Abstract: This study is concerned with the optimization of
fermentation parameters for the hyper production of mannanase from
Fusarium oxysporum SS-25 employing two step statistical strategy
and kinetic characterization of crude enzyme preparation. The
Plackett-Burman design used to screen out the important factors in
the culture medium revealed 20% (w/w) wheat bran, 2% (w/w) each
of potato peels, soyabean meal and malt extract, 1% tryptone, 0.14%
NH4SO4, 0.2% KH2PO4, 0.0002% ZnSO4, 0.0005% FeSO4, 0.01%
MnSO4, 0.012% SDS, 0.03% NH4Cl, 0.1% NaNO3 in brewer’s spent
grain based medium with 50% moisture content, inoculated with
2.8×107 spores and incubated at 30oC for 6 days to be the main
parameters influencing the enzyme production. Of these factors, four
variables including soyabean meal, FeSO4, MnSO4 and NaNO3 were
chosen to study the interactive effects and their optimum levels in
central composite design of response surface methodology with the
final mannanase yield of 193 IU/gds. The kinetic characterization
revealed the crude enzyme to be active over broader temperature and
pH range. This could result in 26.6% reduction in kappa number with
4.93% higher tear index and 1% increase in brightness when used to
treat the wheat straw based kraft pulp. The hydrolytic potential of
enzyme was also demonstrated on both locust bean gum and guar
gum.
Abstract: Taro Scarab beetles (Papuana uninodis, Coleoptera:
Scarabaeidae) inflict severe damage on important root crops and
plants such as Taro or Cocoyam, yam, sweet potatoes, oil palm and
coffee tea plants across Africa and Asia resulting in economic
hardship and starvation in some nations. Scoliid wasps and
Metarhizium anisopliae fungus - bio-control agents; are shown to be
able to control the population of Scarab beetle adults and larvae using
a newly created simulation model based on non-linear ordinary
differential equations that track the populations of the beetle life
cycle stages: egg, larva, pupa, adult and the population of the scoliid
parasitoid wasps, which attack beetle larvae. In spite of the challenge
driven by the longevity of the scarab beetles, the combined effect of
the larval wasps and the fungal bio-control agent is able to control
and drive down the population of both the adult and the beetle eggs
below the environmental carrying capacity within an interval of 120
days, offering the long term prospect of a stable and eco-friendly
environment; where the population of scarab beetles is: regulated by
parasitoid wasps and beneficial soil saprophytes.
Abstract: Drying behavior of blanched sweet potato in a cabinet
dryer using different five air temperatures (40-80°C) and ten sweet
potato varieties sliced to 5mm thickness were investigated. The
drying data were fitted to eight models. The Modified Henderson and
Pabis model gave the best fit to the experimental moisture ratio data
obtained during the drying of all the varieties while Newton (Lewis)
and Wang and Singh models gave the least fit. The values of Deff
obtained for Bophelo variety (1.27 x 10-9 to 1.77 x 10-9 m2/s) was
the least while that of S191 (1.93 x 10-9 to 2.47 x 10-9 m2/s) was the
highest which indicates that moisture diffusivity in sweet potato is
affected by the genetic factor. Activation energy values ranged from
0.27-6.54 kJ/mol. The lower activation energy indicates that drying
of sweet potato slices requires less energy and is hence a cost and
energy saving method. The drying behavior of blanched sweet potato

Abstract: This study was aimed to investigate the effect of
various organic supplements on growth and development of
Dendrobium discolor’s protocorms and seedlings growth of
Dendrobium Judy Rutz. Protocorms of Dendrobium discolor with 2.0
cm. in diameter and seedlings of Dendrobium Judy Rutz at the same
size (0.5 cm. height) were sub-cultured on Hyponex medium
supplemented with cow milk (CM), soy milk (SM), potato extract
(PE) and peptone (P) for 2 months. The protocorms were developed
to seedlings in all treatments after cultured for 2 months. However,
the best results were found on Hyponex medium supplemented with
P was the best in which the maximum fresh and dry weight and
maximum shoot height were obtained in this treatment statistically
different (p ≤ 0.05) to other treatments. Moreover, Hyponex medium
supplemented with P also stimulated the maximum mean number of
5.7 shoots per explant which also showed statistically different (p ≤
0.05) when compared to other treatments. The results of growth of
Dendrobium Judy Rutz seedlings indicated the medium
supplemented with 100 mL/L PE enhanced the maximum fresh and
dry weigh per explants with significantly different (p ≤ 0.05) in fresh
weight from other treatments including the control medium without
any organic supplementation. However, the dry weight was not
significantly different (p ≤ 0.05) from medium supplemented with
SM and P. There was multiple shoots induction in all media with or
without organic supplementation ranging from 2.6 to 3 shoots per
explants. The maximum shoot height was also obtained in the
seedlings cultured on medium supplemented with PE while the
longest root length was found in medium supplemented with SM.
