Abstract: Carbon nanotubes (CNTs) are attractive because of
their excellent chemical durability mechanical strength and electrical
properties. Therefore there is interest in CNTs for not only electrical
and mechanical application, but also biological and medical
application.
In this study, the dispersion power of surfactant-treated multiwalled
carbon nanotubes (MWCNTs) and their effect on the antibacterial
activity were examined. Surfactant was used sodium
dodecyl-benzenesulfonate (SDBS). UV-vis absorbance and
transmission electron microscopy(TEM) were used to characterize the
dispersion of MWCNTs in the aqueous phase, showing that the
surfactant molecules had been adsorbed onto the MWCNTs surface.
The surfactant-treated MWCNTs exhibited antimicrobial activities
to streptococcus mutans. The optical density growth curves and viable
cell number determined by the plating method suggested that the
antimicrobial activity of surfactant-treated MWCNTs was both
concentration and treatment time-dependent.
Abstract: This study was set to determine the antimicrobial
activities of brine salting, chlorinated solution, and oil frying
treatments on enteric bacteria and fungi in Rastrineobola argentea
fish from fish landing beaches within L. Victoria basin of western
Kenya. Statistical differences in effectiveness of the different
treatment methods was determined by single factor ANOVA, and
paired two-tail t-Test was performed to compare the differences in
moisture contents before and after storage. Oil fried fish recorded the
lowest microbial loads, sodium chloride at 10% concentration was
the second most effective and chlorinated solution even at 150ppm
was the least effective against the bacteria and fungi in fish. Moisture
contents of the control and treated fish were significantly lower after
storage. These results show that oil frying of fish should be adopted
for processing and preserving Rastrineobola argentea which is the
most abundant and affordable fish species from Lake Victoria.
Abstract: The biological activity of A. pullulans isolates against
species of the genus Fusarium, bacteria of the genus Azotobacter and
pseudomonads colonizing wheat kernels was evaluated. A field
experiment was carried out in 2009-2011, in north-eastern Poland.
Winter wheat (cv. Bogatka) plants were sprayed with a cell
suspension of A. pullulans at a density of 106 - 108 per cm3 water at
the stem elongation stage and the heading stage. Untreated plants
served as control. The abundance of epiphytic yeasts, bacteria of the
genus Azotobacter, pseudomonads and Fusarium pathogens on wheat
grain was estimated at harvest and after six months’ storage. The
average size of yeast communities was significantly greater on wheat
kernels treated with a cell suspension of A. pullulans, compared with
control samples. In 2010-2011, biological control reduced the
abundance of some species of the genus Fusarium.
Abstract: The main objective of this research was to investigate
the biosorption capacity for biofilms of sulphate reducing bacteria
(SRB) to remove heavy metals, such as Zn, Pb and Cd from
rainwater using laboratory-scale reactors containing mixed support
media. Evidence showed that biosorption had contributed to removal
of heavy metals including Zn, Pb and Cd in presence of SRB and
SRB were also found in the aqueous samples from reactors.
However, the SRB and specific families (Desulfobacteriaceae and
Desulfovibrionaceae) were found mainly in the biomass samples
taken from all reactors at the end of the experiment. EDX-analysis
of reactor solids at end of experiment showed that heavy metals Zn,
Pb and Cd had also accumulated in these precipitates.
Abstract: A series of 1-(1H-benzimidazol-2-yl)-3-(substituted phenyl)-2-propen-1-one were allowed to react with hydrazine hydrate and phenyl hydrazine in submitted reactions to get pyrazoline and phenyl pyrazoline derivatives. All the compounds entered for screening at the Tuberculosis Antimicrobial Acquisition and Coordinating Facility (TAACF) for their in vitro antibacterial activity against Mycobacterium tuberculosis H37Rv strain (ATCC 27294) using Microplate Alamar Blue Assay (MABA) susceptibility test. The results expressed as MIC (minimum inhibitory concentration) in μg/mL. Among the fifteen compounds, eight compounds were found to have MIC values less than 10 μg/mL. These were subjected for cytotoxicity assay in VERO cells to determine CC50 (cytotoxic concentration 50%) values and finally SI (Selectivity Index) were calculated. Compound (XV) 2-[5-(4- fluorophenyl)-1-phenyl-4,5-dihydro-1H-3-pyrazolyl]-1Hbenzimidazole was considered the best candidate of the series that could be a good starting point to develop new lead compounds in the fight against tuberculosis.
