Abstract: In this paper, extract of papaya leaves are used as a
natural dye and combined by variations of solvent concentration
applied on DSSC (Dye-Sensitized Solar Cell). Indonesian geographic
located on the equator line occasions the magnitude of the potential
to develop organic solar cells made from extracts of chlorophyll as a
substitute for inorganic materials or synthetic dye on DSSC material.
Dye serves as absorbing photons which are then converted into
electrical energy. A conductive coated glass layer called TCO
(Transparent Conductive Oxide) is used as a substrate of electrode.
TiO2 nanoparticles as binding dye molecules, redox couple iodide/
tri-iodide as the electrolyte and carbon as the counter electrode in the
DSSC are used. TiO2 nanoparticles, organic dyes, electrolytes, and
counter electrode are arranged and combined with the layered
structure of the photo-catalyst absorption layer. Dye absorption
measurements using a spectrophotometer at 400-800 nm light
spectrum produces a total amount of chlorophyll 80.076 mg/l. The
test cell at 7 watt LED light with 5000 lux luminescence was
obtained Voc and Isc of 235.5 mV and 14 μA, respectively.
Abstract: Enterprise Architecture (EA) is a strategy that is
employed by enterprises in order to align their business and
Information Technology (IT). EA is managed, developed, and
maintained through Enterprise Architecture Implementation
Methodology (EAIM). Effectiveness of EA implementation is the
degree in which EA helps to achieve the collective goals of the
organization. This paper analyzes the results of a survey that aims to
explore the factors that affect the effectiveness of EAIM and
specifically the relationship between factors and effectiveness of the
output and functionality of EA project. The exploratory factor
analysis highlights a specific set of five factors: alignment,
adaptiveness, support, binding, and innovation. The regression
analysis shows that there is a statistically significant and positive
relationship between each of the five factors and the effectiveness of
EAIM. Consistent with theory and practice, the most prominent
factor for developing an effective EAIM is innovation. The findings
contribute to the measuring the effectiveness of EA implementation
project by providing an indication of the measurement
implementation approaches which is used by the Enterprise
Architects, and developing an effective EAIM.
Abstract: The presence of endocrine-disrupting compounds,
such as bisphenol A (BPA), in the environment can cause serious
health problems. However, there are controversial opinions. This
study investigated the reproductive, metabolic, oxidative and
immunologic-disrupting effects of bisphenol A in male rabbits.
Rabbits were divided into five groups. The first four rabbit groups
were administered oral BPA (1, 10, 50, or 100 mg/kg/day) for ten
weeks. The fifth group was administered corn oil as the vehicle. BPA
significantly decreased serum testosterone, estradiol and the free
androgen index (FAI) and significantly increased sex hormone
binding globulin (SHBG) compared with the placebo group. The
higher doses of BPA showed a significant decrease in follicular
stimulating hormone (FSH) and luteinizing hormone (LH). A
significant increase in blood glucose levels was identified in the BPA
groups. The non-significant difference in insulin levels is a novel
finding. The cumulative testicular toxicity of BPA was clearly
demonstrated by the dose-dependent decrease in absolute testes
weight, primary measures of semen quality and a significant increase
in testicular malonaldehyde (MDA). Moreover, BPA significantly
decreased total antioxidant capacity (TAC) and significantly
increased immunoglobulin G (IgG) at the highest concentration. Our
results suggest that BPA, especially at higher doses, is associated
with many adverse effects on metabolism, oxidative stress, immunity,
sperm quality and markers of androgenic action.
Abstract: Natural antimicrobials are used to preserve foods that
can be found in plants, animals, and microorganisms. Antimicrobial
substances are natural or artificial agents that produced by
microorganisms or obtained semi/total chemical synthesis are used at
low concentrations to inhibit the growth of other microorganisms.
Food borne pathogens and spoilage microorganisms are inactivated
by the use of antagonistic microorganisms and their metabolites.
