Abstract: The identification of lipid and soluble sugar components in flour samples of different cultivars belonging to common oat species (Avena sativa L.) was performed: spring oat, winter oat and hulless oat. Fatty acids were extracted from flour samples with n-hexane, and derivatized into volatile methyl esters, using TMSH (trimethylsulfonium hydroxide in methanol). Soluble sugars were then extracted from defatted and dried samples of oat flour with 96% ethanol, and further derivatized into corresponding TMS-oximes, using hydroxylamine hydrochloride solution and BSTFA (N,O-bis-(trimethylsilyl)-trifluoroacetamide). The hexane and ethanol extracts of each oat cultivar were analyzed using GC-MS system. Lipid and simple sugar compositions are very similar in all samples of investigated cultivars. Chemometric tool was applied to numeric values of automatically integrated surface areas of detected lipid and simple sugar components in their corresponding derivatized forms. Hierarchical cluster analysis shows a very high similarity between the investigated flour samples of oat cultivars, according to the fatty acid content (0.9955). Moderate similarity was observed according to the content of soluble sugars (0.50). These preliminary results support the idea of establishing methods for oat flour authentication, and provide the means for distinguishing oat flour samples, regardless of the variety, from flour samples made of other cereal species, just by lipid and simple sugar profile analysis.
Abstract: The effect of calcium salts on the storage stability and on the quality attributes of both fresh and processed product (guava powder) of white flesh guavas (var ‘Allahabad safeda’) was studied. The pulp behavioral studies of fully ripened guava fruits indicated that fruits pretreated with 3% and 4.5% calcium chloride had the least viscosity. The guava pulp powder using spray drying technique was developed and its storage stability and the moisture sorption studies were carried out for product quality evaluation at normal storage condition (27°C; 65%RH). Results revealed that powder obtained from 3% calcium chloride pretreated guavas was found to be at par with the powder obtained from control guavas after 90 days of normal storage. Studies on microbiological quality of guava pulp powder indicated that among the treatments powder obtained from guava fruit pretreated with 3% calcium chloride to be the most effective through restricting microbial counts of total plate count, yeast, mold, Staphylococcus and E. coli below their permissible limit. Moisture sorption studies of guava powder revealed that foil laminate 12μm PET/9 μm foil/38-40 μm is the most suitable packaging material recommended.
Abstract: This paper determines the presence and levels of phthalates in sachet and borehole water source in some parts of Delta State, Nigeria. Sachet and borehole water samples were collected from seven different water packaging facilities and level of phthalates determined using GC-MS instrumentation. Phthalates concentration in borehole samples varied from 0.00-0.01 (DMP), 0.06-0.20 (DEP), 0.10-0.98 (DBP), 0.21-0.36 (BEHP), 0.01-0.03 (DnOP) µg/L and (BBP) was not detectable; while sachet water varied from 0.03-0.95 (DMP), 0.16-12.45 (DEP), 0.57-3.38 (DBP), 0.00-0.03 (BBP), 0.08-0.31 (BEHP) and 0-0.03 (DnOP) µg/L. Phthalates concentration in the sachet water was higher than that of the corresponding boreholes sources and also showed significant difference (p < 0.05) between the two. Sources of these phthalate esters were the interaction between water molecules and plastic storage facilities. Although concentration of all phthalate esters analyzed were lower than the threshold limit value(TLV), over time storage of water samples in this medium can lead to substantial increase with negative effects on individuals consuming them.
Abstract: Today, wine quality is only evaluated by wine experts with their own different personal tastes, even if they may agree on some common features. So producers do not have any unbiased way to independently assess the quality of their products. A tool is here proposed to evaluate wine quality by an objective ranking based upon the variables entering wine elaboration, and analysed through principal component analysis (PCA) method. Actual climatic data are compared by measuring the relative distance between each considered wine, out of which the general ranking is performed.
