Abstract: Response Surface Methodology (RSM) is a powerful
and efficient mathematical approach widely applied in the
optimization of cultivation process. Cellulase enzyme production by
Trichoderma reesei RutC30 using agricultural waste rice straw and
banana fiber as carbon source were investigated. In this work,
sequential optimization strategy based statistical design was
employed to enhance the production of cellulase enzyme through
submerged cultivation. A fractional factorial design (26-2) was applied
to elucidate the process parameters that significantly affect cellulase
production. Temperature, Substrate concentration, Inducer
concentration, pH, inoculum age and agitation speed were identified
as important process parameters effecting cellulase enzyme synthesis.
The concentration of lignocelluloses and lactose (inducer) in the
cultivation medium were found to be most significant factors. The
steepest ascent method was used to locate the optimal domain and a
Central Composite Design (CCD) was used to estimate the quadratic
response surface from which the factor levels for maximum
production of cellulase were determined.
Abstract: Durian husk (DH), a fruit waste, was studied for its ability to remove Basic blue 3 (BB3) from aqueous solutions. Batch kinetic studies were carried out to study the sorption characteristics under various experimental conditions. The optimum pH for the dye removal occurred in the pH range of 3-10. Sorption was found to be concentration and agitation dependent. The kinetics of dye sorption fitted a pseudo-second order rate expression. Both Langmuir and Freundlich models appeared to provide reasonable fittings for the sorption data of BB3 on durian husk. Maximum sorption capacity calculated from the Langmuir model is 49.50 mg g-1.
Abstract: The morphology, mineralogical and chemical
composition of a low-grade nickel ore from Mpumalanga, South
Africa, were studied by scanning electron microscope (SEM), X-ray
diffraction (XRD) and X-ray fluorescence (XRF), respectively. The
ore was subjected to atmospheric agitation leaching using sulphuric
acid to investigate the effects of acid concentration, leaching
temperature, leaching time and particle size on extraction of nickel
and cobalt. Analyses results indicated the ore to be a saprolitic nickel
laterite belonging to the serpentine group of minerals. Sulphuric acid
was found to be able to extract nickel from the ore. Increased acid
concentration and temperature only produced low amounts of nickel
but improved cobalt extraction. As high as 77.44% Ni was achieved
when leaching a -106+75μm fraction with 4.0M acid concentration at
25oC. The kinetics of nickel leaching from the saprolitic ore were
studied and the activation energy was determined to be 18.16kJ/mol.
This indicated that nickel leaching reaction was diffusion controlled.
Abstract: Activated carbon was prepared from agricultural waste “almond (Prunus amygdalus) nut shells" by chemical activation with phosphoric acid as an activating agent at 450 °C for 24 hr soaking time. The physical and chemical properties were analyzed. The adsorption of chromium VI from aqueous solution on almond nut shell activated carbon (ASAC) was investigated. The adsorption process parameters pH, agitation speed, agitation time, adsorbent dose were optimized. 98% of Cr VI was sorbed at pH 2 and stirring speed 200 rpm.. Surface structure showed that ASAC has a spongy type structure showing large number of pores
Abstract: Absorptive characteristics of polyaniline synthesized
in mixture of water and acetonitrile in 50/50 volume ratio was
studied. Synthesized polyaniline in powder shape is used as an
adsorbent to remove toxic hexavalent chromium from aqueous
solutions. Experiments were conducted in batch mode with different
variables such as agitation time, solution pH and initial concentration
of hexavalent chromium. Removal mechanism is the combination of
surface adsorption and reduction. The equilibrium time for removal
of Cr(T) and Cr(VI) was about 2 and 10 minutes respectively. The
optimum pH for total chromium removal occurred at pH 7 and
maximum hexavalent chromium removal took place under acidic
condition at pH 3. Investigating the isothermal characteristics showed
that the equilibrium adsorption data fitted both Freundlich-s and
Langmuir-s isotherms. The maximum adsorption of chromium was
calculated 36.1 mg/g for polyaniline
Abstract: In this study, aerobic digestion of tannery industry
wastewater was carried out using mixed culture obtained from
common effluent treatment plant treating tannery wastewater. The
effect of pH, temperature, inoculum concentration, agitation speed
and initial substrate concentration on the reduction of organic matters
were found. The optimum conditions for COD reduction was found
to be pH - 7 (60%), temperature - 30ÔùªC (61%), inoculum
concentration - 2% (61%), agitation speed - 150rpm (65%) and initial
substrate concentration - 1560 mg COD/L (74%). Kinetics studies
were carried by using Monod model, First order, Diffusional model
and Singh model. From the results it was found that the Monod
model suits well for the degradation of tannery wastewater using
mixed microbial consortium.
Abstract: Xanthan gum is one of the major commercial
biopolymers. Due to its excellent rheological properties xanthan gum
is used in many applications, mainly in food industry. Commercial
production of xanthan gum uses glucose as the carbon substrate;
consequently the price of xanthan production is high. One of the
ways to decrease xanthan price, is using cheaper substrate like
agricultural wastes. Iran is one of the biggest date producer countries.
However approximately 50% of date production is wasted annually.
The goal of this study is to produce xanthan gum from waste date
using Xanthomonas campestris PTCC1473 by submerged
fermentation. In this study the effect of three variables including
phosphor and nitrogen amount and agitation rate in three levels using
response surface methodology (RSM) has been studied. Results
achieved from statistical analysis Design Expert 7.0.0 software
showed that xanthan increased with increasing level of phosphor.
Low level of nitrogen leaded to higher xanthan production. Xanthan
amount, increasing agitation had positive influence. The statistical
model identified the optimum conditions nitrogen amount=3.15g/l,
phosphor amount=5.03 g/l and agitation=394.8 rpm for xanthan. To
model validation, experiments in optimum conditions for xanthan
gum were carried out. The mean of result for xanthan was 6.72±0.26.
