Abstract: Protorhus longifolia is known as a medicinal plant that has been used traditionally to treat various ailments such as hemiplegic paralysis, blood clotting related diseases, diarrhoea, heartburn, etc. The study reports a High-Performance Thin Layer Chromatography (HPTLC) fingerprint profile of Protorhus longifolia methanolic extract and its qualitative analysis of gallic acid, rutin, and quercetin. HPTLC analysis was achieved using CAMAG HPTLC system equipped with CAMAG automatic TLC sampler 4, CAMAG Automatic Developing Chamber 2 (ADC2), CAMAG visualizer 2, CAMAG Thin Layer Chromatography (TLC) scanner and visionCATS CAMAG HPTLC software. Mobile phase comprising toluene, ethyl acetate, formic acid (21:15:3) was used for qualitative analysis of gallic acid and revealed eight peaks while the mobile phase containing ethyl acetate, water, glacial acetic acid, formic acid (100:26:11:11) for qualitative analysis of rutin and quercetin revealed six peaks. HPTLC sillica gel 60 F254 glass plates (10 × 10) were used as the stationary phase. Gallic acid was detected at the Rf = 0.35; while rutin and quercetin were not evident in the extract. Further studies will be performed to quantify gallic acid in Protorhus longifolia leaves and also identify other biomarkers.
Abstract: Immunomodulators are substances that alter immune
system via dynamic regulation of messenger molecules. It can be
divided into immunostimulant and immunosuppressant. It can help to
increase immunity of people with a low immune system, and also can
help to normalize an overactive immune system. Aim of this study is
to investigate the effects of in vitro exposure to low and high doses of
several immunomodulators which include caffeine, kaloba and
quercetin on antigen-stimulated whole blood culture cytokine
production. Whole blood samples were taken from 5 healthy males
(age: 32 ± 12 years; weight: 75.7 ± 6.1 kg; BMI: 24.3 ± 1.5 kg/m2)
following an overnight fast with no vigorous activity during the
preceding 24 h. The whole blood was then stimulated with 50 μl of
100 x diluted Pediacel vaccine and low or high dose of
immunomodulators in the culture plate. After 20 h incubation (5%
CO2, 37°C), it was analysed using the Evidence Investigator to
determine the production of cytokines including IL-2, IL-4, IL-10,
IFN-γ, and IL-1α. Caffeine and quercetin showed a tendency towards
decrease cytokine production as the doses were increased. On the
other hand, an upward trend was evident with kaloba, where a high
dose of kaloba seemed to increase the cytokine production. In
conclusion, we found that caffeine and quercetin have potential as
immunosuppressant and kaloba as immunostimulant.
Abstract: The use of herbs to treat disease is accompanied with the history of human life. This research is aimed to study the anti-inflammatory and antinociceptive effects of hydroalcoholic extract of aerial parts of "Tanacetum balsamita balsamita". In the experimental studies 144 male mice are used. In the inflammatory test, animals were divided into six groups: Control, positive control (receiving Dexamethason at dose of 15mg/kg), and four experimental groups receiving Tanacetum balsamita balsamita hydroalcoholic extract at doses of 25, 50, 100 and 200mg/kg. Xylene was used to induce inflammation. Formalin was used to study the nociceptive effects. Animals were divided into six groups: control group, positive control group (receiving morphine) and four experimental groups receiving Tanacetum balsamita balsamita (Tb.) hydroalcoholic extract at doses of 25, 50, 100 and 200mg/kg. I.p. injection of drugs or normal saline was performed 30 minutes before test. The data were analyzed by using one way Variance analysis and Tukey post test.