Abstract: In this study, the effect of various drying methods (sun drying, shade drying and industrial drying) on final moisture content, shell splitting degree, shrinkage and color change were studied. Sun drying resulted higher degree of pistachio nuts shell splitting on pistachio nuts relative other drying methods. The ANOVA results showed that the different drying methods did not significantly effects on color change of dried pistachio nut. The results illustrated that pistachio nut dried by industrial drying had the lowest moisture content. After the end of drying process, initially, the experimental drying data were fitted with five famous drying models namely Newton, Page, Silva et al., Peleg and Henderson and Pabis. The results indicated that Peleg and Page models gave better results compared with other models to monitor the moisture ratio’s pistachio nut in industrial drying and open sun (or shade drying) methods, respectively.
Abstract: The butterflies are ecologically very important insects. The adults generally feed on nectar and are important as pollinators of flowering plants. However, these pollinators are under threat with their habitat loss. One reason for habitat loss is spread of invasive plants. However, there are even beneficial exotic plants which can directly support for Butterfly Conservation Action Plan of Sri Lanka by attracting butterflies for nectar. Stachytarpheta jamaicensis (L.) is an important nectar plant which attracts a diverse set of butterflies in higher number. It comprises a violet color inflorescence which last for about 37 hours where it attracted a peak of butterflies around 9.00am having around average of 15 butterflies. There were no butterflies in early and late hours where the number goes to very low values as 2 at 1.00pm. it was found that a diverse group of butterflies were attracted from around 15 species including 01 endemic species, 02 endemic subspecies and 02 vulnerable species. Therefore, this is a beneficial exotic plant that could be used in butterfly attraction and conservation however with adequate monitoring of the plant population.
Abstract: The study assessed the effect of crude oil applied at rates, 0, 2, 5, and 10% on the fractional chemical forms and availability of some metals in soils from Usen, Edo State, with no known crude oil contamination and soil from a crude oil spill site in Ubeji, Delta State, Nigeria. Three methods were used to determine the bioavailability of metals in the soils: maize (Zea mays) plant, EDTA and BCR sequential extraction. The sequential extract acid soluble fraction of the BCR extraction (most labile fraction of the soils, normally associated with bioavailability) were compared with total metal concentration in maize seedlings as a means to compare the chemical and biological measures of bioavailability. Total Fe was higher in comparison to other metals for the crude oil contaminated soils. The metal concentrations were below the limits of 4.7% Fe, 190mg/kg Cu and 720mg/kg Zn intervention values and 36mg/kg Cu and 140mg/kg Zn target values for soils provided by the Department of Petroleum Resources (DPR) guidelines. The concentration of the metals in maize seedlings increased with increasing rates of crude oil contamination. Comparison of the metal concentrations in maize seedlings with EDTA extractable concentrations showed that EDTA extracted more metals than maize plant.
Abstract: Nitrogen fertility is an important component for optimum potato yield and quality. Best management practices are necessary in regards to N applications to achieve these goals without applying excess N with may contribute to ground water contamination. Eight potato fields in the Southern San Joaquin Valley were sampled for nitrogen inputs and uptake, tuber and vine dry matter and residual soil nitrate-N. The fields had substantial soil nitrate-N prior to the potato crop. Nitrogen fertilizer was applied prior to planting and in irrigation water as needed based on in-season petiole sampling in accordance with published recommendations. Average total nitrogen uptake was 237 kg ha-1 on 63.5 Mg ha-1 tuber yield and nitrogen use efficiency was very good at 81 percent. Sixty-nine percent of the plant nitrogen was removed in tubers. Soil nitrate-N increased 14 percent from pre-plant to post-harvest averaged across all fields and was generally situated in the upper soil profile. Irrigation timing and amount applied did not move water into the lower profile except for a single location where nitrate also moved into the lower soil profile. Pre-plant soil analysis is important information to be used. Rotation crops having deeper rooting growth would be able to utilize nitrogen that remained in the soil profile.
