Abstract: The objective of the study was to select the survival of
probiotic strains when exposed to acidic and bile salts condition. Four
probiotic strains Lactobacillus casei subsp. rhamnosus TISTR 047,
Lactobacillus casei TISTR 1500, Lactobacillus acidophilus TISTR
1338 and Lactobacillus plantarum TISTR 1465 were cultured in
MRS broth and incubated at 35ºC for 15 hours before being inoculated
into acidic condition 5 M HCl, pH 2 for 2 hours and bile salt 0.3%,
pH 5.8 for 8 hour. The survived probiotics were counted in MRS agar.
Among four stains, Lactobacillus casei subsp. rhamnosus TISTR 047
was the highest tolerance specie. Lactobacillus casei subsp.
rhamnosus TISTR 047 reduced 6.74±0.07 log CFU/ml after growing
in acid and 5.52±0.05 log CFU/ml after growing in bile salt. Then,
double emulsion of microorganisms was chosen to encapsulate before
spray drying. Spray drying was done with the inlet temperature 170ºC
and outlet temperature 80ºC. The results showed that the survival of
encapsulated Lactobacillus casei subsp. rhamnosus TISTR 047 after
spray drying decreased from 9.63 ± 0.32 to 8.31 ± 0.11 log CFU/ml
comparing with non-encapsulated, 9.63 ± 0.32 to 4.06 ± 0.08 log
CFU/ml. Therefore, Lactobacillus casei subsp. rhamnosus TISTR 047
would be able to survive in gastrointestinal and spray drying condition.
Abstract: Red River Gum (Eucalyptus camaldulensis) is a tree
of the genus Eucalyptus widely distributed in Algeria and in the
world. The value of its aromatic secondary metabolites offers new
perspectives in the pharmaceutical industry. This strategy can
contribute to the sustainable development of our country. Preliminary
tests performed on the essential oil of Eucalyptus camendulensis
showed that this oil has antibacterial activity vis-à-vis the bacterial
strains (Enterococcus feacalis, Enterobacter cloaceai, Proteus
microsilis, Escherichia coli, Klebsiella pneumonia, and Pseudomonas
aeruginosa) and antifungic (Fusarium sporotrichioide and Fusarium
graminearum). The culture medium used was nutrient broth Muller
Hinton. The interaction between the bacteria and the essential oil is
expressed by a zone of inhibition with diameters of MIC indirectly
expression of. And we used the PDA medium to determine the fungal
activity. The extraction of the aromatic fraction (essentially oilhydrolat)
of the fresh aerian part of the Eucalyptus camendulensis
was performed by hydrodistillation. The average essential oil yield is
0.99%. The antimicrobial and fungal study of the essential oil and
hydrosol showed a high inhibitory effect on the growth of pathogens.
Abstract: The lactic acid bacteria (LAB) were isolated from
10 samples of fermented foods (Sa-tor-dong and Bodo) in South
locality of Thailand. The 23 isolates of lactic acid bacteria were
selected, which were exhibited a clear zone and growth on MRS
agar supplemented with CaCO3. All of lactic acid bacteria were
tested on morphological and biochemical. The result showed that
all isolates were Gram’s positive, non-spore forming but only
10 isolates displayed catalase negative. The 10 isolates including
BD1 .1, BD 1.2, BD 2.1, BD2.2, BD 2.3, BD 3.1, BD 4.1, BD 5.2,
ST 4.1 and ST 5.2 were selected for inhibition activity
determination. Only 2 strains (ST 4.1 and BD 2.3) showed
inhibition zone on agar, when using Escherichia coli sp. as target
strain. The ST 4.1 showed highest inhibition zone on agar, which
was selected for probiotic property testing. The ST4.1 isolate
could grow in MRS broth containing a high concentration of
sodium chloride 6%, bile salts 7%, pH 4-10 and vary temperature
at 15-45°C.
