Abstract: Soil enzyme activities in Kasuga-yama Hill Primeval Forest (Nara, Japan) were examined to determine levels of mineralization and metabolism. Samples were selected from the soil surrounding laurel-leaved (BB-1) and Carpinus japonica (BB-2 and Pw) trees for analysis. Cellulase, β-xylosidase, and protease activities were higher in BB-1 samples those in BB-2 samples. These activity levels corresponded to the distribution of cellulose and hemicellulose in the soil horizons. Cellulase, β-xylosidase, and chymotrypsin activities were higher in soil from the Pw forest than in that from the BB-2 forest. The relationships between the soil enzymes calculated by Spearman’s rank correlation indicate that the interactions between enzymes in BB-2 samples were more complex than those in Pw samples.
Abstract: The result of suggestion for low energy diet (1,000-1,200 kcal) in obese women to the effect on body weight, waist circumference and body mass index (BMI) in this experiment. Quisi experimental research was used for this study and it is a One-group pretest-posttest designs measurement method. The aim of this study was body weight, waist circumference and body mass index (BMI) reduction by using low energy diet (1,000-1,200 kcal) in obese women, the result found that in 15 of obese women that contained their body mass index (BMI) ≥ 30, after they obtained low energy diet (1,000-1,200 kcal) within 2 weeks. The data were collected before and after of testing the results showed that the average of body weight decrease 3.4 kilogram, waist circumference value decrease 6.1 centimeter and the body mass index (BMI) decrease 1.3 kg.m2 from their previous body weight, waist circumference and body mass index (BMI) before experiment started. After this study, the volunteers got healthy and they can choose or select some food for themselves. For this study, the research can be improved for data development for forward study in the future.
Abstract: The main cause of several neurodegenerative diseases such as Alzhemier, Parkinson and spongiform encephalopathies is formation of amyloid fibrils and plaques in proteins. We have analyzed different sets of proteins and peptides to understand the influence of sequence based features on protein aggregation process. The comparison of 373 pairs of homologous mesophilic and thermophilic proteins showed that aggregation prone regions (APRs) are present in both. But, the thermophilic protein monomers show greater ability to ‘stow away’ the APRs in their hydrophobic cores and protect them from solvent exposure. The comparison of amyloid forming and amorphous b-aggregating hexapeptides suggested distinct preferences for specific residues at the six positions as well as all possible combinations of nine residue pairs. The compositions of residues at different positions and residue pairs have been converted into energy potentials and utilized for distinguishing between amyloid forming and amorphous b-aggregating peptides. Our method could correctly identify the amyloid forming peptides at an accuracy of 95-100% in different datasets of peptides.
Abstract: Despite the advances made in various new
technologies, application of these technologies for agriculture still
remains a formidable task, as it involves integration of diverse
domains for monitoring the different process involved in agricultural
management. Advances in ambient intelligence technology represents
one of the most powerful technology for increasing the yield of
agricultural crops and to mitigate the impact of water scarcity,
climatic change and methods for managing pests, weeds and diseases.
This paper proposes a GPS-assisted, machine to machine solutions
that combine information collected by multiple sensors for the
automated management of paddy crops. To maintain the economic
viability of paddy cultivation, the various techniques used in
agriculture are discussed and a novel system which uses ambient
intelligence technique is proposed in this paper. The ambient
intelligence based agricultural system gives a great scope.
Abstract: Microbes have been used to solve environmental
problems for many years. The role of microorganism to sequester,
precipitate or alter the oxidation state of various heavy metals has
been extensively studied. Treatment using microorganism interacts
with toxic metal are very diverse. The purpose of this research is to
remove the mercury using Pseudomonas putida (P. putida), pure
culture ATTC 49128 at optimum growth parameters such as
techniques of culture, acclimatization time and speed of incubator
shaker. Thus, in this study, the optimum growth parameters of P.
putida were obtained to achieve the maximum of mercury removal.
Based on the optimum parameters of P. putida for specific growth
rate, the removal of two different mercury concentration, 1 ppm and
4 ppm were studied. From mercury nitrate solution, a mercuryresistant
bacterial strain which is able to reduce from ionic mercury
to metallic mercury was used to reduce ionic mercury. The overall
levels of mercury removal in this study were between 80% and 89%.
The information obtained in this study is of fundamental for
understanding of the survival of P. putida ATTC 49128 in mercury
solution. Thus, microbial mercury removal is a potential
bioremediation for wastewater especially in petrochemical industries
in Malaysia.
