Abstract: Hyaluronic acid (HA) consists of linear heteropolysaccharides repeat of D-glucuronic acid and N-acetyl-D-glucosamine. HA has various useful properties to maintain skin elasticity and moisture, reduce inflammation, and lubricate the movement of various body parts without causing immunogenic allergy. HA can be found in several animal tissues as well as in the capsule component of some bacteria including Pasteurella multocida. This study aimed to modify a genome-scale metabolic model of Escherichia coli using computational simulation and flux analysis methods to predict HA productivity under different carbon sources and nitrogen supplement by the addition of two enzymes (GLMU2 and HYAD) from P. multocida to improve the HA production under the specified amount of carbon sources and nitrogen supplements. Result revealed that threonine and aspartate supplement raised the HA production by 12.186%. Our analyses proposed the genome-scale metabolic model is useful for improving the HA production and narrows the number of conditions to be tested further.
Abstract: Kefir is a traditional fermented refreshing beverage which is known for its valuable and beneficial properties for human health. Mainly yeast species, lactic acid bacteria (LAB) strains and fewer acetic acid bacteria strains live together in a natural matrix named “kefir grain”, which is formed from various proteins and polysaccharides. Different microbial species live together in slimy kefir grain and it has been thought that synergetic effect could take place between microorganisms, which belong to different genera and species. In this research, yeast and LAB were isolated from kefir samples obtained from Uludag University Food Engineering Department. The cell morphology of isolates was screened by microscopic examination. Gram reactions of bacteria isolates were determined by Gram staining method, and as well catalase activity was examined. After observing the microscopic/morphological and physical, enzymatic properties of all isolates, they were divided into the groups as LAB and/or yeast according to their physicochemical responses to the applied examinations. As part of this research, the antagonistic/synergistic efficacy of the identified five LAB and five yeast strains to each other were determined individually by disk diffusion method. The antagonistic or synergistic effect is one of the most important properties in a co-culture system that different microorganisms are living together. The synergistic effect should be promoted, whereas the antagonistic effect is prevented to provide effective culture for fermentation of kefir. The aim of this study was to determine microbial interactions between identified yeast and LAB strains, and whether their effect is antagonistic or synergistic. Thus, if there is a strain which inhibits or retards the growth of other strains found in Kefir microflora, this circumstance shows the presence of antagonistic effect in the medium. Such negative influence should be prevented, whereas the microorganisms which have synergistic effect on each other should be promoted by combining them in kefir grain. Standardisation is the most desired property for industrial production. Each microorganism found in the microbial flora of a kefir grain should be identified individually. The members of the microbial community found in the glue-like kefir grain may be redesigned as a starter culture regarding efficacy of each microorganism to another in kefir processing. The main aim of this research was to shed light on more effective production of kefir grain and to contribute a standardisation of kefir processing in the food industry.
Abstract: The tamarind based resin containing hydroxypropane sulphonic acid groups has been synthesized and their adsorption behavior for heavy metal ions has been investigated using batch and column experiments. The hydroxypropane sulphonic acid group has been incorporated onto tamarind by a modified Porath's method of functionalisation of polysaccharides. The tamarind hydroxypropane sulphonic acid (THPSA) resin can selectively remove of heavy metal ions, which are contained in industrial wastewater. The THPSA resin was characterized by FTIR and thermogravimetric analysis. The effects of various adsorption conditions, such as pH, treatment time and adsorbent dose were also investigated. The optimum adsorption condition was found at pH 6, 120 minutes of equilibrium time and 0.1 gram of resin dose. The orders of distribution coefficient values were determined.
Abstract: Sorghum byproducts, namely bran, stalk, and panicle are examples of lignocellulosic biomass. These raw materials contain large amounts of polysaccharides, in particular hemicelluloses, celluloses, and lignins, which if efficiently extracted, can be utilised for the development of a range of added value products with potential applications in agriculture and food packaging sectors. The aim of this study was to characterise fractions extracted from sorghum bran and stalk with regards to their physicochemical properties that could determine their applicability as food-packaging materials. A sequential alkaline extraction was applied for the isolation of cellulosic, hemicellulosic and lignin fractions from sorghum stalk and bran. Lignin content, phenolic content and antioxidant capacity were also investigated in the case of the lignin fraction. Thermal analysis using differential scanning calorimetry (DSC) and X-Ray Diffraction (XRD) revealed that the glass transition temperature (Tg) of cellulose fraction of the stalk was ~78.33 oC at amorphous state (~65%) and water content of ~5%. In terms of hemicellulose, the Tg value of stalk was slightly lower compared to bran at amorphous state (~54%) and had less water content (~2%). It is evident that hemicelluloses generally showed a lower thermal stability compared to cellulose, probably due to their lack of crystallinity. Additionally, bran had higher arabinose-to-xylose ratio (0.82) than the stalk, a fact that indicated its low crystallinity. Furthermore, lignin fraction had Tg value of ~93 oC at amorphous state (~11%). Stalk-derived lignin fraction contained more phenolic compounds (mainly consisting of p-coumaric and ferulic acid) and had higher lignin content and antioxidant capacity compared to bran-derived lignin fraction.