Abstract: A total of 150 meat type chickens comprising 50 each
of Arbor Acre, Marshall and Ross were used for this study which
lasted for 10 weeks at the Federal University of Agriculture,
Abeokuta, Nigeria. Growth performance data were collected from the
third week through week 10 and data obtained were analysed using
the Generalized Linear Model Procedure. Heritability estimates (h2)
for body dimensions carried out on the chicken strains ranged from
low to high. Marshall broiler chicken strain had the highest h2 for
body weight 0.46±0.04, followed by Arbor Acre and Ross with h2
being 0.38±0.12 and 0.26±0.06, respectively. The repeatability
estimates for body weight in the three broiler strains were high, and it
ranged from 0.70 at week 4 to 0.88 at week 10. Relationships
between the body weight and linear body measurements in the broiler
chicken strains were positive and highly significant (p > 0.05).
Abstract: Natural antimicrobials are used to preserve foods that
can be found in plants, animals, and microorganisms. Antimicrobial
substances are natural or artificial agents that produced by
microorganisms or obtained semi/total chemical synthesis are used at
low concentrations to inhibit the growth of other microorganisms.
Food borne pathogens and spoilage microorganisms are inactivated
by the use of antagonistic microorganisms and their metabolites.
Yeasts can produce toxic proteins or glycoproteins (toxins) that cause
inhibition of sensitive bacteria and yeast species. Antimicrobial
substance producing phenotypes belonging different yeast genus
were isolated from different sources. Toxins secreted by many yeast
strains inhibiting the growth of other yeast strains. These strains show
antimicrobial activity, inhibiting the growth of mold and bacteria.
The effect of antimicrobial agents produced by yeasts can be
extremely fast, and therefore may be used in various treatment
procedures. Rapid inhibition of microorganisms is possibly caused by
microbial cell membrane lipopolysaccharide binding and in
activation (neutralization) effect. Antimicrobial agents inhibit the
target cells via different mechanisms of action.
Abstract: In this study, six bacterial isolates of a slightly
thermophilic organism from the Sg. Klah hot spring, Malaysia were
successfully isolated and designated as M7T55D1, M7T55D2,
M7T55D3, M7T53D1, M7T53D2 and M7T53D3 respectively. The
bacterial isolates were screened for their cellulose hydrolytic ability
on Carboxymethlycellulose agar medium. The isolated bacterial
strains were identified morphologically, biochemically and
molecularly with the aid of 16S rDNA sequencing. All of the bacteria
showed their optimum growth at a slightly alkaline pH of 7.5 with a
temperature of 55°C. All strains were Gram-negative, non-spore
forming type, strictly aerobic, catalase-positive and oxidase-positive
with the ability to produce thermostable cellulase. Based on BLASTn
results, bacterial isolates of M7T55D2 and M7T53D1 gave the
highest homology (97%) with similarity to Tepidimonas ignava while
isolates M7T55D1, M7T55D3, M7T53D2 and M7T53D3 showed
their closest homology (97%-98%) with Tepidimonas thermarum.
These cellulolytic thermophiles might have a commercial potential to
produce valuable thermostable cellulase.
Abstract: Abundant and cheap agricultural waste of oil palm trunk (OPT) juice was used to produce bioethanol. Two strains of Saccharomyces cerevisiae and a strain of Pichia stipitis were used to produce bioethanol from the OPT juice. Fermentation was conducted at previously optimized condition at 30oC and without shaking. The kinetic parameters were estimated and calculated. Monod equation and Hinshelwood model is used to relate the specific growth to the concentration of the limiting substrate and also to simulate bioethanol production rate. Among the three strains, single S. cerevisiae Kyokai no. 7 produce the highest ethanol yield of 0.477 g/l.h within the shortest time (12 h). This yeast also produces more than 20 g/l ethanol concentration within 10 h of fermentation.