Abstract: The aim of this study is to present the results of a retrospective survey on the foreign matter found in foods analyzed at the Adolfo Lutz Institute, from July 2001 to July 2015. All the analyses were conducted according to the official methods described on Association of Official Agricultural Chemists (AOAC) for the micro analytical procedures and Food and Drug Administration (FDA) for the macro analytical procedures. The results showed flours, cereals and derivatives such as baking and pasta products were the types of food where foreign matters were found more frequently followed by condiments and teas. Fragments of stored grains insects, its larvae, nets, excrement, dead mites and rodent excrement were the most foreign matter found in food. Besides, foreign matters that can cause a physical risk to the consumer’s health such as metal, stones, glass, wood were found but rarely. Miscellaneous (shell, sand, dirt and seeds) were also reported. There are a lot of extraneous materials that are considered unavoidable since are something inherent to the product itself, such as insect fragments in grains. In contrast, there are avoidable extraneous materials that are less tolerated because it is preventable with the Good Manufacturing Practice. The conclusion of this work is that although most extraneous materials found in food are considered unavoidable it is necessary to keep the Good Manufacturing Practice throughout the food processing as well as maintaining a constant surveillance of the production process in order to avoid accidents that may lead to occurrence of these extraneous materials in food.
Abstract: The aim of this study was to improve the durability and quality of Bali beef (M. Longissimus dorsi) and broiler carcass through the addition of liquid smoke as a natural preservative. This study was using Longissimus dorsi muscle from male Bali beef aged 3 years, broiler breast and thigh aged 40 days. Three types of meat were marinated in liquid smoke with concentrations of 0, 5, and 10% for 30 minutes at the level of 20% of the sample weight (w/w). The samples were storage at 2-5°C for 1 month. This study designed as a factorial experiment 3 x 3 x 4 based on a completely randomized design with 5 replications; the first factor was meat type (beef, chicken breast and chicken thigh); the 2nd factor was liquid smoke concentrations (0, 5, and 10%), and the 3rd factor was storage duration (1, 2, 3, and 4 weeks). Parameters measured were TBA value, total bacterial colonies, water holding capacity (WHC), shear force value both before and after cooking (80°C – 15min.), and cooking loss. The results showed that the type of meat produced WHC, shear force value, cooking loss and TBA differed between the three types of meat. Higher concentration of liquid smoke, the WHC, shear force value, TBA, and total bacterial colonies were decreased; at a concentration of 10% of liquid smoke, the total bacterial colonies decreased by 57.3% from untreated with liquid smoke. Longer storage, the total bacterial colonies and WHC were increased, while the shear force value and cooking loss were decreased. It can be concluded that a 10% concentration of liquid smoke was able to maintain fat oxidation and bacterial growth in Bali beef and chicken breast and thigh.
Abstract: Plants as therapeutic agents are used as drug in many parts of the world. Medicinal plants are mostly used in developing countries due to culture acceptability, belief or due to lack of easy access to primary health care services. Jatropha curcas is a plant from the Euphorbiaceae family which is widely used in Northern Nigeria as an anti-diarrheal agent. This study was conducted to determine the anti-diarrheal effect of the leaf extract on castor oil induced diarrhea in albino rats. The leaves of J. curcas were collected from Balanga Local government in Gombe State, north-eastern Nigeria; due to its bioavailability. The leaves were air-dried at room temperature and ground to powder. Phytochemical screening was done and different concentrations of the extract was prepared and administered to the different categories of experimental animals. From the results, aqueous leaf extract of Jatropha curcas at doses of 200mg/Kg and 400mg/Kg was found to reduce the mean stool score as compared to control rats, however, maximum reduction was achieved with the standard drug of Loperamide (5mg/Kg). Treatment of diarrhea with 200mg/Kg of the extract did not produce any significant decrease in stool fluid content but was found to be significant in those rats that were treated with 400mg/Kg of the extract at 2hours (0.05±0.02) and 4hours (0.01±0.01). A significant reduction of diarrhea in the experimental animals signifies it to possess some anti-diarrheal activity.