Abstract: In order to investigate the prebiotic potential of
oligosaccharides prepared by chemical hydrolysis of water-soluble
polysaccharides (WSP) from Zizyphus lotus leaves, the effect of
oligosaccharides on bacterial growth was studied. The chemical
composition of WSP was evaluated by colorimetric assays revealed
the average values: 7.05±0.73% proteins and 86.21±0.74%
carbohydrates, among them 64.81±0.42% is neutral sugar and the rest
16.25±1.62% is uronic acids. The characterization of
monosaccharides was determined by high performance anion
exchange chromatography with pulsed amperometric detection
(HPAEC-PAD) was found to be composed of galactose (23.95%),
glucose (21.30%), rhamnose (20.28%), arabinose (9.55%), and
glucuronic acid (22.95%). The effects of oligosaccharides on the
growth of lactic acid bacteria were compared with those of fructooligosaccharide
(RP95). The oligosaccharides concentration was
1g/L of Man, Rogosa, Sharpe broth. Bacterial growth was assessed
during 2, 4.5, 6.5, 9, 12, 16 and 24 h by measuring the optical density
of the cultures at 600 nm (OD600) and pH values. During
fermentation, pH in broth cultures decreased from 6.7 to 5.87±0.15.
The enumeration of lactic acid bacteria indicated that
oligosaccharides led to a significant increase in bacteria (P≤0.05)
compared to the control. The fermentative metabolism appeared to be
faster on RP95 than on oligosaccharides from Zizyphus lotus leaves.
Both RP95 and oligosaccharides showed clear prebiotic effects, but
had differences in fermentation kinetics because of to the different
degree of polymerization. This study shows the prebiotic
effectiveness of oligosaccharides, and provides proof for the selection
of leaves of Zizyphus lotus for use as functional food ingredients.
Abstract: Superabsorbent polymers received much attention and
are used in many fields because of their superior characters to
traditional absorbents, e.g., sponge and cotton. So, it is very
important but challenging to prepare highly and fast-swelling
superabsorbents. A reliable, efficient and low-cost technique for
removing heavy metal ions from wastewater is the adsorption using
bio-adsorbents obtained from biological materials, such as
polysaccharides-based hydrogels superabsorbents. In this study, novel multi-functional superabsorbent composites
type semi-interpenetrating polymer networks (Semi-IPNs) were
prepared via graft polymerization of acrylamide onto chitosan
backbone in presence of gelatin, CTS-g-PAAm/Ge, using potassium
persulfate and N,N’-methylene bisacrylamide as initiator and
crosslinker, respectively. These hydrogels were also partially
hydrolyzed to achieve superabsorbents with ampholytic properties
and uppermost swelling capacity. The formation of the grafted
network was evidenced by Fourier Transform Infrared Spectroscopy
(ATR-FTIR) and Thermogravimetric Analysis (TGA). The porous
structures were observed by Scanning Electron Microscope (SEM).
From TGA analysis, it was concluded that the incorporation of the Ge
in the CTS-g-PAAm network has marginally affected its thermal
stability. The effect of gelatin content on the swelling capacities of
these superabsorbent composites was examined in various media
(distilled water, saline and pH-solutions). The water absorbency was
enhanced by adding Ge in the network, where the optimum value was
reached at 2 wt. % of Ge. Their hydrolysis has not only greatly
optimized their absorption capacity but also improved the swelling
kinetic.These materials have also showed reswelling ability. We
believe that these super-absorbing materials would be very effective
for the adsorption of harmful metal ions from wastewater.
Abstract: Rice bran is normally used as a raw material for rice
bran oil production or sold as feed with a low price. Conventionally,
the protein in defatted rice bran was extracted using alkaline
extraction and acid precipitation, which involves in chemical usage
and lowering some nutritious component. This study was conducted
in order to extract of rice bran protein concentrate (RBPC) from
defatted rice bran using enzymes and employing polysaccharides in a
precipitating step. The properties of RBPC obtained will be compared
to those of a control sample extracted using a conventional method.