Abstract: Among agricultural residues, sugarcane bagasse is one of the most convincing raw materials for the production of bioethanol due to its availability, and low cost through enzymatic hydrolysis and yeast fermentation. A pretreatment step is needed to enhance the enzymatic step. In this study, sugarcane bagasse (SCB), one of the most abundant agricultural residues in Thailand, was pretreated biologically with various microorganisms of white-rot fungus—Phanerochaete sordid (SK 7), Cellulomonas sp. (TISTR 784), and strain A 002 (Bacillus subtilis isolated from Thai higher termites). All samples with various microbial pretreatments were further hydrolyzed enzymatically by a commercial enzyme obtained from Aspergillus niger. The results showed that the pretreatment with the white-rot fungus gave the highest glucose concentration around two-fold higher when compared with the others.
Abstract: The aim of this study was to determine the antimicrobial effect of Helichrysum arenarium L. essential oil in "in-vitro" condition on the growth of seven microbial species including Bacillus subtilis, Escherichia coli, Staphylococcus aureus, Saccharomyces cereviciae, Candida albicans, Aspergillus flavus and Aspergillus parasiticus using micro-dilution method. The minimum inhibitory concentration (MIC) and minimum bactericidal or fungicidal concentration (MBC, MFC) were determined for the essential oil at ten concentrations. Finally, the sensitivity of tested microbes to essential oil of H. arenarium was investigated. Results showed that Bacillus subtilis (MIC=781.25 and MBC=6250 µg/ml) was more resistance than two other bacterial species. Among the tested yeasts, Saccharomyces cereviciae (MIC=97.65 and MFC=781.25 µg/ml) was more sensitive than Candida albicans while among the fungal species, growth of Aspergillus parasiticus inhibited at lower concentration of oil than the Aspergillus flavus. The extracted essential oil exhibited the same MIC value in the liquid medium against all fungal strains (48.82 µg/ml), while different activity against A. flavus and A. parasiticus was observed in this medium with MFC values of 6250 and 390.625µg/ml, respectively. Abstract: Endophytic microorganisms are presented in plants of different families growing in the foothills and piedmont plains of Trans-Ili Alatau. It was found that the maximum number of endophytic micromycetes is typical to the Fabaceae family. The number of microscopic fungi in the roots reached (145.9±5.9)×103 CFU/g of plant tissue; yeasts - (79.8±3.5)×102 CFU/g of plant tissue. Basically, endophytic microscopic fungi are typical for underground parts of plants. In contrast, yeasts more infected aboveground parts of plants. Small amount of micromycetes is typical to inflorescence and fruits. Antagonistic activity of selected micromycetes against Fusarium graminearum, Cladosporium sp., Phytophtora infestans and Botrytis cinerea phytopathogens was detected. Strains with a broad, narrow and limited range of action were identified. For further investigations Rh2 and T7 strains were selected, they are characterized by a broad spectrum of fungicidal activity and they formed the large inhibition zones against phytopathogens. Active antagonists are attributed to the Rhodotorula mucilaginosa and Beauveria bassiana species.
Abstract: In this paper, the elasto-plastic and cyclic torsion of a shaft is studied using a finite element method. The Prager kinematic hardening theory of plasticity with the Ramberg and Osgood stress-strain equation is used to evaluate the cyclic loading behavior of the shaft under the torsional loading. The material of shaft is assumed to follow the non-linear strain hardening property based on the Prager model. The finite element method with C1 continuity is developed and used for solution of the governing equations of the problem. The successive substitution iterative method is used to calculate the distribution of stresses and plastic strains in the shaft due to cyclic loads. The shear stress, effective stress, residual stress and elastic and plastic shear strain distribution are presented in the numerical results.
