An Evaluation of Pesticide Stress Induced Proteins in three Cyanobacterial Species-Anabaena Fertilissima, Aulosira Fertilissima and Westiellopsis Prolifica using SDS-PAGE

The whole-cell protein-profiling technique was evaluated for studying differences in banding pattern of three different species of Cyanobacteria i.e. Anabaena fertilissima, Aulosira fertilissima and Westiellopsis prolifica under the influence of four different pesticides-2,4-D (Ethyl Ester of 2,4-Dichloro Phenoxy Acetic Acid), Pencycuron (N-[(4-chlorophenyl)methyl]-Ncyclopentyl- N'–phenylurea), Endosulfan (6,7,8,9,10,10hexachloro- 1,5,5a,6,9,9a-hexahydro-6,9-methano-2,4,3-benzodioxathiepine-3- oxide) and Tebuconazole (1-(4-Chlorophenyl)-4,4-dimethyl-3-(1,2,4- triazol-1-ylmethyl)pentan-3-ol). Whole-cell extracts were obtained by sonication treatment (Sonifier cell disruptor -Branson Digital Sonifier S-450D, USA) and were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). SDS-PAGE analyses of the total protein profile of Anabaena fertilissima, Aulosira fertilissima and Westiellopsis prolifica showed a linear decrease in the protein content with increasing pesticide stress when administered to different concentrations of 2, 4-D, Pencycuron, Endosulfan and Tebuconazole. The results indicate that different stressors exert specific effects on cyanobacterial protein synthesis.

Field Investigation on Modification of Japanese Cedar Pollen Allergen in Urban Air-Polluted Area

Cry j 1 is a causative substance of Japanese cedar pollinosis, and it may deteriorate by Cry j 1 invasion to a lower respiratory tract. We observed airborne particles containing Cry j 1 by an immunofluorescence technique using a fluorescence microscope, and we clarified that Cry j 1 exist as aggregates of airborne fine particles (< 1.1 μm) in the urban atmosphere. Airborne Cry j 1 may react with air pollutants and be denature to a substance deteriorated Japanese cedar pollinosis. Therefore, we applied a sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) to evaluate a Cry j 1 reacted with various air pollutants by liquid phase reaction, and calculated kinetics constants of Cry j 1 extracted from pollens collected in various sites and airborne fine particles containing Cry j 1 by using a surface plasmon resonance (SPR) method. As a result, it is suggested that Cry j 1 may be denatured by air pollutants during the transportation to the urban atmosphere.

Risk Assessment of Acrylamide Intake from Roasted Potatoes in Latvia

From food consumption surveys has been found that potato consumption comparing to other European countries is one of the highest. Hence acrylamide (AA) intake coming from fried potatoes in population might be high as well. The aim of the research was to determine acrylamide content and estimate intake of acrylamide from roasted potatoes bred and cultivated in Latvia. Five common Latvian potato varieties were selected: Lenora, Brasla, Imanta, Zile, and Madara. A two-year research was conducted during two periods: just after harvesting and after six months of storage. Time and temperature (210 ± 5°C) was recorded during frying. AA was extracted from potatoes by solid phase extraction and AA content was determined by LC-MS/MS. estimated intake of acrylamide ranges from 0.012 to 0.496μgkg-1 BW per day.

Synthesis and Analysis of Swelling and Controlled Release Behaviour of Anionic sIPN Acrylamide based Hydrogels

In modern agriculture, polymeric hydrogels are known as a component able to hold an amount of water due to their 3-dimensional network structure and their tendency to absorb water in humid environments. In addition, these hydrogels are able to controllably release the fertilisers and pesticides loaded in them. Therefore, they deliver these materials to the plants' roots and help them with growing. These hydrogels also reduce the pollution of underground water sources by preventing the active components from leaching. In this study, sIPN acrylamide based hydrogels are synthesised by using acrylamide free radical, potassium acrylate, and linear polyvinyl alcohol. Ammonium nitrate is loaded in the hydrogel as the fertiliser. The effect of various amounts of monomers and linear polymer, measured in molar ratio, on the swelling rate, equilibrium swelling, and release of ammonium nitrate is studied.

Thermo-Sensitive Hydrogel: Control of Hydrophilic-Hydrophobic Transition

The study investigated the hydrophilic to hydrophobic transition of modified polyacrylamide hydrogel with the inclusion of N-isopropylacrylamide (NIAM). The modification was done by mimicking micellar polymerization, which resulted in better arrangement of NIAM chains in the polyacrylamide network. The degree of NIAM arrangement is described by NH number. The hydrophilic to hydrophobic transition was measured through the partition coefficient, K, of Orange II and Methylene Blue in hydrogel and in water. These dyes were chosen as a model for solutes with different degree of hydrophobicity. The study showed that the hydrogel with higher NH values resulted in better solubility of both dyes. Moreover, in temperature above the lower critical solution temperature (LCST) of Poly(N-isopropylacrylamide) (PNIAM)also caused the collapse of NIPAM chains which results in a more hydrophobic environment that increases the solubility of Methylene Blue and decreases the solubility of Orange II in the hydrogels with NIPAM present.

