalase (CAT) and biochemical parameters like SGOT, SGPT, and blood glucose were determined. Results- There was significant increase in the level of LPO and decrease in the levels of GSH, SOD and CAT after long term isolation. Biochemical parameters were significantly altered after social isolation. Conclusion- Increased oxidative stress in depression which may leads to alteration of biochemical parameters.
Abstract: Haemodialysis (HD) is a procedure saving patient lives around the world, unfortunately it brings numerous complications. Oxidative stress is one of the major factors which lead to erythrocytes destruction during extracorporeal circulation. Repeated HD procedures destroy blood elements and the organism is not able to keep up with their production. 30 HD procedures on healthy sheep were performed to evaluate effects of such treatment. Oxidative stress study was performed together with an analysis of basic blood parameters and empirical assessment of dialyzer condition after the procedure. A reversible decline in absolute leukocyte count, during first 30 min of HD, was observed. Blood clots were formed in the area of the blood inlet and outlet of the dialyzer. Our results are consistent with outcomes presented throughout the literature specifically with respect to the effects observed in humans and will provide a basis to evaluate methods for blood protection during haemodialysis.
Abstract: Oxidative stress and overwhelming free radicals
associated with diabetes mellitus are likely to be linked with
development of certain complication such as retinopathy,

Olive leaf (Oleaeuropaea), has been endowed with many beneficial
and health promoting properties mostly linked to its antioxidant
activity. This study aimed to evaluate the significance of
supplementation of Olive leaves extract (OLE) in reducing oxidative
stress, hyperglycemia and hyperlipidemia in Sterptozotocin (STZ)-
induced diabetic rats. After induction of diabetes, a significant rise in
plasma glucose, lipid profiles except High density lipoproteincholestrol
(HDLc), malondialdehyde (MDA) and significant decrease
of plasma insulin, HDLc and Plasma reduced glutathione GSH as
well as alteration in enzymatic antioxidants was observed in all
diabetic animals. During treatment of diabetic rats with 0.5g/kg body
weight of Olive leaves extract (OLE) the levels of plasma (MDA)
,(GSH), insulin, lipid profiles along with blood glucose and
erythrocyte enzymatic antioxidant enzymes were significantly
restored to establish values that were not different from normal
control rats. Untreated diabetic rats on the other hand demonstrated
persistent alterations in the oxidative stress marker (MDA), blood
glucose, insulin, lipid profiles and the antioxidant parameters. These
results demonstrate that OLE may be of advantage in inhibiting
hyperglycemia, hyperlipidemia and oxidative stress induced by
diabetes and suggest that administration of OLE may be helpful in
the prevention or at least reduced of diabetic complications
associated with oxidative stress.
Abstract: Hyperglycemia-mediated accumulation of advanced glycation end-products (AGEs) play a pivotal role in the development of diabetic complications by inducing inflammation. In the present study, we evaluated the possible effects of water/ethanol (1/1, v/v) extracts (WEE) and its fractions from Canarium album Raeusch. (Chinese olive) which is a fruit used on AGEs-stimulated oxidative stress and inflammation in monocytes and vascular endothelial cells. Co-incubation of EA.hy926 endothelial cells with WEE and its fractions for 24h resulted in a significant decrease of monocyte–endothelial cell adhesion, the expression of ICAM-1, generation of intracellular ROS and depletion of GSH induced by AGEs. Chinese olive fruit extracts also reduced the expression of pro-inflammatory mediates, such as TNF-α, IL-1β and IL-6 in THP-1 cells. These findings suggested that Chinese olive fruit was able to protect vascular endothelium from dysfunction induced by AGEs.