Abstract: Essential oils have a significant antimicrobial activity.
These oils can successfully replace the antibiotics. So, the
microorganisms show their inefficiencies resistant for the antibiotics.
For this reason, we study the physicochemical analysis and
antimicrobial activity of the essential oil of Daucus carota. The
extraction is done by steam distillation of water which brought us a
very significant return of 4.65%. The analysis of the essential oil is
performed by GC / MS and has allowed us to identify 32 compounds
in the oil of D. carota flowering tops of Bouira. Three of which are in
the majority are the α-Pinene (22.3%), the carotol (21.7%) and the
limonene (15.8%).
Abstract: Paraffinic oils were submitted to microbial action. The
microorganisms consisted of bacteria of the genera Pseudomonas sp.
and Bacillus lincheniforms. The alterations in interfacial tension were
determined using a tensometer and applying the hanging drop
technique at room temperature (299 K ±275 K). The alteration in the
constitution of the paraffins was evaluated by means of gas
chromatography. The microbial activity was observed to reduce
interfacial tension by 54 to 78%, as well as consuming the paraffins
C19 to C29 and producing paraffins C36 to C44. The LIFirr technique
made it possible to determine the microbial action quickly.
Abstract: Lipases constitute one of the most important groups of
industrial enzymes that catalyze the hydrolysis of triacylglycerol to
glycerol and fatty acids. Muscarinic antagonist relieves smooth
muscle spasm of the gastrointestinal tract and effect on the
cardiovascular system. In this research the effect of a muscarinic
antagonist on the lipase activity of Pseudomonas aeruginosa was
studied. Lineweaver–Burk plot showed that the drug inhibited the
enzyme by competitive inhibition. The IC50 value (0.16 mM) and Ki
(0.03 mM) of the drug revealed the drug bound to enzyme with high
affinity. Determination of enzyme activity in various pH and
temperature showed that the maximum activity of lipase was at pH 8
and 60oC both in presence and absence of the drug.
Abstract: Polycyclic aromatic hydrocarbons (PAHs) are formed
during the pyrolysis of scrap tyres to produce tyre pyrolytic oil
(TPO). Due to carcinogenic, mutagenic, and toxic properties PAHs
are priority pollutants. Hence it is essential to remove PAHs from
TPO before utilising TPO as a petroleum fuel alternative (to run the
engine). Agricultural wastes have promising future to be utilized as
biosorbent due to their cost effectiveness, abundant availability, high
biosorption capacity and renewability. Various low cost adsorbents
were prepared from natural sources. Uptake of PAHs present in tyre
pyrolytic oil was investigated using various low-cost adsorbents of
natural origin including sawdust (shisham), coconut fiber, neem bark,
chitin, activated charcoal. Adsorption experiments of different PAHs
viz. naphthalene, acenaphthalene, biphenyl and anthracene have been
carried out at ambient temperature (25°C) and at pH 7. It was
observed that for any given PAH, the adsorption capacity increases
with the lignin content. Freundlich constant Kf and 1/n have been
evaluated and it was found that the adsorption isotherms of PAHs
were in agreement with a Freundlich model, while the uptake
capacity of PAHs followed the order: activated charcoal> saw dust
(shisham) > coconut fiber > chitin. The partition coefficients in
acetone-water, and the adsorption constants at equilibrium, could be
linearly correlated with octanol–water partition coefficients. It is
observed that natural adsorbents are good alternative for PAHs
removal. Sawdust of Dalbergia sissoo, a by-product of sawmills was
found to be a promising adsorbent for the removal of PAHs present in
TPO. It is observed that adsorbents studied were comparable to those
of some conventional adsorbents.
Abstract: Ocimum americanum L (Lamiaceae) is an annual herb
that is native to tropical Africa. The in vitro and in vivo antioxidant
activity of its aqueous extract was carefully investigated by assessing
the DPPH radical scavenging activity, ABTS radical scavenging
activity and hydrogen peroxide radical scavenging activity. The
reducing power, total phenol, total flavonoids and flavonols content
of the extract were also evaluated. The data obtained revealed that the
extract is rich in polyphenolic compounds and scavenged the radicals
in a concentration dependent manner. This was done in comparison
with the standard antioxidants such as BHT and Vitamin C. Also, the
induction of oxidative damage with paracetamol (2000 mg/kg)
resulted in the elevation of lipid peroxides and significant (P < 0.05)
decrease in activities of superoxide dismutase, glutathione
peroxidase, glutathione reductase and catalase in the liver and kidney
of rats. However, the pretreatment of rats with aqueous extract of O.
americanum leaves (200 and 400 mg/kg) and silymarin (100 mg/kg)
caused a significant (P < 0.05) reduction in the values of lipid
peroxides and restored the levels of antioxidant parameters in these
organs. These findings suggest that the leaves of O. americanum have
potent antioxidant properties which may be responsible for its
acclaimed folkloric uses.