Abstract: The increasing usage of antibiotics in the animal
farming industry is an emerging worldwide problem contributing to
the development of antibiotic resistance. The purpose of this work was
to investigate the prevalence and antibiotic resistance profile of
bacterial isolates collected from aquatic environments and meats in a
peri-urban community in Daejeon, Korea. In an antibacterial
susceptibility test, the bacterial isolates showed a high incidence of
resistance (~ 26.04 %) to cefazolin, tetracycline, gentamycin,
norfloxacin, erythromycin and vancomycin. The results from a test for
multiple antibiotic resistance indicated that the isolates were
displaying an approximately 5-fold increase in the incidence of
multiple antibiotic resistance to combinations of two different
antibiotics compared to combinations of three or more antibiotics.
Most of the isolates showed multi-antibiotic resistance, and the
resistance patterns were similar among the sampling groups.
Sequencing data analysis of 16S rRNA showed that most of the
resistant isolates appeared to be dominated by the classes
Betaproteobacteria and Gammaproteobacteria in the phylum
Proteobacteria.
Abstract: According to FDA (Food and Drug Administration of the United States), vinegar is definedas a sour liquid containing at least 4 grams acetic acid in 100 cubic centimeter (4% solution of acetic acid) of solution that is produced from sugary materials by alcoholic fermentation. In the base of microbial starters, vinegars could be contained of more than 50 types of volatile and aromatic substances that responsible for their sweet taste and smelling. Recently the vinegar industry has a great proportion in agriculture, food and microbial biotechnology. The acetic acid bacteria are from the family Acetobacteraceae. Regarding to the latest version of Bergy-s Mannual of Systematic Bacteriology that has categorized bacteria in the base of their 16s RNA differences, the most important acetic acid genera are included Acetobacter (genus I), Gluconacetobacter (genus VIII) and Gluconobacter (genus IX). The genus Acetobacter that is primarily used in vinegar manufacturing plants is a gram negative, obligate aerobe coccus or rod shaped bacterium with the size 0.6 - 0.8 X 1.0 - 4.0 μm, nonmotile or motile with peritrichous flagella and catalase positive – oxidase negative biochemically. Some strains are overoxidizer that could convert acetic acid to carbon dioxide and water. In this research one Acetobacter native strain with high acetic acid productivity was isolated from Iranian white – red cherry. We used two specific culture media include Carr medium [yeast extract, 3%; ethanol, 2% (v/v); bromocresol green, 0.002%; agar, 2% and distilled water, 1000 ml], Frateur medium [yeast extract, 10 g/l; CaCO3, 20 g/l; ethanol, 20 g/l; agar, 20 g/l and distilled water, 1000 ml] and an industrial culture medium. In addition to high acetic acid production and high growth rate, this strain had a good tolerance against ethanol concentration that was examined using modified Carr media with 5%, 7% and 9% ethanol concentrations. While the industrial strains of acetic acid bacteria grow in the thermal range of 28 – 30 °C, this strain was adapted for growth in 34 – 36 °C after 96 hours incubation period. These dramatic characteristics suggest a potential biotechnological strain in production of cherry vinegar with a sweet smell and different nutritional properties in comparison to recent vinegar types. The lack of growth after 24, 48 and 72 hours incubation at 34 – 36 °C and the growth after 96 hours indicates a good and fast thermal flexibility of this strain as a significant characteristic of biotechnological and industrial strains.