Abstract: The present study consisted of an applied test in meat
system to assess the effectiveness of three bio agents bacteriocinproducing
strains: Lm24: Lactobacillus sakei, Lm14and Lm25:
Pediococcus spp. Two tests were carried out: The ex situ test was
intended for three batches added with crude bacteriocin solutions at
12.48 AU/ml for Lm25 and 8.4 AU/ml for Lm14 and Lm24. However, the
in situ one consisted of four batches; three of them inoculated with
one bacteriocinogenic Lm25, Lm14, Lm24, respectively. The fourth one
was used in mixture: Lm14+m24 at approximately of 107 CFU/ml. The
two used tests were done in the presence of the pathogen
St. aureus ATCC 6538, as a test strain at 103 CFU/ml. Another batch
served as a positive or a negative control was used too. The
incubation was performed at 7°C. Total viable counts, staphylococci
and lactic acid bacteria, at the beginning and at selected times with
interval of three days were enumerated. Physico-chemical
determinations (except for in situ test): pH, dry mater, sugars, fat and
total protein, at the beginning and at end of the experiment, were
done, according to the international norms. Our results confirmed the
ex situ effectiveness. Furthermore, the batches affected negatively the
total microbial load over the incubation days, and showed a
significant regression in staphylococcal load at day seven, for Lm14,
Lm24, and Lm25 of 0.73, 2.11, and 2.4 log units. It should be noticed
that, at the last day of culture, staphylococcal load was nil for the
three batches. In the in situ test, the cultures displayed less inhibitory
attitude and recorded a decrease in staphylococcal load, for Lm14,
Lm24, Lm25, Lm14+m24 of 0.73, 0.20, 0.86, 0.032 log units. Therefore,
physicochemical analysis for Lm14, Lm24, Lm25, Lm14+m24 showed an
increase in pH from 5.50 to 5.77, 6.18, 5.96, 7.22, a decrease in dry
mater from 7.30% to 7.05%, 6.87%, 6.32%, 6.00%.This result
reflects the decrease in fat ranging from 1.53% to 1.49%, 1.07%,
0.99%, 0.87%; and total protein from 6.18% to 5.25%, 5.56%,
5.37%, 5.5%. This study suggests that the use of selected strains as
Lm25 could lead to the best results and would help in preserving and
extending the shelf life of lamb meat.
Abstract: In this work we describe the preparation of NanoSilver/methylcellulose hydrogel containing silver nanoparticles (NPs) for topical bactericidal applications. Highly concentrated dispersion of silver NPs as high as of 5g/L of silver with diameter of 10nm was prepared by reduction of AgNO3 via strong reducing agent NaBH4. Silver NPs were stabilized by addition of sodium polyacrylate in order to prevent their aggregation at such high concentration. This way synthesized silver NPs were subsequently incorporated into methylcellulose suspension at elevated temperature resulting in formation of NanoSilver/methylcellulose hydrogel when temperature cooled down to laboratory conditions. In vitro antibacterial activity assay proved high bactericidal and fungicidal efficiency of silver NPs alone in the form of dispersion as well as in the form of hydrogel against broad spectrum of bacteria and yeasts including highly multiresistant strains such as methicillin-resistant Staphylococcus aureus. A very low concentrations of silver as low as 0.84mg/L Ag in as-prepared dispersion gave antibacterial performance. NanoSilver/methylcellulose hydrogel showed antibacterial action at the lowest used silver concentration equal to 25mg/L. Such prepared NanoSilver/methylcellulose hydrogel represent promising topical antimicrobial formulation for treatment of burns and wounds.