Abstract: The abiotic elicitation is one of the methods for
increasing the secondary metabolites production in plant tissue
cultures and it seems to be more effective than traditional strategies.
This study verified the use of silver nitrate as elicitor to enhance
flavonolignans and flavonoid taxifolin production in suspension
culture of Sylibum marianum (L.) Gaertn. Silver nitrate in various
concentrations (5.887.10-3 mol/L, 5.887.10-4 mol/L, 5.887.10-5
mol/L) was used as elicitor. The content of secondary metabolites in
cell suspension cultures was determined by high performance liquid
chromatography. The samples were taken after 6, 12, 24, 48, 72 and
168 hours of treatment. The highest content of taxifolin production
(2.2 mg.g-1) in cell suspension culture of Silybum marianum (L.)
Gaertn. was detected after silver nitrate (5.887.10-4 mol/L) treatment
and 72 h application. Flavonolignans such as silybinA, silybin B,
silydianin, silychristin, isosilybin A, isosilybin B were not produced
by cell suspension culture of S. marianum after elicitor treatment.
Our results show that the secondarymetabolites could be released
from S. marianum cells into the nutrient medium by changed
permeability of cell wall.
Abstract: Pure form of L-Dopa is used to enhance muscular development, fat breakdown and suppress Parkinson disease in humans. However, the L-Dopa in mucuna seed, when present with other antinutritional factors, causes nutritional disorders in monogastric animals. Information on the utilisation of pure L-Dopa in monogastric animals is scanty. Therefore, effect of L-Dopa on growth performance and carcass characteristics in broiler chickens was investigated. Two hundred and forty one-day-old chicks were allotted to six treatments, which consisted of a positive control (PC) with standard energy (3100Kcal/Kg) and negative control (NC) with high energy (3500Kcal/Kg). The rest 4 diets were NC+0.1, NC+0.2, NC+0.3 and NC+0.4% L-Dopa, respectively. All treatments had 4 replicates in a completely randomized design. Body weight gain, final weight, feed intake, dressed weight and carcass characteristics were determined. Body weight gain and final weight of birds fed PC were 1791.0 and 1830.0g, NC+0.1% L-Dopa were 1827.7 and 1866.7g and NC+0.2% L-Dopa were 1871.9 and 1910.9g respectively, and the feed intake of PC (3231.5g), were better than other treatments. The dressed weight at 1375.0g and 1357.1g of birds fed NC+0.1% and NC+0.2% L-Dopa, respectively, were similar but better than other treatments. Also, the thigh (202.5g and 194.9g) and the breast meat (413.8g and 410.8g) of birds fed NC+0.1% and NC+0.2% L-Dopa, respectively, were similar but better than birds fed other treatments. The drum stick of birds fed NC+0.1% L-Dopa (220.5g) was observed to be better than birds on other diets. Meat to bone ratio and relative organ weights were not affected across treatments. L-Dopa extract, at levels tested, had no detrimental effect on broilers, rather better bird performance and carcass characteristics were observed especially at 0.1% and 0.2% L-Dopa inclusion rates. Therefore, 0.2% inclusion is recommended in diets of broiler chickens for improved performance and carcass characteristics.
Abstract: Biochemical and hormonal changes that occur in both follicular fluid and blood are involved in the control of ovarian physiology. The present study was conducted on follicular fluid and serum samples obtained from 708 buffaloes. Samples were examined for estradiol, progesterone, and cholesterol concentrations in relation to seasonal changes, ovarian follicular size, and stage of estrous cycle. The obtained results revealed that follicular fluid and serum levels of estradiol, progesterone, and cholesterol were significantly lower during summer and autumn when compared to winter and spring seasons. With the increase in follicular size, the follicular fluid levels of progesterone and cholesterol were significantly decreased, while estradiol levels were significantly increased. Estradiol and progesterone levels were significantly higher in follicular fluid than blood, while cholesterol was significantly lower in follicular fluid than serum. In conclusion, the current study threw a light on the hormonal changes in the follicular fluid and blood under the effect of heat stress which could be related to the low fertility of buffalo in the summer.