Abstract: Nowadays, several techniques such as; Fuzzy
Inference System (FIS) and Neural Network (NN) are employed for
developing of the predictive models to estimate parameters of water
quality. The main objective of this study is to compare between the
predictive ability of the Adaptive Neuro-Fuzzy Inference System
(ANFIS) model and Artificial Neural Network (ANN) model to
estimate the Biochemical Oxygen Demand (BOD) on data from 11
sampling sites of Saen Saep canal in Bangkok, Thailand. The data is
obtained from the Department of Drainage and Sewerage, Bangkok
Metropolitan Administration, during 2004-2011. The five parameters
of water quality namely Dissolved Oxygen (DO), Chemical Oxygen
Demand (COD), Ammonia Nitrogen (NH3N), Nitrate Nitrogen
(NO3N), and Total Coliform bacteria (T-coliform) are used as the
input of the models. These water quality indices affect the
biochemical oxygen demand. The experimental results indicate that
the ANN model provides a higher correlation coefficient (R=0.73)
and a lower root mean square error (RMSE=4.53) than the
corresponding ANFIS model.
Abstract: Prediction of bacterial virulent protein sequences can
give assistance to identification and characterization of novel
virulence-associated factors and discover drug/vaccine targets against
proteins indispensable to pathogenicity. Gene Ontology (GO)
annotation which describes functions of genes and gene products as a
controlled vocabulary of terms has been shown effectively for a
variety of tasks such as gene expression study, GO annotation
prediction, protein subcellular localization, etc. In this study, we
propose a sequence-based method Virulent-GO by mining informative
GO terms as features for predicting bacterial virulent proteins.
Each protein in the datasets used by the existing method
VirulentPred is annotated by using BLAST to obtain its homologies
with known accession numbers for retrieving GO terms. After
investigating various popular classifiers using the same five-fold
cross-validation scheme, Virulent-GO using the single kind of GO
term features with an accuracy of 82.5% is slightly better than
VirulentPred with 81.8% using five kinds of sequence-based features.
For the evaluation of independent test, Virulent-GO also yields better
results (82.0%) than VirulentPred (80.7%). When evaluating single
kind of feature with SVM, the GO term feature performs much well,
compared with each of the five kinds of features.
Abstract: The consumption of lactose in acid cheese whey
anaerobic fermentation process under fed-batch conditions was
studied. During fermentation for 100 hours the biogas production
(CO2 and CH4) was analyzed online. Among the standard analyses
FT-IR spectroscopy was used to follow the consumption of lactose by
bacteria. The absorption bands at 990, 894 and 787 cm-1 in the 2nd
derivative spectra were shown to be characteristic for lactose and
were used to follow the lactose conversion. It was shown that acid
cheese whey lactose was converted by bacteria in first 7 hours. In the
spectra of 17, 18 and 95 hour fermentation samples lactose was not
identified and these results correlated with the HPLC data.
Abstract: The efficient operation of any biological treatment
process requires pre-treatment of incompatible pollutants such as
acids, bases, oil, toxic substances, etc. which hamper the treatment
of other major components which are otherwise degradable. The
pre-treatment of alkaline waste-waters, generated from various
industries like textile, paper & pulp, potato-processing industries,
etc., having a pH of 10 or higher, is essential. The pre-treatment,
i.e., neutralization of such alkaline waste-waters can be achieved by
chemical as well as biological means. However, the biological pretreatment
offers better package over the chemical means by being
safe and economical. The biological pre-treatment can be
accomplished by using a blend of microorganisms able to withstand
such harsh alkaline conditions. In the present study, for the proper
pre-treatment of alkaline waste-waters, a package of alkalophilic
bacteria is formulated to neutralise the alkaline pH of the industrial
waste-waters. The developed microbial package is cost-effective as
well as environmental friendly.
Abstract: Erwinia carotovora var. carotovora is the main cause of soft rot in potatoes. Hyphaene thebaica was studied for biocontrol of E. carotovora which inhibited growth of E. carotovora on solid medium, a comparative study of classical and ultrasound-assisted extractions of Hyphaene thebaica fruit. The use of ultrasound decreased significant the total time of treatment and increase the total amount of crude extract. The crude extract was subjected to determine the in vitro, by a bioassay technique revealed that the treatment of paper disks with ultrasound extraction of Hyphaene thebaica reduced the growth of pathogen and produced inhibition zones up to 38mm in diameter. The antioxidant activity of ultrasound-ethanolic extract of Doum fruits (Hyphaene thebaica) was determined. Data obtained showed that the extract contains the secondary metabolites such as Tannins, Saponin, Flavonoids, Phenols, Steroids, Terpenoids, Glycosides and Alkaloids.