Abstract: New diagnostic tools are urgently needed to interrupt the transmission of tuberculosis and multidrug-resistant tuberculosis. The microscopic-observation drug-susceptibility (MODS) assay is a rapid, accurate and simple liquid culture method to detect multidrug-resistant tuberculosis (MDR-TB). MODS were evaluated to determine a lower and same concentration of isoniazid and rifampin for detection of MDR-TB. Direct drug-susceptibility testing was performed with the use of the MODS assay. Drug-sensitive control strains were tested daily. The drug concentrations that used for both isoniazid and rifampin were at the same concentration: 0.16, 0.08 and 0.04μg per milliliter. We tested 56 M. tuberculosis clinical isolates and the control strains M. tuberculosis H37RV. All concentration showed same result. Of 53 M. tuberculosis clinical isolates, 14 were MDR-TB, 38 were susceptible with isoniazid and rifampin, 1 was resistant with isoniazid only. Abstract: The modern energy-efficient houses are often founded on a thermal insulation (TI) layer placed under the building’s RC foundation slab.The purpose of the paper is to identify the potential problems of the buildings founded on TI layer from the seismic point of view. The two main goals of the study were to assess the seismic behavior of such buildings, and to search for the critical structural parameters affecting the response of the superstructure as well as of the extruded polystyrene (XPS) layer. As a test building a multi-storeyed RC frame structure with and without the XPS layer under the foundation slab has been investigated utilizing nonlinear dynamic (time-history) and static (pushover) analyses. The structural response has been investigated with reference to the following performance parameters: i) Building’s lateral roof displacements, ii) Edge compressive and shear strains of the XPS, iii) Horizontal accelerations of the superstructure, iv) Plastic hinge patterns of the superstructure, v) Part of the foundation in compression, and vi) Deformations of the underlying soil and vertical displacements of the foundation slab (i.e. identifying the potential uplift). The results have shown that in the case of higher and stiff structures lying on firm soil the use of XPS under the foundation slab might induce amplified structural peak responses compared to the building models without XPS under the foundation slab. The analysis has revealed that the superstructure as well as the XPS response is substantially affected by the stiffness of the foundation slab.
Abstract: Mastitis is one of the most economic disease affecting dairy cows worldwide. Its classic diagnosis using bacterial culture and biochemical findings is a difficult and prolonged method. In this research, using of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) permitted identification of different microorganisms with high accuracy and rapidity (only 24 hours for microbial growth and analysis). During the application of MALDI-TOF MS, one hundred twenty strains of Staphylococcus and Streptococcus species isolated from milk of cows affected by clinical and subclinical mastitis were identified, and the results were compared with those obtained by traditional methods as API and VITEK 2 Systems. 37 of totality 39 strains (~95%) of Staphylococcus aureus (S. aureus) were exactly detected by MALDI TOF MS and then confirmed by a nuc-based PCR technique, whereas accurate identification was observed in 100% (50 isolates) of the coagulase negative staphylococci (CNS) and Streptococcus agalactiae (31 isolates). In brief, our results demonstrated that MALDI-TOF MS is a fast and truthful technique which has the capability to replace conventional identification of several bacterial strains usually isolated in clinical laboratories of microbiology.
Abstract: Malaria constitutes one of the major health problems
in Nigeria. One of the reasons attributed for the upsurge was the
development of resistance of Plasmodium falciparum and the
emergence of multi-resistant strains of the parasite to anti-malaria
drugs. A continued search for other effective, safe and cheap plantbased
anti-malaria agents thus becomes imperative in the face of
these difficulties. The objective of this study is therefore to evaluate
the in vivo anti-malarial efficacy of ethanolic extracts of
Chromolaena odorata and Androgaphis paniculata leaves. The two
plants were evaluated for their anti-malaria efficacy in vivo in a 4-day
curative test assay against Plasmodium berghei strain in mice. The
group treated with 500mg/ml dose of ethanolic extract of A.
paniculata plant showed parasite suppression with increase in Packed
Cell Volume (PCV) value except day 3 which showed a slight
decrease in PCV value. During the 4-day curative test, an increase in
the PCV values, weight measurement and zero count of Plasmodium
berghei parasite values was recorded after day 3 of drug
administration. These results obtained in group treated with A.
paniculata extract showed anti-malarial efficacy with higher
mortality rate in parasitaemia count when compared with
Chromolaena odorata group. These results justify the use of
ethanolic extracts of A. paniculata plant as medicinal herb used in
folklore medicine in the treatment of malaria.
Abstract: The goal of presented work is the development phytoremediation method targeted to cleaning environment polluted with organochlorine pesticides, based on joint application of plants and microorganisms. For this aim the selection of plants and microorganisms with corresponding capabilities towards three organochlorine pesticides (Lindane, DDT and PCP) has been carried out.