Abstract: The safflower seed contains about 25-40% solvent extract and 20-33% fiber. It is well known that dietary phospholipids lower serum cholesterol levels effectively. The nutrient composition of safflower seed changes depending on region, soil and genotypes. This research was made by using of six natural selected (A22, A29, A30, C12, E1, F4, G8, G12, J27) and three commercial (Remzibey, Dincer, Black Sun1) varieties of safflower genotypes. The research was conducted on field conditions for two years (2009 and 2010) in randomized complete block design with three replications in Konya-Turkey ecological conditions. Oil contents, mineral contents and their correlations were determined in the research. According to the results, oil content was ranged from 22.38% to 34.26%, while the minerals were in between the following values: 1469, 04-2068.07 mg kg-1 for Ca, 7.24-11.71 mg kg-1 for B, 13.29-17.41 mg kg-1 for Cu, 51.00-79.35 mg kg-1 for Fe, 3988-6638.34 mg kg-1 for K, 1418.61-2306.06 mg kg-1 for Mg, 11.37-17.76 mg kg-1 for Mn, 4172.33-7059.58 mg kg-1 for P and 32.60-59.00 mg kg-1 for Zn. Correlation analysis that was made separately for the commercial varieties and wild lines showed that high level of oil content was negatively affected by all the investigated minerals except for K and Zn in the commercial varieties.
Abstract: Goat production is one of the activities included in
integrated farming in the Philippines. Goats are raised for its meat
and regardless of breed the animal is slaughtered for this purpose. In
order to document the carcass yield of different goats slaughtered,
five (5) different breeds of goats to include Purebred Boer and
Anglo-nubian, Crossbred Boer and Anglo-nubian and Philippine
Native goat were used in the study. Data on slaughter parameters,
carcass characteristics, and sensory evaluation were gathered and
analyzed using Complete Random Design (CRD) at 5% level of
significance and the results of carcass conformation were assessed
descriptively. Results showed that slaughter data such as
slaughter/live weight, hot and chilled carcass weights, dressing
percentage and percentage drip loss were significantly different
(P>0.05) among breeds. On carcass and meat characteristics, pure
breed and upgraded Boer were found to be moderately muscular
while Native goat was rated as thin muscular. The color of the
carcass also revealed that Purebred and crossbred Boer were
described dark red, while Native goat was noted to be slightly pale.
On sensory evaluation, the results indicated that there was no
significant difference (P>0.05) among breeds evaluated. It is
therefore concluded that purebred goat has heavier carcass, while
both purebred Boer and upgrade are rated slightly muscular. It is
further confirms that regardless of breed, goat will have the same
sensory characteristics. Thus, it is recommended to slaughter heavier
goats to obtain more carcasses with better conformation and quality.
Abstract: Approximately 10,000 different types of dyes and
pigments are being used in various industrial applications yearly,
which include the textile and printing industries. However, these dyes
are difficult to degrade naturally once they enter the aquatic system.
Their high persistency in natural environment poses a potential health
hazard to all form of life. Hence, there is a need for alternative dye
removal strategy in the environment via bioremediation. In this study,
fungi laccase is investigated via commercial agar dyes plates and
submerged fermentation to explore the application of fungi laccase in
textile dye wastewater treatment. Two locally isolated basidiomycetes
were screened for laccase activity using media added with commercial
dyes such as 2, 2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid
(ABTS), guaiacol and Remazol Brillant Blue R (RBBR). Isolate TBB3
(1.70±0.06) and EL2 (1.78±0.08) gave the highest results for ABTS
plates with the appearance of greenish halo on around the isolates.
