Abstract: Obesity as a global health challenge motivates pharmaceutical industries to produce anti-obesity drugs. However, effectiveness of these agents is remained unclear. Because of popularity of dietary supplements, the aim of this study was tp investigate the effects of Conjugated Linoleic Acid (CLA) and L-carnitine (LC) on serum glucose, triglyceride, cholesterol and weight changes in diet induced obese rats. 48 male Wistar rats were randomly divided into two groups: Normal fat diet (n=8), and High fat diet (HFD) (n=32). After eight weeks, the second group which was maintained on HFD until the end of study, was subdivided into four categories: a) 500 mg Corn Oil (as control group), b) 500 mg CLA, c) 200 mg LC, d) 500 mg CLA+ 200 mg LC.All doses are planned per kg body weights, which were administered by oral gavage for four weeks. Body weights were measured and recorded weekly by means of a digital scale. At the end of the study, blood samples were collected for biochemical markers measurement. SPSS Version 16 was used for statistical analysis. At the end of 8th week, a significant difference in weight was observed between HFD and NFD group. After 12 weeks, LC significantly reduced weight gain by 4.2%. Trend of weight gain in CLA and CLA+LC groups was insignificantly decelerated. CLA+LC reduced triglyceride level significantly, but just CLA had significant influence on total cholesterol and insignificant decreasing effect on FBS. Our results showed that an obesogenic diet in a relative short time led to obesity and dyslipidemia which can be modified by LC and CLA to some extent.
Abstract: Nowadays, nanoparticles are being used in pharmacological studies for their exclusive properties such as small size, more surface area, biocompatibility and enhanced solubility. In view of this, the present study aimed to evaluate the antihyperglycemic potential of biologically synthesized silver nanoparticles (BSSNPs) and Gymnema sylvestre (GS) extract. The SEM and SEM analysis divulges that the BSSNPs were spherical in shape. EDAX spectrum exhibits peaks for the presence of silver, carbon, and oxygen atoms in the range of 1.0-3.1 keV. FT-IR reveals the binding properties of active bio-constituents responsible for capping and stabilizing BSSNPs. The results showed increased blood glucose, huge loss in body weight and downturn in plasma insulin. The GS extract (200 mg/kg, 400 mg/kg), BSSNPs (100 mg/kg, 200 mg/kg) and metformin 50 mg/kg were administered to the diabetic rats. BSSNPs at a dose level of 200 mg/kg (b.wt.p.o.) showed significant inhibition of (p
Abstract: Anthocyanins are natural pigments with effective UV
protection but their topical use could be limited due to their
physicochemical characteristics. An attempt to overcome such
limitations by complexation of 2 major anthocyanin-rich sources, C.
ternatea and Z. mays, has potentiated its use as topical antiinflammatory.
Cell studies indicate no cytotoxicity of the
anthocyanin complex (AC) up to 1 mg/ml tested in HaCaT and
human fore head fibroblasts by MTT. Croton oil-induced ear edema
in Wistar rats suggests an effective dose of 5 mg/cm2 of AC as a
topical anti-inflammatory in comparison to 0.5 mg/cm2 of
fluocinolone acetonide. Niosomal encapsulation of the AC
significantly prolonged the anti-inflammatory activity particularly at
8 h after topical application (p = 0.0001). The AC was not cytotoxic
and its anti-inflammatory and activity was dose-dependent and
prolonged by niosomal encapsulation. It has also shown to promote
collagen type 1 production in cell culture. Thus, AC could be a
potential candidate for topical anti-inflammatory agent from natural
resources.
Abstract: The goal of this paper is to examine the effects of laser
radiation on the skin wound healing using infrared thermography as
non-invasive method for the monitoring of the skin temperature
changes during laser treatment. Thirty Wistar rats were used in this
study. A skin lesion was performed at the leg on all rats. The animals
were exposed to laser radiation (λ = 670 nm, P = 15 mW, DP = 16.31
mW/cm2) for 600 s. Thermal images of wound were acquired before
and after laser irradiation. The results have demonstrated that the
tissue temperature decreases from 35.5±0.50°C in the first treatment
day to 31.3±0.42°C after the third treatment day. This value is close
to the normal value of the skin temperature and indicates the end of
the skin repair process. In conclusion, the improvements in the
wound healing following exposure to laser radiation have been
revealed by infrared thermography.