The tolerance of plants to tested pesticides and induction degree of plant detoxification enzymes by these compounds have been used as main criteria for estimating the applicability of plants in proposed technology. Obtained results show that alfalfa, maize and soybean among tested six plant species have highest tolerance to pesticides.
As a result of screening, more than 30 strains from genera Pseudomonas have been selected. As a result of GC analysis of incubation area, 11 active cultures for investigated pesticides are carefully chosen.
Abstract: Altered drug binding may be an important factor in isoniazid (INH) resistance, rather than major changes in the enzyme’s activity as a catalase or peroxidase (KatG). The identification of structural or functional defects in the mutant KatGs responsible for INH resistance remains as an area to be explored. In this connection, the differences in the binding affinity between wild-type (WT) and mutants of KatG were investigated, through the generation of three mutants of KatG, Ser315Thr [S315T], Ser315Asn [S315N], Ser315Arg [S315R] and a WT [S315]) with the help of software-MODELLER. The mutants were docked with INH using the software-GOLD. The affinity is lower for WT than mutant, suggesting the tight binding of INH with the mutant protein compared to WT type. These models provide the in silico evidence for the binding interaction of KatG with INH and implicate the basis for rationalization of INH resistance in naturally occurring KatG mutant strains of Mycobacterium tuberculosis.
Abstract: The aim of the present work was to test in vitro inhibition of food pathogens and spoilage bacteria by crude bacteriocins from autochthonous lactic acid bacteria. Thirty autochthonous lactic acid bacteria isolated previously, belonging to the genera: Lactobacillus, Carnobacterium, Lactococcus, Vagococcus, Streptococcus, and Pediococcus, have been screened by an agar spot test and a well diffusion assay against Gram-positive and Gram-negative harmful bacteria: Bacillus cereus, Bacillus subtilis ATCC 6633, Escherichia coli ATCC 8739, Salmonella typhimurium ATCC 14028, Staphylococcus aureus ATCC 6538, and Pseudomonas aeruginosa under conditions means to reduce lactic acid and hydrogen peroxide effect to select bacteria with high bacteriocinogenic potential. Furthermore, crude bacteriocins semiquantification and heat sensitivity to different temperatures (80, 95, 110°C, and 121°C) were performed. Another exploratory test concerning the response of St. aureus ATCC 6538 to the presence of crude bacteriocins was realized. It has been observed by the agar spot test that fifteen candidates were active toward Gram-positive targets strains. The secondary screening demonstrated an antagonistic activity oriented only against St. aureus ATCC 6538, leading to the selection of five isolates: Lm14, Lm21, Lm23, Lm24, and Lm25 with a larger inhibition zone compared to the others. The ANOVA statistical analysis reveals a small variation of repeatability: Lm21: 0.56%, Lm23: 0%, Lm25: 1.67%, Lm14: 1.88%, Lm24: 2.14%. Conversely, slight variation was reported in terms of inhibition diameters: 9.58± 0.40, 9.83± 0.46 and 10.16± 0.24 8.5 ± 0.40 10 mm for, Lm21, Lm23, Lm25, Lm14and Lm24, indicating that the observed potential showed a heterogeneous distribution (BMS = 0.383, WMS = 0.117). The repeatability coefficient calculated displayed 7.35%. As for the bacteriocins semiquantification, the five samples exhibited production amounts about 4.16 for Lm21, Lm23, Lm25 and 2.08 AU/ml for Lm14, Lm24. Concerning the sensitivity the crude bacteriocins were fully insensitive to heat inactivation, until 121°C, they preserved the same inhibition diameter. As to, kinetic of growth , the µmax showed reductions in pathogens load for Lm21, Lm23, Lm25, Lm14, Lm24 of about 42.92%, 84.12%, 88.55%, 54.95%, 29.97% in the second trails. Inversely, this pathogen growth after five hours displayed differences of 79.45%, 12.64%, 11.82%, 87.88%, 85.66% in the second trails, compared to the control. This study showed potential inhibition to the growth of this food pathogen, suggesting the possibility to improve the hygienic food quality.
