Abstract: Application of health risk assessment methods is important in order to comprehend the risk of human exposure to heavy metals and other dangerous pollutants. Four soil samples were collected at distances of 10, 20, 30 m and the control 100 m away from the dump site at depths of 0.3, 0.6 and 0.9 m. The collected soil samples were examined for Zn, Cu, Pb, Cd and Ni using standard methods. The health risks via the main pathways of human exposure to heavy metal were detected using relevant standard equations. Hazard quotient was calculated to determine non-carcinogenic health risk for each individual heavy metal. Life time cancer risk was calculated to determine the cumulative life cancer rating for each exposure pathway. The estimated health risk values for adults and children were generally lower than the reference dose. The calculated hazard quotient for the ingestion, inhalation and dermal contact pathways were less than unity. This means that there is no detrimental concern to the health on human exposure to heavy metals in contaminated soil. The life time cancer risk 5.4 × 10-2 was higher than the acceptable threshold value of 1 × 10-4 which is reflected to have significant health effects on human exposure to heavy metals in contaminated soil. Good hygienic practices are recommended to ease the potential risk to children and adult who are exposed to contaminated soils. Also, the local authorities should be made aware of such health risks for the purpose of planning the management strategy accordingly.
Abstract: In this research, the risk assessment of radiation hazard for the Research Nuclear Reactor has been studied. In the current study, the MCNPx computational code has been used and coupled with a developed program using MATLAB software to evaluate Total Effective Dose Equivalent (TEDE) and cancer risk according to the BEIR equations for various human organs. In this study, the risk assessment of cancer has been calculated for ten years after exposure, in each of body organs of different ages and sexes. Also, the risk assessment of cancer has been calculated in each of body organs of different ages and sexes due to exposure after the retirement of the reactor staff. According to obtained results, a conservative whole-body dose rate, during a year, is 0.261 Sv and the probability the cancer risk for women is more than men and for children is more than adults. It has been shown that thyroid cancer was more possible than others.
Abstract: Associations between life events and various forms of cancers have been identified. The purpose of a recent random-effects meta-analysis was to identify studies that examined the association between adverse events associated with changes to financial status including decreased income and breast cancer risk. The same association was studied in four separate studies which displayed traits that were not consistent between studies such as the study design, location, and time frame. It was of interest to pool information from various studies to help identify characteristics that differentiated study results. Two random-effects Bayesian meta-analysis models are proposed to combine the reported estimates of the described studies. The proposed models allow major sources of variation to be taken into account, including study level characteristics, between study variance and within study variance, and illustrate the ease with which uncertainty can be incorporated using a hierarchical Bayesian modelling approach.
Abstract: The role and relative importance of intrinsic and extrinsic factors in the development of complex diseases such as cancer still remains a controversial issue. Determining the amount of variation explained by these factors needs experimental data and statistical models. These models are nevertheless based on the occurrence and accumulation of random mutational events during stem cell division, thus rendering cancer development a stochastic outcome. We demonstrate that not only individual genome sequencing is uninformative in determining cancer risk, but also assigning a unique genome sequence to any given individual (healthy or affected) is not meaningful. Current whole-genome sequencing approaches are therefore unlikely to realize the promise of personalized medicine. In conclusion, since genome sequence differs from cell to cell and changes over time, it seems that determining the risk factor of complex diseases based on genome sequence is somewhat unrealistic, and therefore, the resulting data are likely to be inherently uninformative.
Abstract: An assessment of the air quality of Győr (Hungary) was performed by determining the ambient concentrations of PM10-bound carcinogenic polycyclic aromatic hydrocarbons (cPAHs) in different seasons. A high volume sampler was used for the collection of ambient aerosol particles, and the associated cPAH compounds (benzo[a]pyrene (BaP), benzo[a]anthracene, benzofluoranthene isomers, indeno[123-cd]pyrene and dibenzo[ah]anthracene) were analyzed by a gas chromatographic method. Higher mean concentrations of total cPAHs were detected in samples collected in winter (9.62 ng/m3) and autumn (2.69 ng/m3) compared to spring (1.05 ng/m3) and summer (0.21 ng/m3). The calculated BaP toxic equivalent concentrations have also reflected that the local population appears to be exposed to significantly higher cancer risk in the heating seasons. Moreover, the concentration levels of cPAHs determined in this study were compared to other Hungarian urban sites.