Abstract: The excessive use of agricultural pesticides and the
resulting contamination of food and beds of rivers have been a
recurring problem nowadays. Some of these substances can cause
changes in endocrine balance and impair reproductive function of
human and animal population. In the present study, we evaluated the
possible effects of the fungicide cuprous copper oxide Sandoz® on
pregnant Wistar rats. They received a daily oral administration of 103
or 3.103 mg/kg of the fungicide from the 6th to the 15th day of
gestation. On day 21 of gestation, the maternal and fetal toxicity
parameters and indices were determined. The administration of
cuprous oxide (Copper Sandoz) in Wistar rats, the period of
organogenesis, revealed no evidence of maternal toxicity or embryo
at the studied doses.
Abstract: Anxiety is a common disorder that attacks many people in society and often accompanied by physiological sensations such as tachycardia, chest pain, shortness of breath, insensitivity and etc. The purpose of this study is to characterize the putative anxiolytic-like effects of DEV (dichloromethane extracts of valerian) using the elevated plus maze (EPM) in rats. DEV was dissolved in DMSO and orally administered at different doses to adult male wistar rats, 0.5, 1.5 and 3 hours before behavioral evaluation in an EPM respectively. Control rats were treated with an equal volume of DMSO. Single treatment of DEV (at 0.1,0.2. 0.3, and 0.4 g/kg) significantly increased time-spent and arm entries into open arms of EPM versus control groups (p
Abstract: Recent studies demonstrated that high-fat diet increases oxidative stress in plasma and in a variety of tissues. Many researchers have been looking for natural products, which can reverse the effect of high fat diet. Recently, buckwheat is becoming common ingredient in functional food because of it properties. In study on buckwheat, it is known that, this plant plays roles as anti-oxidative, anti-inflammatory and anti-hypertensive. Nevertheless still little is known about buckwheat groats. The aim of this study was to investigate the effects of addition of buckwheat groats to the fat diet (30% lard), on some antioxidant and oxidant stress parameters in plasma and selected tissues in Wistar rats. The experiment was carried out with three months old male Wistar rats ca. 250g of body weight fed for 5 weeks with either a high-fat (30% of lard) diet or control diet, with or without addition of buckwheat groats. In plasma biochemistry and the activities of the antioxidant enzymes were measured selected tissues: glutathione peroxidase (GPX), catalase (CAT) and the levels of total and reduced glutathione (GSH), free thiol groups (pSH), antioxidant potential of plasma (FRAP) and oxidant stress indices - proteins carbonyl groups (CO) and malonyldialdehyde concentration (MDA). Activity of catalase (CAT) in plasma of rats was significantly increased in buckwheat groats groups and activity of GPx3 in plasma of rats was decreased in buckwheat groups as compared to control group. The reduced glutathione (GSH) in plasma of rats was significantly increased and protein CO was significantly decreased in buckwheat groups as compared to controls. The lowered concentration of GSH was found in serum of rats fed buckwheat groats addition but it accompanied in 7-fold increase in reduced-to-oxidized glutatione ratio, significant increase in HDL and decrease in nonHDL concentration. Conclusions: Buckwheat groats indicate a beneficial effect in inhibiting protein and lipid peroxidation in rats and improved lipid profile. These results suggest that buckwheat groats exert a significant antioxidant potential and may be used as normal food constituent to ameliorate the oxidant-induced damage in organism.