Yeasts can produce toxic proteins or glycoproteins (toxins) that cause
inhibition of sensitive bacteria and yeast species. Antimicrobial
substance producing phenotypes belonging different yeast genus
were isolated from different sources. Toxins secreted by many yeast
strains inhibiting the growth of other yeast strains. These strains show
antimicrobial activity, inhibiting the growth of mold and bacteria.
The effect of antimicrobial agents produced by yeasts can be
extremely fast, and therefore may be used in various treatment
procedures. Rapid inhibition of microorganisms is possibly caused by
microbial cell membrane lipopolysaccharide binding and in
activation (neutralization) effect. Antimicrobial agents inhibit the
target cells via different mechanisms of action.
Abstract: Two new metal-based anticancer chemotherapeutic
agents, [(Ph2Sn)2(HGuO)2(phen)Cl2] 1 and [(Ph3Sn)(HGuO)(phen)]-
Cl.CH3OH.H2O 2, were designed, prepared and characterized by
analytical and spectral (IR, ESI-Mass, 1H, 13C and 119Sn NMR)
techniques. The proposed geometry of Sn(IV) in 1 and 2 is distorted
octahedral and distorted trigonal-bipyramidal, respectively. Both 1
and 2 exhibit potential cytotoxicity in vitro against MCF-7, HepG-2
and DU-145 cell lines. The intrinsic binding constant (Kb) values of 1
(2.33 × 105 M-1) and 2 (2.46 × 105 M-1) evaluated from UV-Visible
absorption studies suggest non-classical electrostatic mode of
interaction via phosphate backbone of DNA double helix. The Stern-
Volmer quenching constant (Ksv) of 1 (9.74 × 105 M-1) and 2 (2.9 ×
106 M-1) determined by fluorescence studies suggests the groove
binding and intercalation mode for 1 and 2, respectively. Effective
cleavage of pBR322 DNA is induced by 1.Their interaction with
DNA of cancer cells may account for potency.
Abstract: This article describes to what extent the addition of energy by-products into the structures of the technical hemp filling materials influence their properties. The article focuses on the changes in physical-mechanical and thermal technical properties of materials after the addition of ash or FBC ash or slag in the binding component of material. Technical hemp filling materials are made of technical hemp shives bonded by the mixture of cement and dry hydrate lime. They are applicable as fillers of vertical or horizontal structures or roofs. The research used eight types of energy by-products of power or heating plants in the Czech Republic. Secondary energy products were dispensed in three different percentage ratios as a replacement of cement in the binding component. Density, compressive strength and determination of the coefficient of thermal conductivity after 28, 60 and 90 days of curing in a laboratory environment were determined and subsequently evaluated on the specimens produced.
Abstract: We developed new processes which can collect and detect rapidly airborne pathogens such as the avian flu virus for the pandemic prevention. The fluorescence antibody technique is known as one of high-sensitive detection methods for viruses, but this needs up to a few hours to bind sufficient fluorescence dyes to viruses for detection. In this paper, we developed a mist-labeling can detect substitution viruses in a short time to improve the binding rate of fluorescent dyes and substitution viruses by the micro reaction process. Moreover, we developed the rapid detection system with the above “mist labeling”. The detection system set with a sampling bag collecting patient’s breath and a cartridge can detect automatically pathogens within 10 minutes.
Abstract: Altered drug binding may be an important factor in isoniazid (INH) resistance, rather than major changes in the enzyme’s activity as a catalase or peroxidase (KatG). The identification of structural or functional defects in the mutant KatGs responsible for INH resistance remains as an area to be explored. In this connection, the differences in the binding affinity between wild-type (WT) and mutants of KatG were investigated, through the generation of three mutants of KatG, Ser315Thr [S315T], Ser315Asn [S315N], Ser315Arg [S315R] and a WT [S315]) with the help of software-MODELLER. The mutants were docked with INH using the software-GOLD. The affinity is lower for WT than mutant, suggesting the tight binding of INH with the mutant protein compared to WT type. These models provide the in silico evidence for the binding interaction of KatG with INH and implicate the basis for rationalization of INH resistance in naturally occurring KatG mutant strains of Mycobacterium tuberculosis.