The Suitability of Potato Cultivars in Production of Chips and Sticks by Using Microwave-Vacuum Drier

The aim of present experiment was to evaluate the influence of cultivar to quality parameters of dried potato chips and sticks produced in microwave-vacuum drier. The potatoes before drying were blanched in oil and water at 180ºC and at 85ºC respectively. The moisture content, crispiness, the colour (CIE L*a*b*), the content of ascorbic acid, total carotenoids and total fat content of dried potato chips and sticks was determined The highest ascorbic acid content, high content of carotenoids, low total fat content, low acrylamide content and good crispiness (low breaking force) especially for sticks was determined in the samples of Gundega cultivar.

pH-Responsiveness Properties of a Biodigradable Hydrogels Based on Carrageenan-g-poly(NaAA-co-NIPAM)

A novel thermo-sensitive superabsorbent hydrogel with salt- and pH-responsiveness properties was obtained by grafting of mixtures of acrylic acid (AA) and N-isopropylacrylamide (NIPAM) monomers onto kappa-carrageenan, kC, using ammonium persulfate (APS) as a free radical initiator in the presence of methylene bisacrylamide (MBA) as a crosslinker. Infrared spectroscopy was carried out to confirm the chemical structure of the hydrogel. Moreover, morphology of the samples was examined by scanning electron microscopy (SEM). The effect of MBA concentration and AA/NIPAM weight ratio on the water absorbency capacity has been investigated. The swelling variations of hydrogels were explained according to swelling theory based on the hydrogel chemical structure. The hydrogels exhibited salt-sensitivity and cation exchange properties. The temperature- and pH-reversibility properties of the hydrogels make the intelligent polymers as good candidates for considering as potential carriers for bioactive agents, e.g. drugs.

Protein Production by Bacillus Subtilis Atcc 21332 in the Presence of Cymbopogon Essential Oils

Proteins levels produced by bacteria may be increased in stressful surroundings, such as in the presence of antibiotics. It appears that many antimicrobial agents or antibiotics, when used at low concentrations, have in common the ability to activate or repress gene transcription, which is distinct from their inhibitory effect. There have been comparatively few studies on the potential of antibiotics or natural compounds in nature as a specific chemical signal that can trigger a variety of biological functions. Therefore, this study was focusing on the effect of essential oils from Cymbopogon flexuosus and C. nardus in regulating proteins production by Bacillus subtilis ATCC 21332. The Minimum Inhibition Concentrations (MICs) of both essential oils on B. subtilis were determined by using microdilution assay, resulting 0.2% and 1.56% for each C. flexuosus and C. nardus subsequently. The bacteria were further exposed to each essential oils at concentration of 0.01XMIC for 2 days. The proteins were then isolated and analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Protein profile showed that a band with approximate size of 250 kD was appeared for the treated bacteria with essential oils. Thus, Bacillus subtilis ATCC 21332 in stressful condition with the presence of essential oils at low concentration could induce the protein production.

Optimization of Enzymatic Hydrolysis of Manihot Esculenta Root Starch by Immobilizeda-Amylase Using Response Surface Methodology

Enzymatic hydrolysis of starch from natural sources finds potential application in commercial production of alcoholic beverage and bioethanol. In this study the effect of starch concentration, temperature, time and enzyme concentration were studied and optimized for hydrolysis of cassava (Manihot esculenta) starch powder (of mesh 80/120) into glucose syrup by immobilized (using Polyacrylamide gel) a-amylase using central composite design. The experimental result on enzymatic hydrolysis of cassava starch was subjected to multiple linear regression analysis using MINITAB 14 software. Positive linear effect of starch concentration, enzyme concentration and time was observed on hydrolysis of cassava starch by a-amylase. The statistical significance of the model was validated by F-test for analysis of variance (p < 0.01). The optimum value of starch concentration temperature, time and enzyme concentration were found to be 4.5% (w/v), 45oC, 150 min, and 1% (w/v) enzyme. The maximum glucose yield at optimum condition was 5.17 mg/mL.

Effects of Polymers and Alkaline on Recovery Improvement from Fractured Models

In this work, several ASP solutions were flooded into fractured models initially saturated with heavy oil at a constant flow rate and different geometrical characteristics of fracture. The ASP solutions are constituted from 2 polymers i.e. a synthetic polymer, hydrolyzed polyacrylamide as well as a biopolymer, a surfactant and 2types of alkaline. The results showed that using synthetic hydrolyzed polyacrylamide polymer increases ultimate oil recovery; however, type of alkaline does not play a significant rule on oil recovery. In addition, position of the injection well respect to the fracture system has remarkable effects on ASP flooding. For instance increasing angle of fractures with mean flow direction causes more oil recovery and delays breakthrough time. This work can be accounted as a comprehensive survey on ASP flooding which considers most of effective factors in this chemical EOR method.
