Abstract: Legume crops are able to fix atmospheric nitrogen by the symbiotic relation with specific bacteria, which allows the use of the mineral nitrogen-fertilizer to be reduced, or even excluded, resulting in more profit for the farmers and less pollution for the environment. Soybean (Glycine max (L.) Merrill) is one of the most important legumes with its high content of both protein and oil. However, it is recommended to combine the two nitrogen sources under stress conditions in order to overcome its negative effects. Drought stress is one of the most important abiotic stresses that increasingly limits soybean yields. A precise rate of mineral nitrogen under drought conditions is not confirmed, as it depends on many factors; soybean yield-potential and soil-nitrogen content to name a few. An experiment was conducted during 2017 growing season in Debrecen, Hungary to investigate the effects of nitrogen source on the physiology and the yield of the soybean cultivar 'Boglár'. Three N-fertilizer rates including no N-fertilizer (0 N), 35 kg ha-1 of N-fertilizer (35 N) and 105 kg ha-1 of N-fertilizer (105 N) were applied under three different irrigation regimes; severe drought stress (SD), moderate drought stress (MD) and control with no drought stress (ND). Half of the seeds in each treatment were pre-inoculated with Bradyrhizobium japonicum inoculant. The overall results showed significant differences associated with fertilization and irrigation, but not with inoculation. Increasing N rate was mostly accompanied with increased chlorophyll content and leaf area index, whereas it positively affected the plant height only when the drought was waived off. Plant height was the lowest under severe drought, regardless of inoculation and N-fertilizer application and rate. Inoculation increased the yield when there was no drought, and a low rate of N-fertilizer increased the yield furthermore; however, the high rate of N-fertilizer decreased the yield to a level even less than the inoculated control. On the other hand, the yield of non-inoculated plants increased as the N-fertilizer rate increased. Under drought conditions, adding N-fertilizer increased the yield of the non-inoculated plants compared to their inoculated counterparts; moreover, the high rate of N-fertilizer resulted in the best yield. Regardless of inoculation, the mean yield of the three fertilization rates was better when the water amount increased. It was concluded that applying N-fertilizer to provide the nitrogen needed by soybean plants, with the absence of N2-fixation process, is very important. Moreover, adding relatively high rate of N-fertilizer is very important under severe drought stress to alleviate the drought negative effects. Further research to recommend the best N-fertilizer rate to inoculated soybean under drought stress conditions should be executed.
Abstract: Microgravity is known to be a major abiotic stress in space which affects plants depending on the duration of exposure. In this work, tomatoes seeds were exposed to long hours of simulated microgravity condition using a one-axis clinostat. The seeds were sown on a 1.5% combination of plant nutrient and agar-agar solidified medium in three Petri dishes. One of the Petri dishes was mounted on the clinostat and allowed to rotate at the speed of 20 rpm for 72 hours, while the others were subjected to the normal gravity vector. The anatomical sections of both clinorotated and normal gravity plants were made after 72 hours and observed using a Phase-contrast digital microscope. The percentage germination, as well as the growth rate of the normal gravity seeds, was higher than the clinorotated ones. The germinated clinorotated roots followed different directions unlike the normal gravity ones which grew towards the direction of gravity vector. The clinostat was able to switch off gravistimulation. Distinct cellular arrangement was observed for tomatoes under normal gravity condition, unlike those of clinorotated ones. The root epidermis and cortex of normal gravity are thicker than the clinorotated ones. This implied that under long-term microgravity influence, plants do alter their anatomical features as a way of adapting to the stress condition.
Abstract: The semidental sweet maize of Armenian population under drought stress and pollution by some heavy metals (HMs) in sites along the river Debet was studied. Accordingly, the objective of this work was to investigate the antioxidant status of maize plant in order to identify simple and reliable criteria for assessing the degree of adaptation of plants to abiotic stress of drought and HMs. It was found that in the case of removal from the mainstream of the river, the antioxidant status of the plant varies. As parameters, the antioxidant status of the plant has been determined by the activity of malondialdehyde (MDA) and Ferric Reducing Ability of Plasma (FRAP), taking into account the characteristics of natural drought of this region. The possibility of using some indicators which characterized the antioxidant status of the plant was concluded. The criteria for assessing the extent of environmental pollution could be HMs. This fact can be used for the early diagnosis of diseases in the population who lives in these areas and uses corn as the main food.
Abstract: In Arabidopsis, several genes encoding proteins with ankyrin repeats and transmembrane domains (AtANKTM) have been identified as mediators of biotic and abiotic stress responses. It has been known that the expression of an AtANKTM gene, At2g24600, is induced in response to abiotic stress and that there are four splicing variants derived from this locus. In this study, by RT-PCR and sequencing analysis, an unknown splicing variant of the At2g24600 transcript was identified. Based on differences in the predicted amino acid sequences, the five splicing variants are divided into three groups. The three predicted proteins are highly homologous, yet have different numbers of ankyrinrepeats and transmembrane domains. It is generally considered that ankyrin repeats mediate protein-protein interaction and that the number oftransmembrane domains affects membrane topology of proteins. The protein variants derived from the At2g24600 locus may have different molecular functions each other.