Abstract: In Indonesia, goat milk is often consumed and
believed as anti-allergy. The objective of this research was to study
the effect of goat milk and their fractions (casein and whey)
supplementation on total serum IgE concentrations and leukocytes
count in rat sensitized with contact allergen dinitrochlorobenzene
(DNCB). Female Wistar rats 6-8 weeks old were divided into four
groups: 1) whey, 2) casein, 3) whole milk supplementation and 4)
phosphate-buffered saline/PBS (control). The results showed that
supplementation of goat milk on rats did not affects on total serum
IgE concentrations and number of leukocytes. After sensitized with
DNCB, the monocyte percentage in rats was higher (P
Abstract: The neurogenic potential of many herbal extracts used
in Indian medicine is hitherto unknown. Extracts derived from
Clitoria ternatea Linn have been used in Indian Ayurvedic system of
medicine as an ingredient of “Medhya rasayana", consumed for
improving memory and longevity in humans and also in treatment of
various neurological disorders. Our earlier experimental studies with
oral intubation of Clitoria ternatea aqueous root extract (CTR) had
shown significant enhancement of learning and memory in postnatal
and young adult Wistar rats. The present study was designed to
elucidate the in vitro effects of 200ng/ml of CTR on proliferation,
differentiation and growth of anterior subventricular zone neural
stem cells (aSVZ NSC-s) derived from prenatal and postnatal rat
pups. Results show significant increase in proliferation and growth of
neurospheres and increase in the yield of differentiated neurons of
aSVZ neural precursor cells (aSVZNPC-s) at 7 days in vitro when
treated with 200ng/ml of CTR as compared to age matched control.
Results indicate that CTR has growth promoting neurogenic effect on
aSVZ neural stem cells and their survival similar to neurotrophic
factors like Survivin, Neuregulin 1, FGF-2, BDNF possibly the basis
for enhanced learning and memory.
Abstract: Dexamethasone (Dex) is a synthetic glucocorticoid
that is used in therapy. However prolonged treatments with high
doses are often required. This causes side effects that interfere with
the activity of several endocrine systems, including the gonadotropic
axis.
The aim of our study is to determine the effect of Dex on testicular
function in prepubertal Wistar rats.
Newborn Wistar rats are submitted to intraperitoneal injection of
Dex (1μg of Dex dissolved in NaCl 0.9% / 5g bw) for 20 days and
then sacrificed at the age of 40days. A control group received NaCl
0.9%. The rat is weighed daily. The plasmatic levels of testosterone,
LH and FSH were measured by radioimmunoassay. A histomorphometric
study was performed on sections of testis.
Treated groups showed a significant decrease in body weight (p
Abstract: Leonotisleonurus a shrub indigenous to Southern
Africa is widely used in traditional medicine to treat a variety of
conditions ranging from skin diseases and cough to epileptic fits and
‘heart problems’. Studies on the aqueous extract of the leaves have
indicated cycloxegenase enzyme inhibitory activity and an
antihypertensive effect.
Five methanol leaf extract fractions (MLEa - MLEe) of L.
leonurus were tested on anaesthetized normotensive male Wistar rats
(AWR) and isolated perfused working rat hearts (IWH). Fraction
MLEc (0.01mg/kg – 0.05mg/kg) induced significant increases in BP
and HR in AWR and positive chronotropic and inotropic effects in
IWH (1.0mg/ml – 5.0mg/ml). Pre-administration of atenolol
(2.0mg/kg) and prazosin (60μg/kg) significantly inhibited MLEc
effect on HR and MAP respectively in vivo, while atenolol
(7.0mg/ml) pre-perfusion significantly inhibited MLEc effect in vitro.
The hypertensive effect of MLEc is probably via β1agonism.
Results also indicate the presence of multiple cardioactive
compounds in L. leonurus.
Abstract: Goat milk has an hypoallergenic effects, and allergic
diseases related to abnormal of intestinal flora. Probiotic microorganisms
do exert an activity on the immune system in the skin of
the individual. The purpose of this study are to determine the number
of leukocyte and lymphocyte proliferation in rat supplemented with
fermented goat milk (acidophilus milk and kefir) and sensitized with
dinitrochlorobenzene (DNCB). Female Wistar rats 6-8 weeks olds
were divided into 3 treatment groups. The first group supplemented
goat milk kefir, second group acidophilus goat milk, and third group
as control. During 28-day experiment, on day 15 rat sensitized with
allergen DNCB on the dorsal of the body, and on day 24 was
challenged with DNCB on the ear. Sampling of blood and tissue of
intestinal Peyer'patch (PP) were performed on day 14 (before DNCB
sensitized) and on day 28 (after DNCB sensitized). The results
showed the number of neutrophils in rats supplemented with
acidophilus milk was higher (P