Abstract: Food borne illnesses have been reported to be a global
health challenge. Annual incidences of food–related diseases involve
76 million cases, of which only 14 million can be traced to known
pathogens. Poor hygienic practices have contributed greatly to this. It
has been reported that in the year 2000 about 2.1 million people died
from diarrheal diseases, hence, there is a need to ensure food safety at
all level. This study focused on the sterility examination and
inhibitory effect of honey samples on selected gram negative and
gram positive food borne pathogen from South West Nigeria. The
laboratory examinations revealed the presence of some bacterial and
fungal contaminations of honey samples and that inhibitory activity
of the honey sample was more pronounced on the gram negative
bacteria than the gram positive bacterial isolates. Antibiotic
sensitivity test conducted on the different bacterial isolates also
showed that honey was able to inhibit the proliferation of the tested
bacteria than the employed antibiotics.
Abstract: Antimicrobial resistant is becoming a major factor in
virtually all hospital acquired infection may soon untreatable is a
serious public health problem. These concerns have led to major
research effort to discover alternative strategies for the treatment of
bacterial infection. Nanobiotehnology is an upcoming and fast
developing field with potential application for human welfare. An
important area of nanotechnology for development of reliable and
environmental friendly process for synthesis of nanoscale particles
through biological systems In the present studies are reported on the
use of fungal strain Aspergillus species for the extracellular synthesis
of bionanoparticles from 1 mM silver nitrate (AgNO3) solution. The
report would be focused on the synthesis of metallic bionanoparticles
of silver using a reduction of aqueous Ag+ ion with the
culture supernatants of Microorganisms. The bio-reduction of the
Ag+ ions in the solution would be monitored in the aqueous
component and the spectrum of the solution would measure through
UV-visible spectrophotometer The bionanoscale particles were
further characterized by Atomic Force Microscopy (AFM), Fourier
Transform Infrared Spectroscopy (FTIR) and Thin layer
chromatography. The synthesized bionanoscale particle showed a
maximum absorption at 385 nm in the visible region. Atomic Force
Microscopy investigation of silver bionanoparticles identified that
they ranged in the size of 250 nm - 680 nm; the work analyzed the
antimicrobial efficacy of the silver bionanoparticles against various
multi drug resistant clinical isolates. The present Study would be
emphasizing on the applicability to synthesize the metallic
nanostructures and to understand the biochemical and molecular
mechanism of nanoparticles formation by the cell filtrate in order to
achieve better control over size and polydispersity of the
nanoparticles. This would help to develop nanomedicine against
various multi drug resistant human pathogens.
Abstract: Spent Sulfidic Caustic was biologically treated and
regenerated for reusing by Thiobacillus denitrificans bacteria, sulfide
content oxidized and RSNa reduced dramatically.PH in this test was
11.8 and no neutralization has been done on spent caustic, so spent
caustic as the most difficult of industrial wastes to dispose could be
regenerate and reuse instead of disposing to sea or deep wells
Abstract: Today, biogenic magnetite nanoparticles among
magnetic nanoparticles have unique attracted attention because of
their magnetic characteristics and potential applications in various
fields such as therapeutic and diagnostic. A well known example of
these biogenic nanoparticles is magnetosomes of magnetotactic
bacteria. In this research, we used two different types of technique for
the isolation and purification of magnetosome nanoparticles from the
isolated magnetotactic bacterial cells, heat-alkaline treatment and
sonication. Also we evaluated pyrogen content and sterility of
synthesized the isolated individual magnetosome by the Limulus
Amoebocyte Lysate test and direct impedimetric method
respectively.