Abstract: The main objective of this study was to assess the
seasonal variation of atmospheric polycyclic aromatic hydrocarbon
(PAH) concentrations associated with PM10 in an urban site of Győr,
Hungary. A total of 112 PM10 aerosol samples were collected in the
years of 2012 and 2013 and analyzed for PAHs by gas
chromatography method. The total PAH concentrations (sum of the
concentrations of 19 individual PAH compounds) ranged from 0.19
to 70.16 ng/m3 with the mean value of 12.29 ng/m3. Higher
concentrations of both total PAHs and benzo[a]pyrene (BaP) were
detected in samples collected in the heating seasons. Using BaPequivalent
potency index on the carcinogenic PAH concentration
data, the local population appears to be exposed to significantly
higher cancer risk in the heating seasons. However, the comparison
of the BaP and total PAH concentrations observed for Győr with
other cities it was found that the PAH levels in Győr generally
corresponded to the EU average.
Abstract: Breast cancer is a major health burden worldwide being a major cause of death amongst women. In this paper, Fuzzy Inference Systems (FIS) are developed for the evaluation of breast cancer risk using Mamdani-type and Sugeno-type models. The paper outlines the basic difference between Mamdani-type FIS and Sugeno-type FIS. The results demonstrated the performance comparison of the two systems and the advantages of using Sugeno- type over Mamdani-type.
Abstract: Before performing polymerase chain reactions (PCR), a feasible primer set is required. Many primer design methods have been proposed for design a feasible primer set. However, the majority of these methods require a relatively long time to obtain an optimal solution since large quantities of template DNA need to be analyzed. Furthermore, the designed primer sets usually do not provide a specific PCR product. In recent years, evolutionary computation has been applied to PCR primer design and yielded promising results. In this paper, a particle swarm optimization (PSO) algorithm is proposed to solve primer design problems associated with providing a specific product for PCR experiments. A test set of the gene CYP1A1, associated with a heightened lung cancer risk was analyzed and the comparison of accuracy and running time with the genetic algorithm (GA) and memetic algorithm (MA) was performed. A comparison of results indicated that the proposed PSO method for primer design finds optimal or near-optimal primer sets and effective PCR products in a relatively short time.
Abstract: The progress of concentrations of particular heavy metals was assessed in chosen localities in region Moravia, the Czech Republic, from 2007 to 2009. Particular metals were observed in localities with various types and characterization of zone. Pb, Ni, As and Cd were emphasized as a result of their toxicity and potential adverse health effect to the exposed population. The progress of metal concentrations and their health effects in the most polluted localities were examined. According to the results, the air pollution limit values were not exceeded. Based on the health risk assessment, the probability of developing tumorous diseases is acceptable, except for the increased probability of cancer risk from long-term exposure to As.
Abstract: The p53 tumor suppressor gene plays two important
roles in genomic stability: blocking cell proliferation after DNA
damage until it has been repaired, and starting apoptosis if the
damage is too critical. Codon 72 exon4 polymorphism (Arg72Pro) of
the P53 gene has been implicated in cancer risk. Various studies have
been done to investigate the status of p53 at codon 72 for arginine
(Arg) and proline (Pro) alleles in different populations and also the
association of this codon 72 polymorphism with various tumors. Our
objective was to investigate the possible association between P53
Arg72Pro polymorphism and susceptibility to colorectal cancer
among Isfahan and Chaharmahal Va Bakhtiari (a part of south west
of Iran) population. We investigated the status of p53 at codon 72 for
Arg/Arg, Arg/Pro and Pro/Pro allele polymorphisms in blood
samples from 145 colorectal cancer patients and 140 controls by
Nested-PCR of p53 exon 4 and digestion with BstUI restriction
enzyme and the DNA fragments were then resolved by
electrophoresis in 2% agarose gel. The Pro allele was 279 bp, while
the Arg allele was restricted into two fragments of 160 and 119 bp.
Among the 145 colorectal cancer cases 49 cases (33.79%) were
homozygous for the Arg72 allele (Arg/Arg), 18 cases (12.41%) were
homozygous for the Pro72 allele (Pro/Pro) and 78 cases (53.8%)
found in heterozygous (Arg/Pro).
In conclusion, it can be said that p53Arg/Arg genotype may be
correlated with possible increased risk of this kind of cancers in south
west of Iran.