The result was closed to the predicted value by using RSM.
Abstract: This research is to design and implement a new kind
of agitators called differential agitator. The Differential Agitator is an
electro- mechanic set consists of two shafts. The first shaft is the
bearing axis while the second shaft is the axis of the quartet upper
bearing impellers group and the triple lower group which are called
as agitating group. The agitating group is located inside a cylindrical
container equipped especially to contain square directors for the
liquid entrance and square directors called fixing group for the liquid
exit. The fixing group is installed containing the agitating group
inside any tank whether from upper or lower position. The agitating
process occurs through the agitating group bearing causing a lower
pressure over the upper group leading to withdrawing the liquid from
the square directors of the liquid entering and consequently the liquid
moves to the denser place under the quartet upper group. Then, the
liquid moves to the so high pressure area under the agitating group
causing the liquid to exit from the square directors in the bottom of
the container. For improving efficiency, parametric study and shape
optimization has been carried out. A numerical analysis,
manufacturing and laboratory experiments were conducted to design
and implement the differential agitator. Knowing the material
prosperities and the loading conditions, the FEM using ANSYS11
was used to get the optimum design of the geometrical parameters of
the differential agitator elements while the experimental test was
performed to validate the advantages of the differential agitators to
give a high agitation performance of lime in the water as an example.
In addition, the experimental work has been done to express the
internal container shape in the agitation efficiency. The study ended
up with conclusions to maximize agitator performance and optimize
the geometrical parameters to be used for manufacturing the
differential agitator
Abstract: An immunomodulator bioproduct is prepared in a
batch bioprocess with a modified bacterium Pseudomonas
aeruginosa. The bioprocess is performed in 100 L Bioengineering
bioreactor with 42 L cultivation medium made of peptone, meat
extract and sodium chloride. The optimal bioprocess parameters were
determined: temperature – 37 0C, agitation speed - 300 rpm, aeration
rate – 40 L/min, pressure – 0.5 bar, Dow Corning Antifoam M-max.
4 % of the medium volume, duration - 6 hours. This kind of
bioprocesses are appreciated as difficult to control because their
dynamic behavior is highly nonlinear and time varying. The aim of
the paper is to present (by comparison) different models based on
experimental data.
The analysis criteria were modeling error and convergence rate.
The estimated values and the modeling analysis were done by using
the Table Curve 2D.
The preliminary conclusions indicate Andrews-s model with a
maximum specific growth rate of the bacterium in the range of
0.8 h-1.
Abstract: The present work presents the extraction of copper(II)
from sulphuric acid solutions with Sodium diethyldithiocarbamate
(SDDT), and six different organic diluents: Dichloromethane,
Chloroform, Carbon tetrachloride, Toluene, xylene and Cyclohexane,
were tested. The pair SDDT/Chloroform showed to be the most
selective in removing the copper cations, and hence was considered
throughout the experimental study.
The effects of operating parameters such as the initial concentration
of the extracting agent, the agitation time, the agitation speed and the
acid concentration were considered.
For an initial concentration of Cu (II) of 63 ppm in a 0.5 M sulphuric
acid solution, both with a mass of the extracting agent of 20 mg, an
extraction percentage of about 97.8 % and a distribution coefficient
of 44.42 were obtained, respectively, confirming the performance
of the SDDT-Chloroform pair.
Abstract: In this study, the sorption of Malachite green (MG) on Hydrilla verticillata biomass, a submerged aquatic plant, was investigated in a batch system. The effects of operating parameters such as temperature, adsorbent dosage, contact time, adsorbent size, and agitation speed on the sorption of Malachite green were analyzed using response surface methodology (RSM). The proposed quadratic model for central composite design (CCD) fitted very well to the experimental data that it could be used to navigate the design space according to ANOVA results. The optimum sorption conditions were determined as temperature - 43.5oC, adsorbent dosage - 0.26g, contact time - 200min, adsorbent size - 0.205mm (65mesh), and agitation speed - 230rpm. The Langmuir and Freundlich isotherm models were applied to the equilibrium data. The maximum monolayer coverage capacity of Hydrilla verticillata biomass for MG was found to be 91.97 mg/g at an initial pH 8.0 indicating that the optimum sorption initial pH. The external and intra particle diffusion models were also applied to sorption data of Hydrilla verticillata biomass with MG, and it was found that both the external diffusion as well as intra particle diffusion contributes to the actual sorption process. The pseudo-second order kinetic model described the MG sorption process with a good fitting.

of 10% sucrose was used as a control medium. Results indicated that
the optimum period for xanthan gum production in this condition was
4 days.
Abstract: Fructooligosaccharides derived from microbial enzyme especially from fungal sources has been received particular attention due to its beneficial effects as prebiotics and mass production. However, fungal fermentation is always cumbersome due to its broth rheology problem that will eventually affect the production of FOS. This study investigated the efficiency of immobilized cell system using rotating fibrous bed bioreactor (RFBB) in producing fructooligosaccharides (FOS). A comparative picture with respect to conventional stirred tank bioreactor (CSTB) and RFBB has been presented. To demonstrate the effect of agitation intensity and aeration rate, a laboratory-scale bioreactor 2.5 L was operated in three phases (high, medium, low) for 48 hours. Agitation speed has a great influence on P. simplicissimum fermentation for FOS production, where the volumetric FOS productivity using RFBB is increased with almost 4 fold compared to the FOS productivity in CSTB that only 0.319 g/L/h. Rate of FOS production increased up to 1.2 fold when immobilized cells system was employed at aeration rate similar to the freely suspended cells at 2.0 vvm.
