Abstract: In this study, we developed and simulated nano-drug delivery systems efficacy in compare to free drug prescription. Computational models can be utilized to accelerate experimental steps and control the experiments high cost. Molecular dynamics simulation (MDS), in particular NAMD was utilized to better understand the anti-cancer drug interaction with cell membrane model. Paclitaxel (PTX) and dipalmitoylphosphatidylcholine (DPPC) were selected for the drug molecule and as a natural phospholipid nanocarrier, respectively. This work focused on two important interaction parameters between molecules in terms of center of mass (COM) and van der Waals interaction energy. Furthermore, we compared the simulation results of the PTX interaction with the cell membrane and the interaction of DPPC as a nanocarrier loaded by the drug with the cell membrane. The molecular dynamic analysis resulted in low energy between the nanocarrier and the cell membrane as well as significant decrease of COM amount in the nanocarrier and the cell membrane system during the interaction. Thus, the drug vehicle showed notably better interaction with the cell membrane in compared to free drug interaction with the cell membrane.
Abstract: This study aims to evaluate the diversity of microorganisms in filters PM2.5 and PM10; and determine the genotoxic and cytotoxic activity of the complex mixture present in PM2.5 filters used in the AburrĂ¡ Valley Air Quality Monitoring Network (Colombia). The research results indicate that particulate matter PM2.5 of different monitoring stations are bacteria; however, this study of detection of bacteria and their phylogenetic relationship is not complete evidence to connect the microorganisms with pathogenic or degrading activities of compounds present in the air. Additionally, it was demonstrated the damage induced by the particulate material in the cell membrane, lysosomal and endosomal membrane and in the mitochondrial metabolism; this damage was independent of the PM2.5 concentrations in almost all the cases.
Abstract: Liver disorders are one of the major problems of the
world. Despite its frequent occurrence, high morbidity and high
mortality, its medical management is currently inadequate. This study
was designed to evaluate the hepatoprotective effect of saponin
extract of the root of Garcinia kola on the integrity of the liver of
paracetamol induced wistar albino rats. Twenty five (25) male adult
wistar albino rats were divided into five (5) groups. Group I was the
Control group that received distilled water only, group II was the
negative control that received 2 g/kg of paracetamol on the 13th day,
and group III, IV and V were pre-treated with 100, 200 and
400mg/kg of the saponin extract before inducing the liver damage on
the 13th day with 2 g/kg of paracetamol. Twenty four (24) h after
administration, the rats were sacrificed and blood samples were
collected. The serum Alanine Transaminase (ALT), Aspartate
Transaminase (AST), Alkaline Phosphatase (ALP) activities,
Bilirubin and conjugated bilirubin, glucose and protein
concentrations were evaluated. The liver was fixed immediately in
Formalin and was processed and stained in Haematoxylin and Eosin
(H&E). Administration of saponin extract from the root of Garcinia
kola significantly decreased paracetamol induced elevated enzymes
in the test group. Also histological observations showed that saponin
extract of the root of Garcinia kola exhibited a significant liver
protection against the toxicant as evident by the cells trying to return
to normal. Saponin extract from the root of Garcinia kola indicated a
protection of structural integrity of the hepatocytic cell membrane
and regeneration of the damaged liver.
Abstract: Natural antimicrobials are used to preserve foods that
can be found in plants, animals, and microorganisms. Antimicrobial
substances are natural or artificial agents that produced by
microorganisms or obtained semi/total chemical synthesis are used at
low concentrations to inhibit the growth of other microorganisms.
Food borne pathogens and spoilage microorganisms are inactivated
by the use of antagonistic microorganisms and their metabolites.
Yeasts can produce toxic proteins or glycoproteins (toxins) that cause
inhibition of sensitive bacteria and yeast species. Antimicrobial
substance producing phenotypes belonging different yeast genus
were isolated from different sources. Toxins secreted by many yeast
strains inhibiting the growth of other yeast strains. These strains show
antimicrobial activity, inhibiting the growth of mold and bacteria.