Abstract: A collection of thirty cultivars/clones of a red pitaya
was used to investigate flowering response to lighting
supplementation in the winter season of 2013-2014 in southern
Taiwan. The night-breaking treatment was conducted during the
period of 10 Oct. 2013 to 5 Mar. 2014 with 4-continuous hours
(22.00 – 02.00 hrs) of additional lighting daily using incandescent
bulbs (100W). Among cultivars and clones tested, twenty-three
genotypes, most belonging to the red-magenta flesh type, were found
to have positively flowering response to the lighting treatment. The
duration of night-breaking treatment for successful flowering
initiation varied from 33- 48 days. The lighting-sensitive genotypes
bore 1-2 flowering flushes. Floral and fruiting stages took 21-26 and
46-59 days, respectively. Among sixteen fruiting genotypes, the
highest fruit set rates were found in Damao 9, D4, D13, Chaozou
large, Chaozhou 5, Small Nick and F22. Five cultivars and clones
(Orejona, D4, Chaozhou large, Chaozhou 5 and Small Nick) produced
fruits with an average weight of more than 300 g per fruit which were
higher than those of the fruits formed in the summer of 2013. Fruits
produced during off-season containing total soluble solids (TSS)
from 17.5 to 20.7oBrix, which were higher than those produced inseason.
Abstract: This study is concerned with the optimization of
fermentation parameters for the hyper production of mannanase from
Fusarium oxysporum SS-25 employing two step statistical strategy
and kinetic characterization of crude enzyme preparation. The
Plackett-Burman design used to screen out the important factors in
the culture medium revealed 20% (w/w) wheat bran, 2% (w/w) each
of potato peels, soyabean meal and malt extract, 1% tryptone, 0.14%
NH4SO4, 0.2% KH2PO4, 0.0002% ZnSO4, 0.0005% FeSO4, 0.01%
MnSO4, 0.012% SDS, 0.03% NH4Cl, 0.1% NaNO3 in brewer’s spent
grain based medium with 50% moisture content, inoculated with
2.8×107 spores and incubated at 30oC for 6 days to be the main
parameters influencing the enzyme production. Of these factors, four
variables including soyabean meal, FeSO4, MnSO4 and NaNO3 were
chosen to study the interactive effects and their optimum levels in
central composite design of response surface methodology with the
final mannanase yield of 193 IU/gds. The kinetic characterization
revealed the crude enzyme to be active over broader temperature and
pH range. This could result in 26.6% reduction in kappa number with
4.93% higher tear index and 1% increase in brightness when used to
treat the wheat straw based kraft pulp. The hydrolytic potential of
enzyme was also demonstrated on both locust bean gum and guar
gum.
Abstract: Fructo-oligosaccharides (FOS) are produced from
sucrose by Aureobasidium pullulans in yields between 40-60%
(w/w). To increase the amount of FOS it is necessary to remove the
small, non-prebiotic sugars, present. Two methods for producing
high-purity FOS have been developed: the use of microorganisms
able to consume small saccharides; and the use of continuous
chromatography to separate sugars: simulated moving bed (SMB). It
is herein proposed the combination of both methods. The aim of this
study is to optimize the composition of the fermentative broth (in
terms of salts and sugars) that will be further purified by SMB. A
yield of 0.63 gFOS.gSucrose^-1 was obtained with A. pullulans using low
amounts of salts in the initial fermentative broth. By removing the
small sugars, Saccharomyces cerevisiae and Zymomonas mobilis
increased the percentage of FOS from around 56.0% to 83% (w/w) in
average, losing only 10% (w/w) of FOS during the recovery process.