Abstract: This study aimed to investigate effect of different organic supplements on growth of Vanda and Mokara seedlings. Vanda and Mokara seedlings approximately 0.2 and 0.3 cm. in height were sub-cultured onto VW supplemented with 150 ml/L coconut water, 100 g/L potato extract, 100 g/L ‘Gros Michel’ banana (AAA group) and 100 g/L ‘Namwa’ banana (ABB group). The explants were sub-cultured onto the same medium every month for 3 months. The best medium increased stem height to 0.52 and 0.44 Cm. in Vanda and Mokara respectively was supplemented with coconut water. The maximum fresh weight of Vanda (0.59 g) was found on medium supplemented with ‘Gros Michel’ banana while Mokara cultured on medium supplemented with Potato extract had the maximum fresh weight (0.27 g) and number of roots (5.20 roots/shoot) statistically different (p≤ 0.05) to other treatments. However, Vanda cultured on medium supplemented with ‘Namwa’ banana had the maximum number of roots (3.80 roots/shoot). Our results suggested that growth of different orchid genera was responded diversely to different organic supplements.
Abstract: Microbial production of antimicrobials as biopreservatives is the major area of focus nowadays due to increased interest of consumers towards natural and safe preservation of ready to eat food products. The agro-industrial byproduct based medium and optimized process conditions can contribute in economical production of bacteriocins. Keeping this in view, the present investigation was carried out on agro-industrial byproducts utilization for the production of bacteriocin using Enterococcus faecium BS13 isolated from local fermented food. Different agro-industrial byproduct based carbon sources (whey, potato starch liquor, kinnow peel, deoiledrice bran and molasses), nitrogen sources (soya okra, pea pod and corn steep liquor), metal ions and surfactants were tested for optimal bacteriocin production. The effect of various process parameters such as pH, temperature, inoculum level, agitation and time were also tested on bacteriocin production. The optimized medium containing whey, supplemented with 4%corn steep liquor and polysorbate-80 displayed maximum bacteriocin activity with 2% inoculum, at pH 6.5, temperature 40oC under shaking conditions (100 rpm).
Abstract: Humic acids (HA) were produced by a Trichoderma
viride strain under submerged fermentation in a medium based on the
oil palm empty fruit bunch (EFB) and the main variables of the
process were optimized by using response surface methodology. A
temperature of 40°C and concentrations of 50g/L EFB, 5.7g/L potato
peptone and 0.11g/L (NH4)2SO4 were the optimum levels of the
variables that maximize the HA production, within the
physicochemical and biological limits of the process. The optimized
conditions led to an experimental HA concentration of 428.4±17.5
mg/L, which validated the prediction from the statistical model of
412.0mg/L. This optimization increased about 7–fold the HA
production previously reported in the literature. Additionally, the
time profiles of HA production and fungal growth confirmed our
previous findings that HA production preferably occurs during fungal
sporulation. The present study demonstrated that T. viride
successfully produced HA via the submerged fermentation of EFB
and the process parameters were successfully optimized using a
statistics-based response surface model. To the best of our
knowledge, the present work is the first report on the optimization of
HA production from EFB by a biotechnological process, whose
feasibility was only pointed out in previous works.