The results showed that extraction of protein from rice bran using
enzymes exhibited the higher protein recovery compared to that
extraction with alkaline. The extraction conditions using alcalase 2%
(v/w) at 50 C, pH 9.5 gave the highest protein (2.44%) and yield
(32.09%) in extracted solution compared to other enzymes. Rice bran
protein concentrate powder prepared by a precipitation step using
alginate (protein in solution: alginate 1:0.016) exhibited the highest
protein (27.55%) and yield (6.84%). Precipitation using alginate was
better than that of acid. RBPC extracted with alkaline (ALK) or
enzyme alcalase (ALC), then precipitated with alginate (AL)
(samples RBP-ALK-AL and RBP-ALC-AL) yielded the precipitation
rate of 75% and 91.30%, respectively. Therefore, protein
precipitation using alginate was then selected. Amino acid profile of
control sample, and sample precipitated with alginate, as compared to
casein and soy protein isolated, showed that control sample showed
the highest content among all sample. Functional property study of
RBP showed that the highest nitrogen solubility occurred in pH 8-10.
There was no statically significant between emulsion capacity and
emulsion stability of control and sample precipitated by alginate.
However, control sample showed a higher of foaming capacity and
foaming stability compared to those of sample precipitated with
alginate. The finding was successful in terms of minimizing
chemicals used in extraction and precipitation steps in preparation of
rice bran protein concentrate. This research involves in a production
of value-added product in which the double amount of protein (28%)
compared to original amount (14%) contained in rice bran could be
beneficial in terms of adding to food products e.g. healthy drink with
high protein and fiber. In addition, the basic knowledge of functional
property of rice bran protein concentrate was obtained, which can be
used to appropriately select the application of this value-added
product from rice bran.
Abstract: Brown seaweeds are abundant in Portuguese coastline
and represent an almost unexploited marine economic resource. One
of the most common species, easily available for harvesting in the
northwest coast, is Saccorhiza polyschides grows in the lowest shore
and costal rocky reefs. It is almost exclusively used by local farmers
as natural fertilizer, but contains a substantial amount of valuable
compounds, particularly alginates, natural biopolymers of high
interest for many industrial applications.
Alginates are natural polysaccharides present in cell walls of
brown seaweed, highly biocompatible, with particular properties that
make them of high interest for the food, biotechnology, cosmetics
and pharmaceutical industries. Conventional extraction processes are
based on thermal treatment. They are lengthy and consume high
amounts of energy and solvents. In recent years, microwave-assisted
extraction (MAE) has shown enormous potential to overcome major
drawbacks that outcome from conventional plant material extraction
(thermal and/or solvent based) techniques, being also successfully
applied to the extraction of agar, fucoidans and alginates. In the
present study, acid pretreatment of brown seaweed Saccorhiza
polyschides for subsequent microwave-assisted extraction (MAE) of
alginate was optimized. Seaweeds were collected in Northwest
Portuguese coastal waters of the Atlantic Ocean between May and
August, 2014. Experimental design was used to assess the effect of
temperature and acid pretreatment time in alginate extraction.
Response surface methodology allowed the determination of the
optimum MAE conditions: 40 mL of HCl 0.1 M per g of dried
seaweed with constant stirring at 20ºC during 14h. Optimal acid
pretreatment conditions have enhanced significantly MAE of
alginates from Saccorhiza polyschides, thus contributing for the
development of a viable, more environmental friendly alternative to
conventional processes.
Abstract: Shellac is a natural polyester resin secreted by insects. Pectins are natural, non-toxic and water-soluble polysaccharides extracted from the peels of citrus fruits or the leftovers of apples. Both polymers are allowed for the use in the pharmaceutical industry and as a food additive. SSB Aquagold® is the aqueous solution of shellac and can be used for a coating process as an enteric or controlled drug release polymer. In this study, tablets containing 10 mg methylene blue as a model drug were prepared with a rotary press. Those tablets were coated with mixtures of shellac and one of the pectin different types (i.e. CU 201, CU 501, CU 701 and CU 020) mostly in a 2:1 ratio or with pure shellac in a small scale fluidized bed apparatus. A stable, simple and reproducible three-stage coating process was successfully developed. The drug contents of the coated tablets were determined using UV-VIS spectrophotometer. The characterization of the surface and the film thickness were performed with the scanning electron microscopy (SEM) and the light microscopy. Release studies were performed in a dissolution apparatus with a basket. Most of the formulations were enteric coated. The dissolution profiles showed a delayed or sustained release with a lagtime of at least 4 h. Dissolution profiles of coated tablets with pure shellac had a very long lagtime ranging from 13 to 17.9 h and the slopes were quite high. The duration of the lagtime and the slope of the dissolution profiles could be adjusted by adding the proper type of pectin to the shellac formulation and by variation of the coating amount. In order to apply a coating formulation as a colon delivery system, the prepared film should be resistant against gastric fluid for at least 2 h and against intestinal fluid for 4-6 h. The required delay time was gained with most of the shellac-pectin polymer mixtures. The release profiles were fitted with the modified model of the Korsmeyer-Peppas equation and the Hixson-Crowell model. A correlation coefficient (R²)> 0.99 was obtained by Korsmeyer-Peppas equation.