Abstract: The thyroid gland is the largest classic endocrine
organ that effects many organs of the body and plays a significant
role in the process of Metabolism in animals. The aim of this study
was to investigate the prevalence of thyroid disorders diagnosed by
ultrasound and microscopic Lesions of the thyroid during the
slaughter of apparently healthy One Humped Camels (Camelus
dromedarius) in Iran. Randomly, 520 male camels (With an age
range of 4 to 8 years), were studied in 2012 to 2013. The Camels’
thyroid glands were evaluated by sonographic examination. In both
longitudinal and transverse view and then tissue sections were
provide and stained with H & E and finally examined by light
microscopy. The results obtained indicated the following:
hyperplastic goiter (21%), degenerative changes (12%), follicular
cysts (8%), follicular atrophy (4%), nodular hyperplasia (3%),
adenoma (1%), carcinoma (1%) and simple goiter colloid (1%).
Ultrasound evaluation of thyroid gland in adenoma and carcinoma
showed enlargement and irregular of the gland, decreased
echogenicity, and the heterogeneous thyroid parenchyma. Also, in
follicular cysts were observed in the enlarged gland with no echo
structures of different sizes and decreased echogenicity as a local or
general. In nodular hyperplasia, increase echogenicity and
heterogeneous parenchymal were seen. These findings suggest the
use of sonography and pathology as a screening test in the diagnosis
of complications of thyroid disorders.
Abstract: Complexation of anthocyanins to mimic natural
copigmentation process was investigated. Cyanidin-rich extracts from
Zea mays L. ceritina Kulesh. and delphinidin-rich extracts from
Clitoria ternatea L. were used to form 4 anthocyanin complexes,
AC1, AC2, AC3 and AC4, in the presence of several polyphenols and
a trace metal. Characterizations of the ACs were conducted by UV,
FTIR, DSC/TGA and morphological observations. Bathochromic
shifts of the UV spectra of 4 formulas of ACs were observed at peak
wavelengths of about 510-620 nm by 10 nm suggesting complex
formation. FTIR spectra of the ACs indicate shifts of peaks from
1,733 cm-1 to 1,696 cm-1 indicating interactions and a decrease in the
peak areas within the wavenumber of 3,400-3,500 cm-1 indicating
changes in hydrogen bonding. Thermal analysis of all of the ACs
suggests increases in melting temperature after complexation. AC
with the highest melting temperature was morphologically observed
by SEM and TEM to be crystal-like particles within a range of 50 to
200 nm. Particle size analysis of the AC by laser diffraction gave a
range of 50-600 nm, indicating aggregation. This AC was shown to
have no cytotoxic effect on cultured HGEPp0.5 and HGF (all p>
0.05) by MTT. Therefore, complexation of anthocyanins was simple
and self-assembly process, potentially resulting in nanosized particles
of anthocyanin complex.
Abstract: Anthocyanins are natural pigments with effective UV
protection but their topical use could be limited due to their
physicochemical characteristics. An attempt to overcome such
limitations by complexation of 2 major anthocyanin-rich sources, C.
ternatea and Z. mays, has potentiated its use as topical antiinflammatory.
Cell studies indicate no cytotoxicity of the
anthocyanin complex (AC) up to 1 mg/ml tested in HaCaT and
human fore head fibroblasts by MTT. Croton oil-induced ear edema
in Wistar rats suggests an effective dose of 5 mg/cm2 of AC as a
topical anti-inflammatory in comparison to 0.5 mg/cm2 of
fluocinolone acetonide. Niosomal encapsulation of the AC
significantly prolonged the anti-inflammatory activity particularly at
8 h after topical application (p = 0.0001). The AC was not cytotoxic
and its anti-inflammatory and activity was dose-dependent and
prolonged by niosomal encapsulation. It has also shown to promote
collagen type 1 production in cell culture. Thus, AC could be a
potential candidate for topical anti-inflammatory agent from natural
resources.
Abstract: Food is widely packaged with plastic materials to
prevent microbial contamination and spoilage. Ionizing radiation is
widely used to sterilize the food-packaging materials. Sterilization by
γ-radiation causes degradation such as embrittlement, stiffening,
softening, discoloration, odour generation, and decrease in molecular
weight. Many antioxidants can prevent γ-degradation but most of
them are toxic. The migration of antioxidants to its environment
gives rise to major concerns in case of food packaging plastics. In
this attempt, we have aimed to utilize synergistic mixtures of
stabilizers which are approved for food-contact applications.
Ethylene-propylene-diene terpolymer has been melt-mixed with
hindered amine stabilizers (HAS), phenolic antioxidants and organophosphites
(hydroperoxide decomposer). Results were discussed by
comparing the stabilizing efficiency of mixtures with and without
phenol system. Among phenol containing systems where we mostly
observed discoloration due to the oxidation of hindered phenol, the
combination of secondary HAS, tertiary HAS, organo-phosphite and
hindered phenol exhibited improved stabilization efficiency than
single or binary additive systems. The mixture of secondary HAS and
tertiary HAS, has shown antagonistic effect of stabilization.