Abstract: Awareness of value addition of sweet potato has received comparatively little attention in Nigeria despite its potential to reduce perishability and enhanced utilization of the crop in diverse products forms. This study assessed the awareness of value addition of sweet potato in Osun State, Nigeria. Multi-stage random sampling technique was used to select 120 respondents for the study. Data obtained were analyzed using descriptive statistics and multiple regression analysis. Findings showed that most (75.00%) of the respondents were male with mean age of 42.10 years and 96.70% of the respondents had formal education. The mean farm size was 2.30 hectares. Majority (75.00%) of the respondents had more than 10 years farming experience. Awareness of value addition of sweet potato was very low among the respondents. It was recommended that sweet potato farmers should be empowered through effective and efficient extension training on the use of modern processing techniques in order to enhance value addition of sweet potato.
Abstract: Diversification of the processing of crops is a very important way of reducing food insecurity, perishability of most perishable crops and generates verities. Sweet potato has been diversified in various ways by researchers through processing into different forms for consumption. The study considered diversifying the crop into different drinks by combining it with different high nutrient acceptable cereal. There was significant relationship between the educational background of the respondents and level of acceptability of the sweet potato drinks (χ 2 = 1.033 and P = 0.05). Interestingly, significant relationship existed between the most preferred sweet potato drink by the respondents and level of acceptability of the sweet potato drinks (r = 0.394, P = 0.031). The high level of acceptability of the drinks will lead to enhanced production of the crops required for the drinks that would assist in income generation and alleviating food and nutrition insecurity.
Abstract: There has been studied the distribution character of potato stem nematode (Ditylenchus destructor Thorne, 1945) on the potato fields in four municipalities (Tsalka, Bolnisi, Marneuli, Gardabani) of Kvemo Kartli (Eastern Georgia).
As a result of scientific research there is stated the extensiveness of pathogens invasion, accompanying composition of fauna species, environmental groups of populations and quantity.
During the research process in the studied ecosystems there were registered 160 forms of free-living and Phyto-parasitic nematodes, from which 118 forms are determined as species and 42 as genus.
It was found that in almost the entire studied ecosystem there is dominated pathogenic nematodes Ditylenchus destructor. The large number of exemplars (almost uncountable) was found in tubers material of Bolnisi and Gardabani.
Abstract: Date production in North Africa is facing a worrying slowdown and a decline because of Fusarium wilt or bayoud date palm (Phoenix dactylifera L., caused by Fusarium oxysporum f. sp. albedinis (F. o. a). The objective of this work is to study the in vitro effect of flavonoid aglycones extracted from the roots of two cultivars of date palm (one sensitive to bayoud (Deglet Nour) and the other resistant (Takerboucht)) on the growth and production fusaric acid of the pathogen. Results show that during the first week of development of F. o. a on potato dextrose liquid medium, the flavonoid aglycones extracts of the susceptible cultivar roots stimulates mycelial growth as well as conidiogenesis of F.o.a, nevertheless it has no effect on the synthesis of fusaric acid. However, the flavonoid aglycones extract of resistant cultivar roots stimulates mycelial growth and decreases both the number of conidia production and fusaric acid. It therefore appears possible that the resistant cultivar aglycones have two types of action: they either inhibit the synthesis of fusaric acid, or they metabolize this toxin into hydrosoluble product, this is called detoxification.