Abstract: The principal objective of a water treatment plant is to
produce water that satisfies a set of drinking water quality standards
at a reasonable price to the consumers. The gravel-bed flocculator
provide a simple and inexpensive design for flocculation in small
water treatment plants (less than 5000 m3/day capacity). The packed
bed of gravel provides ideal conditions for the formation of compact
settleable flocs because of continuous recontact provided by the
sinuous flow of water through the interstices formed by the gravel.
The field data which were obtained from the operation of the
water supply treatment unit cover the physical, chemical and
biological water qualities of the raw and settled water as obtained by
the operation of the treatment unit. The experiments were carried out
with the aim of assessing the efficiency of the gravel filter in
removing the turbidity, pathogenic bacteria, from the raw water. The
water treatment plant, which was constructed for the treatment of
river water, was in principle a rapid sand filter.
The results show that the average value of the turbidity level of
the settled water was 4.83 NTU with a standard deviation of turbidity
2.893 NTU. This indicated that the removal efficiency of the
sedimentation tank (gravel filter) was about 67.8 %. for pH values
fluctuated between 7.75 and 8.15, indicating the alkaline nature of
the raw water of the river Shatt Al-Hilla, as expected. Raw water pH
is depressed slightly following alum coagulation. The pH of the
settled water ranged from 7.75 to a maximum of 8.05.
The bacteriological tests which were carried out on the water
samples were: total coliform test, E-coli test, and the plate count test.
In each test the procedure used was as outlined in the Standard
Methods for the Examination of Water and Wastewater (APHA,
AWWA, and WPCF, 1985). The gravel filter exhibit a low
performance in removing bacterial load. The percentage bacterial
removal, which is maximum for total plate count (19%) and
minimum for total coliform (16.82%).
Abstract: 17α-ethynylestradiol (EE2) is a synthetic estrogen
used as a key ingredient in an oral contraceptives pill. EE2 is an
endocrine disrupting compound, high in estrogenic potency.
Although EE2 exhibits low degree of biodegradability with common
microorganisms in wastewater treatment plants (WWTPs), this
compound can be biotransformed by ammonia-oxidizing bacteria
(AOB) via a co-metabolism mechanism in WWTPs. This study
aimed to investigate the effect of real wastewater on
biotransformation of EE2 by AOB. A preliminary experiment on the
effect of nitrite and pH levels on abiotic transformation of EE2
suggested that the abiotic transformation occurred at only pH
Abstract: Lectins have a good scope in current clinical
microbiology research. In the present study evaluated the
antimicrobial activities of a D-galactose binding lectin (PnL) was
purified from the annelid, Perinereis nuntia (polychaeta) by affinity
chromatography. The molecular mass of the lectin was determined to
be 32 kDa as a single polypeptide by SDS-PAGE under both reducing
and non-reducing conditions. The hemagglutinating activity of the
PnL showed against trypsinized and glutaraldehyde-fixed human
erythrocytes was specifically inhibited by D-Gal, GalNAc,
Galβ1-4Glc and Galα1-6Glc. PnL was evaluated for in vitro
antibacterial screening studies against 11 gram-positive and
gram-negative microorganisms. From the screening results, it was
revealed that PnL exhibited significant antibacterial activity against
gram-positive bacteria. Bacillus megaterium showed the highest
growth inhibition by the lectin (250 μg/disc). However, PnL did not
inhibit the growth of gram-negative bacteria such as Vibrio cholerae
and Pseudomonas sp. PnL was also examined for in vitro antifungal
activity against six fungal phytopathogens. PnL (100 μg/mL) inhibited
the mycelial growth of Alternaria alternata (24.4%). These results
indicate that future findings of lectin applications obtained from
annelids may be of importance to life sciences.
Abstract: Effect of viscosity of media on kinetic parameters of the coupled enzyme system NADH:FMN-oxidoreductase–luciferase was investigated with addition of organic solvents (glycerol and sucrose), because bioluminescent enzyme systems based on bacterial luciferases offer a unique and general tool for analysis of the many analytes and enzymes in the environment, research and clinical laboratories and other fields. The possibility of stabilization and increase of activity of the coupled enzyme system NADH:FMN-oxidoreductase–luciferase activity in vicious aqueous-organic mixtures have been shown.