Abstract: The study reports about the influence of binding of orthosteric ligands as well as point mutations on the conformational dynamics of β-2-adrenoreceptor. Using molecular dynamics simulation we found that there was a little fraction of active states of the receptor in its apo (ligand free) ensemble corresponded to its constitutive activity. Analysis of MD trajectories indicated that such spontaneous activation of the receptor is accompanied by the motion in intracellular part of its alpha-helices. Thus receptor’s constitutive activity directly results from its conformational dynamics. On the other hand the binding of a full agonist resulted in a significant shift of the initial equilibrium towards its active state. Finally, the binding of the inverse agonist stabilized the receptor in its inactive state. It is likely that the binding of inverse agonists might be a universal way of constitutive activity inhibition in vivo. Our results indicate that ligand binding redistribute pre-existing conformational degrees of freedom (in accordance to the Monod-Wyman-Changeux-Model) of the receptor rather than cause induced fit in it. Therefore, the ensemble of biologically relevant receptor conformations is encoded in its spatial structure, and individual conformations from that ensemble might be used by the cell in conformity with the physiological behavior.
Abstract: In this paper we report the quantitative structure activity relationship of novel bis-triazole derivatives for predicting the activity profile. The full model encompassed a dataset of 46 Bis- triazoles. Tripos Sybyl X 2.0 program was used to conduct CoMSIA QSAR modeling. The Partial Least-Squares (PLS) analysis method was used to conduct statistical analysis and to derive a QSAR model based on the field values of CoMSIA descriptor. The compounds were divided into test and training set. The compounds were evaluated by various CoMSIA parameters to predict the best QSAR model. An optimum numbers of components were first determined separately by cross-validation regression for CoMSIA model, which were then applied in the final analysis. A series of parameters were used for the study and the best fit model was obtained using donor, partition coefficient and steric parameters. The CoMSIA models demonstrated good statistical results with regression coefficient (r2) and the cross-validated coefficient (q2) of 0.575 and 0.830 respectively. The standard error for the predicted model was 0.16322. In the CoMSIA model, the steric descriptors make a marginally larger contribution than the electrostatic descriptors. The finding that the steric descriptor is the largest contributor for the CoMSIA QSAR models is consistent with the observation that more than half of the binding site area is occupied by steric regions.
Abstract: This paper is focused on the investigation of productivity (total productivity and partial productivity). The value productivity is an indicator of level and changes in technical economic efficiency of production factors. It represents an important factor in achieving corporate objectives. This text works with the contemporary concept of value productivity that means that indicators of the productivity express the effect of economic efficiency not only of inputs consumption, but also of inputs binding efficiency. This approach is based on principles of the economic profit, respectively the economic value added (EVA). The research is done on the sample of Czech enterprises operating in the automotive industry in the regions of Liberec and the Central Bohemia. The data sample covers the time period 2006-2011 which allows the comparison of development before crisis and during crisis period. It enables to discover the companies' reaction during crises and the regional comparison allows to showing if there are significant differences between regions.
Abstract: The bound state energy of three quark systems is studied in the framework of a non- relativistic spin independent phenomenological model. The hyper- spherical coordinates are considered for the solution this system. According to Jacobi coordinate, we determined the bound state energy for (uud) and (ddu) quark systems, as quarks are flavorless mass, and it is restrict that choice potential at low and high range in nucleon bag for a bound state.
Abstract: Timber-concrete structures were recently introduced in Brazil as a viable option for bridge construction on side roads. Binding between timber and concrete is fundamentally important to assure the rigidity and performance of this structural system. The objective of this study was to assess the structural performance of a timber-concrete bridge prototype with width of 170cm and span of 400cm, whose binding among timber beams and concrete slabs was made with metal pins, obtained from CA 50 construction steel bars of 12.5mm diameter. It was possible to conclude, from the results obtained experimentally in laboratory, that the timber-concrete bridge prototype showed a good structural performance. This structural system provides an economical, rapid implementation solution, which may be used on side roads, favoring regional integration and agricultural production flow.