The effect of antimicrobial agents produced by yeasts can be
extremely fast, and therefore may be used in various treatment
procedures. Rapid inhibition of microorganisms is possibly caused by
microbial cell membrane lipopolysaccharide binding and in
activation (neutralization) effect. Antimicrobial agents inhibit the
target cells via different mechanisms of action.
Abstract: This work deals with the synthesis and the determination of some surface properties of a new anionic surfactant belonging to sulfonamide derivatives. The interest in this new surfactant is that its behavior in aqueous solution is interesting both from a fundamental and a practice point of view. Indeed, it is well known that this kind of surfactant leads to the formation of bilayer structures, and the microstructures obtained have applications in various fields, ranging from cosmetics to detergents, to biological systems such as cell membranes and bioreactors. The surfactant synthesized from pure n-alkane by photosulfochlorination and derivatized using N-ethanol amine is a mixture of position isomers. These compounds have been analyzed by Gas Chromatography coupled to Mass Spectrometry by Electron Impact mode (GC -MS/IE), and IR. The surface tension measurements were carried out, leading to the determination of the critical micelle concentration (CMC), surface excess and the area occupied per molecule at the interface. The foaming power has also been determined by Bartsch method, and the results have been compared to those of commercial surfactants. The stability of the foam formed has also been evaluated. These compounds show good foaming power characterized in most cases by dry foam.
Abstract: The possibility of intrinsic electromagnetic fields
within living cells and their resonant self-interaction and interaction
with ambient electromagnetic fields is suggested on the basis of a
theoretical and experimental study. It is reported that intrinsic
electromagnetic fields are produced in the form of radio-frequency
and infra-red photons within atoms (which may be coupled or
uncoupled) in cellular structures, such as the cell cytoskeleton and
plasma membrane. A model is presented for the interaction of these
photons among themselves or with atoms under a dipole-dipole
coupling, induced by single-photon or two-photon processes. This
resonance is manifested by conspicuous field amplification and it is
argued that it is possible for these resonant photons to undergo
tunnelling in the form of evanescent waves to a short range (of a few
nanometers to micrometres). This effect, suggested as a resonant
photon tunnelling mechanism in this report, may enable these fields
to act as intracellular signal communication devices and as bridges
between macromolecules or cellular structures in the cell
cytoskeleton, organelles or membrane. A brief overview of an
experimental technique and a review of some preliminary results are
presented, in the detection of these fields produced in living cell
membranes under physiological conditions.
Abstract: Transcription factor p53 has a powerful tumor
suppressing function that is associated with many cancers. However,
p53 of the molecular weight was higher make the limitation across to
skin or cell membrane. Thymidine dinucleotide (pTT), an
oligonucleotide, can activate the p53 transcription factor. pTT is a
hydrophilic and negative charge oligonucleotide, which delivery in to
cell membrane need an appropriate carrier. The aim of this study was
to improve the bioavailability of the nucleotide fragment, thymidine
dinucleotide (pTT), using elasic liposome carriers to deliver the drug
into the skin. The study demonstrate that dioleoylphosphocholine
(DOPC) incorporated with sodium cholate at molar ratio 1:1 can
archived the particle size about 220 nm. This elastic liposome could
penetration through skin from stratum corneum to whole epidermis by
confocal laser scanning microscopy (CLSM). Moreover, we observed
the the slight increase in generation of p53 by western blot.
Abstract: Topical photodynamic therapy (PDT) with
5-aminolevulinic acid (ALA) is an alternative therapy for treating
superficial cancer, especially for skin or oral cancer. ALA, a precursor
of the photosensitizer protoporphyrin IX (PpIX), is present as
zwitterions and hydrophilic property which make the low permeability
through the cell membrane. Collagen is a traditional carrier; its
molecular composed various amino acids which bear positive charge
and negative charge. In order to utilize the ion-pairs with ALA and
collagen, the study employed various pH values adjusting the net
charge. The aim of this study was to compare a series collagen form,
including solution, gel and sponge to investigate the topical delivery
behavior of ALA. The in vivo confocal laser scanning microscopy
(CLSM) study demonstrated that PpIX generation ability was different
pattern after apply for 6 h. Gel type could generate high PpIX, and
archived more deep of skin depth.