However, the combination of organo-phosphite with secondary HAS,
tertiary HAS and phenol antioxidants have been found to give
synergistic even at higher doses of Gamma-irradiation. The effects
have been explained through the interaction between the stabilizers.
After γ-irradiation, the consumption of oligomeric stabilizer
significantly depends on the components of stabilization mixture. The
effect of the organo-phosphite antioxidant on the overall stability has
been discussed.
Abstract: Liver disorders are one of the major problems of the
world. Despite its frequent occurrence, high morbidity and high
mortality, its medical management is currently inadequate. This study
was designed to evaluate the hepatoprotective effect of saponin
extract of the root of Garcinia kola on the integrity of the liver of
paracetamol induced wistar albino rats. Twenty five (25) male adult
wistar albino rats were divided into five (5) groups. Group I was the
Control group that received distilled water only, group II was the
negative control that received 2 g/kg of paracetamol on the 13th day,
and group III, IV and V were pre-treated with 100, 200 and
400mg/kg of the saponin extract before inducing the liver damage on
the 13th day with 2 g/kg of paracetamol. Twenty four (24) h after
administration, the rats were sacrificed and blood samples were
collected. The serum Alanine Transaminase (ALT), Aspartate
Transaminase (AST), Alkaline Phosphatase (ALP) activities,
Bilirubin and conjugated bilirubin, glucose and protein
concentrations were evaluated. The liver was fixed immediately in
Formalin and was processed and stained in Haematoxylin and Eosin
(H&E). Administration of saponin extract from the root of Garcinia
kola significantly decreased paracetamol induced elevated enzymes
in the test group. Also histological observations showed that saponin
extract of the root of Garcinia kola exhibited a significant liver
protection against the toxicant as evident by the cells trying to return
to normal. Saponin extract from the root of Garcinia kola indicated a
protection of structural integrity of the hepatocytic cell membrane
and regeneration of the damaged liver.
Abstract: The present study was done to evaluate the presence
of tetracycline resistance genes in Lactococcus garvieae isolated
from cultured rainbow trout, West Iran. The isolates were examined
for antimicrobial resistance using disc diffusion method. Of the 49
strains tested, 19 were resistant to tetracycline (38.7%), 32 to
enrofloxacin (65.3%), 21 to erythromycin (42.8%), 20 to
chloramphenicol and trimetoprim-sulfamethoxazole (40.8%). The
strains were then characterized for their genotypic resistance profiles.
The results revealed that all 49 isolates contained at least one of the
tetracycline resistance genes. Tet (A) was found in 89.4% of
tetracycline resistant isolates and the frequency of other gene were as
follows: tet (E) 42.1%, tet (B) 47.3%, tet (D) 15.7%, tet (L) 26.3%,
tet (K) 52.6%, tet (G) 36.8%, tet (34) 21%, tet (S) 63.1%, tet (C)
57.8%, tet (M) 73.6%, tet (O) 42.1%. The results revealed high levels
of antibiotic resistance in L. garvieae strains which is a potential
danger for trout culture as well as for public health.
Abstract: Welfare may be considered to be a subjective
experience; it has a biological function that is related to the fitness
and survival of the animal accordingly, researches have suggested
that welfare is compromised when the animal's evolutionary fitness is
reduced. This study was carried out to explain the effect of some
managerial stressors as handling and restraint on behavioral patterns
and biochemical parameters of rats. A total of 24 (12 males & 12
females) Sprague-Dawley rats (12 months & 150-180g) were allotted
into 3 groups, handled group (4 male & 4 female), restrained group (4
male & 4 female) and control group (4 males & 4 females).
The obtained results revealed that time spent feeding, drinking,
movement and cage exploration frequencies increased significantly in
handled rats than other groups, while lying time and licking increased
significantly in restrained rats than handled and controls. Moreover,
social behavior decreased in both stressed groups than control.
Triglycerides were significantly increased in handled rats than
other groups, while total lipid, total protein and globulin significantly
increased in both treated groups than control. Corticosterone
increased in restrained and handled rats than control ones. Moreover,
there was an increment in packed cell volume significantly in
restrained rats than others. These deducted that if we want to study
the effect of stress on animal welfare it is necessary to study the
effect of such stressors on animal’s behavior and physiological
responses.