Abstract: Cattle manure and mineral fertilizers are two source
of Nitrogen, which can affect the growth and quantity of potato. In
this research the effects of the use of cattle manure (5, 10, 15 and 20
ton ha-1), Nitrogen fertilizer (50, 100 and 150 kg N ha-1) and their
interaction on potato growth were evaluated during field
experiments in 2008 with the help of Randomized Complete Block
(RCB) with the factorial arrangement of three experimental
replications in Iran. At the 75 th day after emergence, dry weight of
Shoots, leaf area index (LAI) and plant height were recorded. Results
showed that, dry weight of Shoots, LAI and plant height increased
linearly and very significantly in response to the application of
manure and Nitrogen fertilizer. While the interaction between
manure and Nitrogen fertilizer just on the LAI and plant height was
significant, somehow the maximum amount of plant height( 73 cm)
was obtained by using 150 kg Nitrogen + 15 tons of manure per
hectare, and maximum LAI ( 5.36) was obtained by using 150 kg
Nitrogen + 20 tons of manure per hectare. Also in this experiment
maximum tuber yield (36.8 tons ha-1) was obtained by the utilization
of 150 kg Nitrogen per hectare + 20 tons manure.
Abstract: To study the effect of suitable methods for
propagation of True Potato Seed (TPS) progenies, transplant and
selection of the best progenies, a factorial experiment base on a
randomized complete block design was carried out in the research
field of Sahneh region, Kermanshah, Iran during 2009-2010. Five
selective progenies from CIP (International Potato Center) including
CIP.994013, CIP.994002, CIP.994014, CIP.888006, and
CIP.994001 and two transplant preparation methods (Paper pot
preparation for mechanical cultivation and preparation in transplant
trays for manual cultivation) were studied in three replications.
Results showed that different progenies had no significant effect on
plant height (cm) and tuber yield (t ha-1), whereas had a significant
effect on number of tubers per unit area (m2). There was significant
difference between transplant preparation methods for plant height
and tuber yield. The interaction effect of progenies and transplant
preparation method was not significant for these traits. CIP.888006
progeny and paper pot preparation method produced the highest
tuber yields. Also CIP.994002 and CIP.994014 progenies considered
as the best progenies under paper pot preparation method due to high
yields.
Abstract: Using strength Pulse Electrical Field (PEF) in food
industries is a non-thermal process that can deactivate
microorganisms and increase penetration in plant and animals tissues
without serious impact on food taste and quality. In this paper designing and fabricating of a PEF generator has been presented. Pulse generation methods have been surveyed and the best of them
selected. The equipment by controller set can generate square pulse with adjustable parameters such as amplitude 1-5kV, frequency 0.1-10Hz, pulse width 10-100s, and duty cycle 0-100%. Setting the number of pulses, and presenting the output voltage and current
waveforms on the oscilloscope screen are another advantages of this
equipment. Finally, some food samples were tested that yielded the satisfactory results. PEF applying had considerable effects on potato, banana and purple cabbage. It caused increase Brix factor from 0.05
to 0.15 in potato solution. It is also so effective in extraction color material from purple cabbage. In the last experiment effects of PEF
voltages on color extraction of saffron scum were surveyed (about 6% increasing yield).
Abstract: Potatoes are a good source of carotenoids, which are lipophilic compounds synthesized in plastids from isoprenoids. The aim of this research was to determine the content of carotenoids in relationship with the colour of organically and conventionally cultivated potato genotypes before and after period of storage. In cooperation with the State Priekuli Plant Breeding Institute (Latvia), six potato genotypes were studied: 'Agrie dzeltenie', 'Prelma', 'Imanta', 'S-03135-10', 'S-99108-8' and 'S-01063-5'. All the genotypes were cultivated under three different conditions: organically and conventionally (two conditions). The content of carotenoids was determined by using spectrophotometer and the colour – L*a*b* system. The results of current research show that after the period of storage, carotenoid amount has increased and in conventionally cultivated potatoes it varies from 228.514 to 552.434 μg 100 g-1 while in organically cultivated potato genotypes – from 45.485 to 662.699 μg 100 g-1 FW. Colour of potato flesh was changing during storage.