Abstract: This research studied the hypoglycemic effect of
water soluble polysaccharide (WSP) extracted from yam (Dioscorea
hispida) tuber by three different methods: aqueous extraction, papain
assisted extraction, and tempeh inoculums assisted extraction. The
two later extraction methods were aimed to remove WSP binding
protein to have more pure WSP. The hypoglycemic activities were
evaluated by means in vivo test on alloxan induced hyperglycemic
rats, glucose response test (GRT), in situ glucose absorption test
using everted sac, and short chain fatty acids (SCFAs) analysis. All
yam WSP extracts exhibited ability to decrease blood glucose level in
hyperglycemia condition as well as inhibited glucose absorption and
SCFA formation. The order of hypoglycemic activity was tempeh
inoculums assisted- >papain assisted- >aqueous WSP extracts. GRT
and in situ glucose absorption test showed that order of inhibition
was papain assisted- >tempeh inoculums assisted- >aqueous WSP
extracts. Digesta of caecum of yam WSP extracts oral fed rats had
more SCFA than control. Tempeh inoculums assisted WSP extract
exhibited the most significant hypoglycemic activity.
Abstract: The effects of coatings based on sodium alginate (S.A) and carboxyl methyl cellulose (CMC) on the color and moisture characteristics of potato round slices were investigated. It is the first time that this combination of polysaccharides is used as edible coating which alone had the best performance as inhibitor of potato color discoloration during the storage of 15 days at 4oC. When ascorbic acid (AA) and green tea (GT) were added in the above edible coating its effects on potato round slices changed. The mixtures of sodium alginate and carboxyl methyl cellulose with ascorbic acid or with green tea behave as a potential moisture barrier, resulting to the extent of potato samples self–life. These data suggests that both GT and AA are potential inhibitors of dehydration in potatoes and not only natural antioxidants.
Abstract: Consumer demand for products with low fat or sugar content and low levels of food additives, as well as cost factors, make exopolysaccharides (EPS) a viable alternative. EPS remain an interesting tool to modulate the sensory properties of yoghurt. This study was designed to evaluate EPS production potential of commercial yoghurt starter cultures (Yo-Flex starters: Harmony 1.0, TWIST 1.0 and YF-L902, Chr.Hansen, Denmark) and their influence on an apparent viscosity of yoghurt samples. The production of intracellularly synthesized EPS by different commercial yoghurt starters varies roughly from 144,08 to 440,81 mg/l. Analysing starters’ producing EPS, they showed large variations in concentration and supposedly composition. TWIST 1.0 had produced greater amounts of EPS in MRS medium and in yoghurt samples but there wasn’t determined significant contribution to development of texture as well as an apparent viscosity of the final product. YF-L902 and Harmony 1.0 starters differed considerably in EPS yields, but not in apparent viscosities (p>0.05) of the final yoghurts. Correlation between EPS concentration and viscosity of yoghurt samples was not established in the study.
Abstract: This study presents the performance of membrane
bioreactor in treating high phosphate wastewater. The laboratory
scale MBR was operated at permeate flux of 25 L/m2.h with a hollow
fiber membrane (polypropylene, approx. pore size 0.01 - 0.2 μm) at
hydraulic retention time (HRT) of 12 hrs. Scanning electron
microscopy (SEM) and energy diffusive X-ray (EDX) analyzer were
used to characterize the membrane foulants. Results showed that the
removal efficiencies of COD, TSS, NH3-N and PO4
3- were 93, 98, 80
and 30% respectively. On average 91% of influent soluble microbial
products (SMP) were eliminated, with the eliminations of
polysaccharides mostly above 80%. The main fouling resistance was
cake resistance. It should be noted that SMP were found in major
portions of mixed liquor that played a relatively significant role in
membrane fouling. SEM and EDX analyses indicated that the
foulants covering the membrane surfaces comprises not only organic
substances but also inorganic elements including Mg, Ca, Al, K and
P.