Abstract: Various cis-regulatory module (CRM) predictors have been proposed in the last decade. Several well-established CRM predictors adopted different categories of prediction strategies, including window clustering, probabilistic modeling and phylogenetic footprinting. Appropriate integration of them has a potential to achieve high quality CRM prediction. This study analyzed four existing CRM predictors (ClusterBuster, MSCAN, CisModule and MultiModule) to seek a predictor combination that delivers a higher accuracy than individual CRM predictors. 465 CRMs across 140 Drosophila melanogaster genes from the RED fly database were used to evaluate the integrated CRM predictor proposed in this study. The results show that four predictor combinations achieved superior performance than the best individual CRM predictor.
Abstract: In May 2009, the International Maritime Organization (IMO) adopted the Hong Kong International Convention for the Safe and Environmentally Sound Recycling of Ships to address the growing concerns about the environmental, occupational health and safety risks related to ship recycling. The aim of the Hong Kong Convention is to provide a legally binding instrument which ensures that the process of ship recycling does not pose risks to human health, safety and to the environment. In this paper, critical analysis of the Hong Kong Convention has been carried out in order to study the effectiveness of the Convention to meet its objectives. The Convention has been studied in detail including its background, main features, major stakeholders, strengths and weaknesses. The Convention, though having several deficiencies, is a major breakthrough in not only recognizing but also dealing with the ill-practices associated with ship recycling.
Abstract: In this study, WC-Ni-based cemented carbides having different nickel contents were used as the substrate for cutting tool materials. Hardened steel was turned by a (Ti,W,Si)N-coated WC-Ni-based cemented carbide tool, and the tool wear was experimentally investigated. The following results were obtained: (1) In the (Ti,W,Si)N-coated WC-Ni-based cemented carbide, the hardness of the coating film was not much different from the content of the binding material, Ni, and the adhesion strength increased with a decrease in Ni content. (2) There is little difference between the wear progress of the (Ti,W,Si)N-coated WC-7%Ni-based cemented carbide tool and that of the (Ti,W,Si)N-coated WC-6%Co-based cemented carbide tool. (3) The wear progress of the (Ti,W,Si)N-coated WC-Ni-based cemented carbide became slower with a decrease in Ni content.
From the above, it is has become clear that WC-Ni-based cemented carbide can be used as a substrate for cutting tool materials.
Abstract: An expressed sequence tag (EST) analysis provideus portions of expressed genes. We have constructed cDNA library and determined randomly sequences from cDNA library clones of T. molitor injected with acholeplasma lysate. We identified the homologous to a galectin gene. As the result of cloning and characterization of novel, we found that the protein has an open reading frame (ORF) of 495 bp, with 164 amino acid residues and molecular weight of 18.5 kDa. To characterize the role of novel Tm-galectin in immune system, we quantified the mRNA level of galectin at different times after treatment with immune elicitors. The galectin mRNA was up-regulated about 7-folds within 18 hrs. This suggests that Tm-galectin is a novel member of animal lectins, and has a role in the process of pathogen recognition. Our study would be helpful for the study on immune defense system and signaling cascade.