Abstract: To evaluate genetic variation of wheat (Triticum aestivum) affected by heat and drought stress on eight Australian wheat genotypes that are parents of Doubled Haploid (HD) mapping populations at the vegetative stage, the water stress experiment was conducted at 65% field capacity in growth room. Heat stress experiment was conducted in the research field under irrigation over summer. Result show that water stress decreased dry shoot weight and RWC but increased osmolarity and means of Fv/Fm values in all varieties except for Krichauff. Krichauff and Kukri had the maximum RWC under drought stress. Trident variety was shown maximum WUE, osmolarity (610 mM/Kg), dry mater, quantum yield and Fv/Fm 0.815 under water stress condition. However, the recovery of quantum yield was apparent between 4 to 7 days after stress in all varieties. Nevertheless, increase in water stress after that lead to strong decrease in quantum yield. There was a genetic variation for leaf pigments content among varieties under heat stress. Heat stress decreased significantly the total chlorophyll content that measured by SPAD. Krichauff had maximum value of Anthocyanin content (2.978 A/g FW), chlorophyll a+b (2.001 mg/g FW) and chlorophyll a (1.502 mg/g FW). Maximum value of chlorophyll b (0.515 mg/g FW) and Carotenoids (0.234 mg/g FW) content belonged to Kukri. The quantum yield of all varieties decreased significantly, when the weather temperature increased from 28 ÔùªC to 36 ÔùªC during the 6 days. However, the recovery of quantum yield was apparent after 8th day in all varieties. The maximum decrease and recovery in quantum yield was observed in Krichauff. Drought and heat tolerant and moderately tolerant wheat genotypes were included Trident, Krichauff, Kukri and RAC875. Molineux, Berkut and Excalibur were clustered into most sensitive and moderately sensitive genotypes. Finally, the results show that there was a significantly genetic variation among the eight varieties that were studied under heat and water stress.
Abstract: In most wheat growing moderate regions and
especially in the north of Iran climate, is affected grain filling by
several physical and abiotic stresses. In this region, grain filling often
occurs when temperatures are increasing and moisture supply is
decreasing. The experiment was designed in RCBD with split plot
arrangements with four replications. Four irrigation treatments
included (I0) no irrigation (check); (I1) one irrigation (50 mm) at
heading stage; (I2) two irrigation (100 mm) at heading and anthesis
stage; and (I3) three irrigation (150 mm) at heading, anthesis and
early grain filling growth stage, two wheat cultivars (Milan and
Shanghai) were cultured in the experiment. Totally raining was 453
mm during the growth season. The result indicated that biological
yield, grain yield and harvest index were significantly affected by
irrigation levels. I3 treatment produced more tillers number in m2,
fertile tillers number in m2, harvest index and biological yield. Milan
produced more tillers number in m2, fertile tillers in m2, while
Shanghai produced heavier tillers and grain 1000 weight. Plant height
was significant in wheat varieties while were not statistically
significant in irrigation levels. Milan produced more grain yield,
harvest index and biological yield. Grain yield shown that I1, I2, and
I3 produced increasing of 5228 (21%), 5460 (27%) and 5670 (29%)
kg ha-1, respectively. There was an interaction of irrigation and
cultivar on grain yields. In the absence of the irrigation reduced grain
1000 weight from 45 to 40 g. No irrigation reduced soil moisture
extraction during the grain filling stage. Current assimilation as a
source of carbon for grain filling depends on the light intercepting
viable green surfaces of the plant after anthesis that due to natural
senescence and the effect of various stresses. At the same time the
demand by the growing grain is increasing. It is concluded from
research work that wheat crop irrigated Milan cultivar could increase
the grain yield in comparison with Shanghai cultivar. Although, the
grain yield of Shanghai under irrigation was slightly lower than
Milan. This grain yield also was related to weather condition, sowing
date, plant density and location conditions and management of
fertilizers, because there was not significant difference in biological
and straw yield. The best result was produced by I1 treatment. I2 and
I3 treatments were not significantly difference with I1 treatment.
Grain yield of I1 indicated that wheat is under soil moisture
deficiency. Therefore, I1 irrigation was better than I0.
Abstract: The mechanism of abiotic stress tolerance is crucial
for plants to survive in harsh condition and the knowledge of this
mechanism can be use to solve the problem of declining productivity
of plants or crops around the world. However in-depth description is
still unclear and it is argued, in particular that there is a relationship
between high salinity tolerance and the ability to tolerate high light
condition. In this study, Dunaliella salina, which can withstand high
salt was used as a model. Chlorophyll fluorometer for nonphotochemical
quenching (NPQ) measurement and high-performance
liquid chromatography for pigment determination was used. The
results show that NPQ value and the amount of pigment were
increased along with the levels of salinity. However, it establish a
clear relationship between high salt and high light but the further
study to optimized the solutions mentioned above is still required.