Abstract: The purpose of study was to design and construction
the semi-automatic sliced ginger machine for reduce production times
in sheet and slice ginger procedure furthermore, reduced amount of
labor of slides and cutting method. Take consider into clean and safety of workers and consumers. The principle of machines, used 1
horsepower motor, rotation speed of sliced blade 967 rpm, the diameter of sliced dish 310 mm, consists of 2 blades for sheet cutting
ginger and the power from motor which transfer to rotate the sliced blade roller, rotation speed 440 rpm. The slice cutter roller was sliced
ginger from sheet ginger to line ginger. The conveyer could
adjustment level of motors, used to the beginning area that sheet
ginger was transference to the roller for sheet and sliced cutting in next process. The cover of sliced cutting had channel for 1 tuber of
ginger. The semi-automatic sliced ginger machine could produced sheet ginger 81.8 kg/h (6.2 times of labor) and line ginger 17.9 kg/h
(2.5 times of labor) compare with, labor work could produced sheet
ginger 13.2 kg/h and line ginger 7.1 kg/h, and when timekeeper, the
total times of semi auto machine 30.86 kg/h and labor 4.6 kg/h, there
for the semi auto machine was 6.7 times of labor. The semiautomatic
sliced ginger machine convenient, easy for use and
maintain, in addition to reduce fatigue of body and seriousness from
works; must be used high skill, and protection accident in slicing
procedure. Beside, machine could used with other vegetables for
example potato, carrot .etc
Abstract: Considering the serious health hazards of air pollutants from automobiles, the present study was aimed to estimate the genotoxic/tumor inducing potential of three soil samples collected from junctions of Bus stand (BS), Crystal (CT) and Railway station (RS) of Amritsar, Punjab (India) using Allium cepa root chromosomal aberration assay (AlRCAA) and potato disc tumor assay (PDTA). The genotoxic potential in AlRCAA was 41.27% and 41.26% for BS; 37.89% and 43.38% for RS and 33.76% and 37.83% for CT during in situ and root dip treatments, respectively. The maximum number of tumors were induced in RS sample (64) followed by BS (21) and CT (9) during PDTA. The physicochemical parameters of soil sample were also studied and the concentration of lead was found to be 95.21 mg/Kg in RS, 35.30 mg/Kg in BS and 24.59 mg/Kg in CT samples.
Abstract: Enzymatic hydrolysis of starch from natural sources
finds potential application in commercial production of alcoholic
beverage and bioethanol. In this study the effect of starch
concentration, temperature, time and enzyme concentration were
studied and optimized for hydrolysis of Potato starch powder (of
mesh 80/120) into glucose syrup by immobilized (using Sodium
arginate) α-amylase using central composite design. The
experimental result on enzymatic hydrolysis of Potato starch was
subjected to multiple linear regression analysis using MINITAB 14
software. Positive linear effect of starch concentration, enzyme
concentration and time was observed on hydrolysis of Potato starch
by α-amylase. The statistical significance of the model was validated
by F-test for analysis of variance (p ≤ 0.01). The optimum value of
starch concentration, enzyme concentration, temperature, time and
were found to be 6% (w/v), 2% (w/v), 40°C and 80min respectively.
The maximum glucose yield at optimum condition was 2.34 mg/mL.
Abstract: Many of the compounds present in potato are important because of their beneficial effects on health, therefore, are highly desirable in the human diet. Potato tubers contain significant amounts of anthocyanins. The aim of this research was to determine the content of anthocyanins and its relationship with the colour of organically and conventionally cultivated potato varieties. In the research eight potato samples of three potato varieties were analyzed on anthocyanins, dry matter content and color. Abstract: From food consumption surveys has been found that potato consumption comparing to other European countries is one of the highest. Hence acrylamide (AA) intake coming from fried potatoes in population might be high as well. The aim of the research was to determine acrylamide content and estimate intake of acrylamide from roasted potatoes bred and cultivated in Latvia. Five common Latvian potato varieties were selected: Lenora, Brasla, Imanta, Zile, and Madara. A two-year research was conducted during two periods: just after harvesting and after six months of storage. Time and temperature (210 ± 5°C) was recorded during frying. AA was extracted from potatoes by solid phase extraction and AA content was determined by LC-MS/MS. estimated intake of acrylamide ranges from 0.012 to 0.496μgkg-1 BW per day.