Abstract: Mammalian genomes contain large number of
retroelements (SINEs, LINEs and LTRs) which could affect
expression of protein coding genes through associated transcription
factor binding sites (TFBS). Activity of the retroelement-associated
TFBS in many genes is confirmed experimentally but their global
functional impact remains unclear. Human SINEs (Alu repeats) and
mouse SINEs (B1 and B2 repeats) are known to be clustered in GCrich
gene rich genome segments consistent with the view that they
can contribute to regulation of gene expression. We have shown
earlier that Alu are involved in formation of cis-regulatory modules
(clusters of TFBS) in human promoters, and other authors reported
that Alu located near promoter CpG islands have an increased
frequency of CpG dinucleotides suggesting that these Alu are
undermethylated. Human Alu and mouse B1/B2 elements have an
internal bipartite promoter for RNA polymerase III containing
conserved sequence motif called B-box which can bind basal
transcription complex TFIIIC. It has been recently shown that TFIIIC
binding to B-box leads to formation of a boundary which limits
spread of repressive chromatin modifications in S. pombe. SINEassociated
B-boxes may have similar function but conservation of
TFIIIC binding sites in SINEs located near mammalian promoters
has not been studied earlier. Here we analysed abundance and
distribution of retroelements (SINEs, LINEs and LTRs) in annotated
sequences of the Database of mammalian transcription start sites
(DBTSS). Fractions of SINEs in human and mouse promoters are
slightly lower than in all genome but >40% of human and mouse
promoters contain Alu or B1/B2 elements within -1000 to +200 bp
interval relative to transcription start site (TSS). Most of these SINEs
is associated with distal segments of promoters (-1000 to -200 bp
relative to TSS) indicating that their insertion at distances >200 bp
upstream of TSS is tolerated during evolution. Distribution of SINEs
in promoters correlates negatively with the distribution of CpG
sequences. Using analysis of abundance of 12-mer motifs from the
B1 and Alu consensus sequences in genome and DBTSS it has been
confirmed that some subsegments of Alu and B1 elements are poorly
conserved which depends in part on the presence of CpG
dinucleotides. One of these CpG-containing subsegments in B1
elements overlaps with SINE-associated B-box and it shows better
conservation in DBTSS compared to genomic sequences. It has been
also studied conservation in DBTSS and genome of the B-box
containing segments of old (AluJ, AluS) and young (AluY) Alu
repeats and found that CpG sequence of the B-box of old Alu is
better conserved in DBTSS than in genome. This indicates that Bbox-
associated CpGs in promoters are better protected from
methylation and mutation than B-box-associated CpGs in genomic
SINEs. These results are consistent with the view that potential
TFIIIC binding motifs in SINEs associated with human and mouse
promoters may be functionally important. These motifs may protect
promoters from repressive histone modifications which spread from
adjacent sequences. This can potentially explain well known
clustering of SINEs in GC-rich gene rich genome compartments and
existence of unmethylated CpG islands.

Abstract: Integrins are a large family of multidomain α/β cell
signaling receptors. Some integrins contain an additional inserted I
domain, whose earliest expression appears to be with the chordates,
since they are observed in the urochordates Ciona intestinalis (vase
tunicate) and Halocynthia roretzi (sea pineapple), but not in integrins
of earlier diverging species. The domain-s presence is viewed as a
hallmark of integrins of higher metazoans, however in vertebrates,
there are clearly three structurally-different classes: integrins without
I domains, and two groups of integrins with I domains but separable
by the presence or absence of an additional αC helix. For example,
the αI domains in collagen-binding integrins from Osteichthyes
(bony fish) and all higher vertebrates contain the specific αC helix,
whereas the αI domains in non-collagen binding integrins from
vertebrates and the αI domains from earlier diverging urochordate
integrins, i.e. tunicates, do not. Unfortunately, within the early
chordates, there is an evolutionary gap due to extinctions between the
tunicates and cartilaginous fish. This, coupled with a knowledge gap
due to the lack of complete genomic data from surviving species,
means that the origin of collagen-binding αC-containing αI domains
remains unknown. Here, we analyzed two available genomes from
Callorhinchus milii (ghost shark/elephant shark; Chondrichthyes –
cartilaginous fish) and Petromyzon marinus (sea lamprey;
Agnathostomata), and several available Expression Sequence Tags
from two Chondrichthyes species: Raja erinacea (little skate) and
Squalus acanthias (dogfish shark); and Eptatretus burgeri (inshore
hagfish; Agnathostomata), which evolutionary reside between the
urochordates and osteichthyes. In P. marinus, we observed several
fragments coding for the αC-containing αI domain, allowing us to
shed more light on the evolution of the collagen-binding integrins.