Submerged fermentation performed on Isolate TBB3 with the
productivity 3.9067 U/ml/day, whereas the laccase activity for Isolate
EL2 was much lower (0.2097 U/ml/day). As isolate TBB3 showed
higher laccase production, it was subjected to molecular
characterization by DNA isolation, PCR amplification and sequencing
of ITS region of nuclear ribosomal DNA. After being compared with
other sequences in National Center for Biotechnology Information
(NCBI database), isolate TBB3 is probably from species Trametes
hirsutei. Further research work can be performed on this isolate by
upscale the production of laccase in order to meet the demands of the
requirement for higher enzyme titer for the bioremediation of textile
dyes.
Abstract: The present study investigated the protective effect of
thymoquinone (TQ), against cadmium-induced kidney injury in rats.
Cadmium chloride (1.2 mg Cd/kg/day, s.c.), was given for nine
weeks. TQ treatment (40 mg/kg/day, p.o.) started on the same day of
cadmium administration and continued for nine weeks. TQ
significantly decreased serum creatinine, renal malondialdehyde and
nitric oxide, and significantly increased renal reduced glutathione in
rats received cadmium. Histopathological examination showed that
TQ markedly minimized renal tissue damage induced by cadmium.
Immunohistochemical analysis revealed that TQ markedly decreased
the cadmium-induced expression of inducible nitric oxide synthase,
tumor necrosis factor-α, cyclooxygenase-2, and caspase-3 in renal
tissue. It was concluded that TQ significantly protected against
cadmium nephrotoxicity in rats, through its antioxidant, antiinflammatory,
and antiapoptotic actions.
Abstract: Not all proteins have the same nutritional value, since
protein quality strongly depends on its amino acid composition and
digestibility. The meat of game animals could be a high protein
source because of its well-balanced essential amino acids
composition. Investigations about biochemical composition of game
meat such as wild boar (Sus scrofa scrofa), roe deer (Capreolus
capreolus) and beaver (Castor fiber) are not very much. Therefore,
the aim of the investigation was evaluate protein composition of
game meat hunted in Latvia. The biochemical analysis, evaluation of
connective tissue and essential amino acids in meat samples were
done, the amino acids score were calculate. Results of analysis
showed that protein content 20.88-22.05% of all types of meat
samples is not different statistically. The content of connective tissue
from 1.3% in roe deer till 1.5% in beaver meat allowed classified
game animal as high quality meat. The sum of essential amino acids
in game meat samples were determined 7.05–8.26g100g-1. Roe deer
meat has highest protein content and lowest content of connective
tissues among game meat hunted in Latvia. Concluded that amino
acid score for limiting amino acids phenylalanine and tyrosine is high
and shows high biological value of game meat.
Abstract: Problem of food preservation is extremely important
for mankind. Viscous damage ("illness") of bread results from
development of Bacillus spp. bacteria. High temperature resistant
spores of this microorganism are steady against 120°C) and remain in
bread during pastries, potentially causing spoilage of the final
product. Scientists are interested in further characterization of bread
spoiling Bacillus spp. species. Our aim was to find weather yeast
Saccharomyces cerevisiae strains that are able to produce natural
antimicrobial killer factor can preserve bread illness. By diffusion
method, we showed yeast antagonistic activity against spore-forming
bacteria. Experimental technological parameters were the same as for
bakers' yeasts production on the industrial scale. Risograph test
during dough fermentation demonstrated gas production. The major
finding of the study was a clear indication of the presence of killer
yeast strain antagonistic activity against rope in bread causing
bacteria. After demonstrating antagonistic effect of S. cerevisiae on
bacteria using solid nutrient medium, we tested baked bread under
provocative conditions. We also measured formation of carbon
dioxide in the dough, dough-making duration and quality of the final
products, when using different strains of S. cerevisiae. It is
determined that the use of yeast S. cerevisiae RCAM 01730 killer
strain inhibits appearance of rope in bread. Thus, natural yeast
antimicrobial killer toxin, produced by some S. cerevisiae strains is
an anti-rope in bread protector.
